Bridging divides: The role of social science in operationalising transdisciplinarity for epidemic preparedness in sub-Saharan Africa, 2012-2023.

This paper investigates the role of transdisciplinarity, social science, and communities' knowledge in epidemic and pandemic preparedness and response in sub-Saharan Africa. It addresses how social science contributions have evolved over the past decade, explores the ways in which transdisciplinary approaches have been implemented and developed and analyses how social science can help to operationalise transdisciplinarity. The research applies a conceptual framework focusing on the intersection of power, knowledge, and crisis, and adopts a definition of transdisciplinarity that emphasises mult

Social Structure of Sheep Flocks at Points of the Production Cycle and Relationship to Disease Spread, Using a Simulated Epidemic of Footrot.

Footrot is one of the top five globally important diseases of sheep and causes lameness, leading to poor welfare and productivity. Transmission of Dichelobacter nodosus, the causative agent, occurs via surfaces such as pasture or bedding and persistence occurs from diseased sheep shedding bacteria into the environment; D. nodosus cannot replicate off host. High resolution proximity sensors were deployed on a flock of Poll Dorset sheep for 10-17 days at several points of the production cycle (teasing, tupping, pregnancy, and lactation (<6-week-old lambs)) between July 2018 and May 2021. Genetic Diversity, Recombination, and Pathogenicity of Porcine Epidemic Diarrhea Virus Strains Circulating in China During 2023-2024.

Porcine epidemic diarrhea virus (PEDV) is an enteric alphacoronavirus that causes severe diarrhea and high mortality in neonatal piglets, posing a persistent threat to the global swine industry. Despite extensive vaccination, PEDV continues to circulate in China, underscoring the need for updated molecular epidemiological surveillance. Here, we investigated the prevalence, genetic diversity, and pathogenicity of PEDV strains circulating in China during 2023-2024. A total of 714 clinical samples were collected from diarrheic pigs in seven major swine-producing regions. PEDV RNA was detected in

Ten millennia of purifying selection on HLA-B27 reveals an ancient epidemic-scale burden of spondyloarthritis in West Eurasia.

HLA-B27 exemplifies an evolutionary trade-off between protection against infection and susceptibility to inflammatory disease. To investigate its long-term population history, this study examined three HLA-B27-tagging variants-rs116488202, rs4349859, and rs116666910-in present-day populations from UK biobank and 1000 Genomes Project, and in 15,836 ancient West Eurasian individuals. The estimated frequency of HLA-B27 reached 49.0% approximately 8500 years before present, then declined progressively to 3.9% in the present-day reference population. Comparison with genome-wide association study (G

PRMT3 restricts porcine epidemic diarrhea virus replication by disrupting the interaction between VAPA and the viral nucleocapsid protein.

Porcine epidemic diarrhea virus (PEDV) represents a severe threat to the global swine industry. Its infection process involves intricate virus-host interactions and immune evasion mechanisms, but effective therapeutic targets remain elusive. In this study, we identified protein arginine methyltransferase 3 (PRMT3) as a novel regulatory factor that significantly modulates PEDV infection via genome-wide CRISPR/Cas9 knockout library screening. Knockout or inhibition of PRMT3 markedly enhanced PEDV infection in multiple cell lines, including LLC-PK1, IPEC-J2, and primary porcine intestinal epithel

4-octyl itaconate inhibits porcine epidemic diarrhea virus replication by modulating the ferroptosis axis and the autophagy pathway.

Porcine epidemic diarrhea virus (PEDV) continues to pose a significant threat to the swine industry, leading to high mortality in neonatal piglets and substantial economic losses. The emergence of antigenic variants often compromises vaccine efficacy, highlighting the need for novel antiviral strategies. This study investigates the potential of 4-octyl itaconate (4-OI) as a novel therapeutic agent against PEDV. In vitro studies demonstrated that 4-OI exhibited dose-dependent antiviral activity across multiple cell lines, directly reduced viral infectivity, and displayed broad-spectrum inhibito

Viral genotype and the pace of epidemic waves: an assessment from SARS-CoV 2 genomics and wastewater surveillance data.

