Engineering exosomes with iRGD for targeted RNAi therapy against pancreatic cancer mediated by long non-coding RNA PLBD1-AS1.

Tumor-derived exosomes play critical roles in pancreatic ductal adenocarcinoma (PDAC) progression by mediating intercellular communication within the tumor microenvironment. This study identifies the long non-coding RNA PLBD1-AS1 as a functional oncogenic lncRNA enriched in PDAC exosomes. We demonstrate that PLBD1-AS1 promotes tumor cell proliferation, migration, and invasion by interacting with the glycolytic enzyme ALDOA and enhancing glycolytic flux. Furthermore, tumor exosomes deliver PLBD1-AS1 to pancreatic stellate cells (PSC), augmenting their glycolysis and facilitating their activatio

Atmosphere-Controlled Valence Engineering of Eu in Aluminate Crystals Enabling High-Performance Temperature Sensing and Flexible X-Ray Imaging.

Developing scintillators that simultaneously deliver high light yield, radiation stability, flexibility, and environmental sustainability remains a long-standing challenge, while optical thermometry is further limited by narrow working ranges and insufficient sensitivity. Here, we propose a buried-carbon reduction concept that enables precise regulation of Eu valence states through controlled carbon burial depth from a single precursor, thereby unlocking multifunctionality within one material system. By such strategy, fully reduced SrMgAl10O17:Eu2+ is utilized in x-ray detection and imaging. H

Bioinspired Nanomaterials for Parkinson's Disease: Engineering Membrane Interfaces to Overcome the Blood-Brain Barrier and Enable Neuron-Targeted Delivery.

Parkinson's disease (PD) is a progressive neurodegenerative disorder marked by the selective loss of dopaminergic neurons in the substantia nigra, which gives rise to characteristic motor and non-motor behavioral impairments. Disease-modifying therapies remain elusive, largely due to the restrictive nature of the blood-brain barrier (BBB) and the lack of effective neuron-targeting strategies. Bioinspired nanomaterials-including cell membrane-derived vesicles, engineered biomimetic nanocarriers, and liposomes-have emerged as versatile platforms to address these limitations. By recapitulating ke

The Role of Morphology and Phase Engineering in Molybdenum Sulfide for Robust Photoelectrochemical Hydrogen Evolution on III-V Photoelectrodes.

Solar-driven hydrogen production via photoelectrochemical (PEC) water splitting offers a direct route to decarbonized fuels without grid dependence. Although III-V semiconductors exhibit exceptional optoelectronic properties, their reliance on platinum group cocatalysts limits scalability. Here, we report a platinum group metal-free (PGM-free) hybrid photocathode integrating an epitaxially grown GaAs/Si:p junction with mixed-phase MoS2 nanostructures for efficient hydrogen evolution in acidic media. Through systematic control of morphology, crystallinity, and phase composition, we demonstrate

Multifunctional Nanoreactor Based on Catalyst Surface Engineering Combines pH-Adaptive Catalysis With Tumor Microenvironment Modulation for Synergistic Chemodynamic-Chemotherapy.

Chemodynamic therapy (CDT) is a novel therapeutic strategy based on the Fenton reaction, which utilizes endogenous hydrogen peroxide (H2O2) to generate toxic hydroxyl radicals (•OH) for killing tumor cells. However, the mildly acidic pH and limited H2O2 levels in the tumor microenvironment (TME) restrict the activity of the Fenton reaction, severely limiting the therapeutic efficacy of CDT. Here, we construct a multifunctional nanoreactor, Fe2O3@MoS2-Pt/FA, which combines catalyst surface engineering with intracellular metabolic regulation to achieve highly efficient synergy between CDT and ch…

Engineering the oxidative myocardium: ROS-responsive biomaterials for precision cardiovascular delivery in ischemia-reperfusion injury and post-infarction remodeling.

Myocardial ischemia-reperfusion injury (MIRI) and post-infarction remodeling are characterized by dynamic oxidative stress that varies across myocardial regions and disease stages. Reactive oxygen species (ROS)-responsive and redox-modulating biomaterials seek to exploit this environment through stimulus-triggered release or degradation, ROS scavenging, targeted delivery, local retention, and stage-specific repair. This review critically evaluates preclinical strategies for MIRI, myocardial infarction, and post-infarction remodeling. Major platform classes differ substantially in mechanism and

Multilevel cytochrome P450 engineering integrating protein language model-guided evolution enables de novo biosynthesis of amentoflavone in Escherichia coli.

Amentoflavone-type biflavonoids exhibit potent neuroprotective activities but are limited by scarce natural abundance and inefficient chemical synthesis routes. Here, we establish a stable Escherichia coli platform for de novo amentoflavone biosynthesis through multilevel engineering of a cytochrome P450-dependent oxidative coupling module. Systematic optimization of redox partner pairing, N-terminal expression regulation, and intrinsic catalytic activity substantially enhances the performance of the key P450 enzyme GbCYP90J6. Protein language model-guided directed evolution identifies high-ac…

Electrochemical Gating of d-Band Engineering in Hierarchically Bridged Dual-Site Nanozymes for Synergistic Cascade Catalysis and Wearable Biosensing.