The SARS-CoV-2 pandemic was characterized by multiple epidemic waves, whose geographical non-synchronicity remains not fully understood. This study aimed to characterize this phenomenon at local and regional scales. To this end, we compared SARS-CoV-2 epidemics in a geographically isolated city (Trelew, Chubut, Argentina) with country-wide data between April 2020 and February 2022. This period encompassed distinct phases defined by non-pharmacological interventions, changes in population immunity, and shifts in dominant viral lineages. Routine daily case reports and clinical genomics data were…

Rapid identification of Sporothrix brasiliensis by MALDI-TOF MS directly from clinical cultures in an Amazonian epidemic setting.

Sporotrichosis caused by Sporothrix brasiliensis is a highly virulent zoonotic mycosis rapidly expanding in Brazil, with increasing records in Pará since 2018. The reference diagnosis relies on culture and the time-consuming induction of fungal dimorphism, which delays taxonomic confirmation for weeks. The objective of the study was to develop and validate an in-house spectral library by MALDI-TOF MS for the rapid identification of S. brasiliensis in the Amazon region of Pará, prioritizing the feasibility of direct identification from the filamentous phase (primary growth). Respiratory syncytial virus as an independent driver of a subsequent influenza epidemic in Berlin before and after the COVID-19 pandemic.

OBJECTIVES: Following the withdrawal of non-pharmaceutical interventions (NPI) implemented during the coronavirus disease 2019 (COVID-19) pandemic, the resurgence of respiratory viral infections in children deviated from established pre-pandemic seasonal patterns. We aimed to use the disruption in the seasonality of respiratory viral infections caused by NPI measures to identify key drivers of seasonality before and since the COVID-19 pandemic. METHODS: We applied a Bayesian model to crossover-predict the incidence of infections with human metapneumovirus, influenza viruses, and respiratory s

Resistance by applied immunology: fabricated typhus epidemic as civil protection in occupied Poland during World War II.

During World War II, Polish physicians Eugeniusz Łazowski and Stanisław Matulewicz fabricated a typhus epidemic, which they reported in detail only after the war. They injected inactivated Proteus bacteria to patients suffering from mild, flu-like ailments in order to trigger a positive Weil-Felix reaction in their serum, the standard diagnostic tool for typhus at that time. These falsely labelled typhus patients would then be protected from seizure by German occupiers, who were much concerned about transmission of this highly deadly disease. According to Łazowski and Matulewicz, this action s

Evolution of HIV epidemic trends and diagnostic delays in French Guiana: how can we make further progress?

BACKGROUND: As antiretroviral treatment indications evolved, an increasing proportion of patients have been treated; since 2017, most patients receive antiretroviral treatment, irrespective of their CD4 count, in French Guiana and neighboring countries. In this context, we aimed to model the epidemic and to study the evolution of the estimates of the intervals between HIV-infection and HIV diagnosis in French Guiana. METHODS: The study was descriptive and comparative. Anonymous data from all persons in the DAT'AIDS HIV cohort based on quality-controlled clinical records was aggregated into ye

Water, Hygiene and Inequality: Keys to Understanding and Combating the Current Cholera Epidemic in the Democratic Republic of Congo-A Narrative Review.

BACKGROUND AND AIM: The Democratic Republic of the Congo (DRC) is among the countries most affected by cholera in the world. Since the 1970s, endemic foci have persisted in the eastern lake provinces, where chronic armed conflict, massive population displacement, and extreme socioeconomic inequalities create particularly favorable conditions for transmission. Despite a national elimination plan involving more than 22 ministries, the epidemic continues: 46,800 cases and 1362 deaths were recorded as of August 2025. This review synthesizes recent data on the structural determinants, public health

Production of Recombinant S1 Protein of Porcine Epidemic Diarrhea Virus in Recombinant CHO Cells for Application in Indirect ELISA.

The N-terminal S1 subunit of the spike protein (S1 protein) of porcine epidemic diarrhea virus (PEDV) is recognized as a potential diagnostic antigen for detecting anti-PEDV immunoglobulin A (IgA) levels in colostrum, a key indicator for assessing passive immunity against PEDV infection. Given the advantages of producing Fc-fusion proteins in mammalian cells, it is important to investigate how the addition of the Fc region affects the diagnostic performance of the PEDV-S1 protein. In this study, we successfully produced full-length PEDV-S1 protein fused with an Fc region (PEDV-S1-Fc protein) v