Cascade nanozymes for biosensing are fundamentally hampered by diffusion limitations and passive catalytic sites. Herein, we report a strategy of electrochemical gating of d-band engineering within a hierarchically bridged dual-site nanozyme (CuNCs@FeMOP) to achieve dynamic control over cascading catalysis. This architecture spatially confines the ascorbic acid oxidase-mimicking copper nanocluster (CuNC) core and the peroxidase-mimicking iron-based microporous organic polymer (FeMOP) shell, eliminating intermediate diffusion losses. More critically, synergistic electronic coupling via a histid…

Regulation of the Thermal and Electrical Transport Properties of Large-Pore Borophene Nanoribbons Based on Molecular Surface Adsorption Engineering.

Large-pore borophene, a novel two-dimensional (2D) material with exceptional electrical properties and unique electronic structure, shows thermal and electrical potential in nanoribbon configurations due to quantum confinement effects. Surface adsorption is an effective strategy to tune the physicochemical properties of low-dimensional materials due to enhanced electron/phonon scattering effects. In this work, thermal/electric transport behavior of large-pore borophene nanoribbons (LH_ψ) with surface-adsorbed DBHD molecules was systematically investigated by using density functional theory (DF

Overcoming the Thermodynamic Diffusion Barrier in DNA Cascade Amplifiers via Spatially Confined Entropy Reduction: A Versatile Kinetic Engineering Framework.

The reaction efficiency of solution-phase DNA cascade amplifiers is fundamentally constrained by stochastic Brownian motion, creating a thermodynamic diffusion bottleneck that limits sensitivity and speed. To overcome this, we present a general kinetic engineering framework based on spatially confined entropy reduction. By integrating a DNA tetrahedron scaffold with a cascade amplification circuit, we successfully transition the reaction from a stochastic dilute-phase regime to a deterministic pseudosolid-phase regime. For the first time, we employ phase-space kinetic mapping to visualize the …

Overcoming redox barriers in black phosphorus negative electrodes through lattice P-N engineering for fast-charging Li-ion batteries.

Electrochemical energy storage devices commonly face a trade-off between specific energy and power capability. Black phosphorus offers a high theoretical capacity of 2596 mA h g-1, but its practical application is limited by slow reaction kinetics during multiphase phosphorus redox processes. These sluggish transformations, involving bond rearrangement and soluble intermediates, hinder fast charging and compromise cycling stability. Here we show that introducing phosphorus-nitrogen bonds into a black phosphorus/carbon composite regulates the reaction pathway and accelerates charge transport. T

Thermal-Ionic Coupling in Transpiration-Inspired Janus Nanofiber Membranes: Synergistic Dry-Wet Interface Engineering for Solar-Driven Electricity Generation.

Harnessing solar evaporation for simultaneous steam and electricity generation is an emerging technology for sustainable energy-water systems, yet its development is fundamentally constrained by the inability to maintain a stable interfacial potential gradient in homogeneous materials. Here, we introduce a dry-wet interface engineering strategy that enables persistent thermal-ionic coupling for continuous electricity generation. Inspired by leaf transpiration, an asymmetric Janus nanofiber membrane is constructed to form a stable hydrophilic-hydrophobic interface that simultaneously regulates …

Interfacial Engineering of Dual-Layer Au-Shelled Ag Nanowires: Decoupling Chemical Stability From Ultrasensitive Surface-Enhanced Raman Spectroscopy Activity.

One-dimensional (1D) silver nanowires are promising surface-enhanced Raman spectroscopy (SERS) substrates, yet their plasmonic hotspots are spatially confined to sharp tips or junctions, leaving the extensive body optically inactive. Additionally, realizing high-performance Ag-based substrates faces a fundamental trade-off between chemical stability and surface activity. To simultaneously resolve these geometric and chemical limitations, we present a stepwise interfacial engineering strategy to synthesize 3D hierarchical dual-layer porous@smooth Au-shelled Ag nanowires. This architecture decou…

Covalent Engineering of a Durable Zwitterionic Sulfobetaine Interface on Titanium via Phosphonate Bridging and Anhydride Activation for Biofouling Resistance.

Biofouling of titanium implants, initiated by nonspecific protein adsorption and followed by bacterial and cellular adhesion, remains a critical challenge that compromises long-term implant performance. Herein, we report a covalent interfacial engineering strategy to construct a robust zwitterionic sulfobetaine interface on titanium via a stepwise grafting approach. This method integrates phosphonate anchoring, maleic anhydride activation, and subsequent ring-opening immobilization of sulfonated amines, enabling the formation of a chemically stable and high-density zwitterionic layer directly …

Molecular engineering of interactions between 1,4-butanediol diglycidyl ether and cellulose in hydrated ionic solvents for sustainable packaging materials.

The development of robust and wet-stable cellulose-based plastics is pivotal for replacing conventional plastics in packaging. This study addresses the critical challenge of balancing cellulose dissolution and crosslinking within a Lewis acid solvent system (AlCl3/ZnCl2/H2O). We introduce 1,4-butanediol diglycidyl ether (BDDE) as a crosslinker and systematically investigate its multifaceted role. Our findings reveal that BDDE, beyond its crosslinking function, significantly alters the solvent microstructure by strengthening water-water hydrogen bonds and reducing free water activity and polari
