Attenuation of diet-induced hepatic steatosis by capric acid-rich MLM structured lipids in an obese mouse model.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has emerged as a major hepatic manifestation of obesity and insulin resistance and represents a growing global health burden. Dietary lipid structure plays a decisive role in lipid metabolic fate and hepatic lipid accumulation, highlighting structured lipids as a promising nutritional strategy to modulate metabolic dysfunction. To evaluate whether capric acid (C10)-based medium-long-medium (MLM) structured lipids can remodel hepatic lipid metabolism and promote the reversal of established MASLD in a diet-induced obese mouse model

Nutritional status of healthcare professionals in primary health and social care.

Balanced nutrition is crucial for healthcare workers' health and performance. Despite extensive knowledge of healthy eating, they face organizational and behavioral barriers leading to suboptimal diets. This study aimed to compare anthropometric status and dietary intake among male physicians, female physicians, and nurses in primary healthcare, and to identify deficiencies or excesses of key nutrients. A descriptive cross-sectional study was conducted in six clinics in Karaganda (2023-2024), involving 202 participants (22 male physicians, 67 female physicians, and 113 nurses). Body compositio

The Changes in Microbiotic Composition of Different Intestinal Tracts and the Effects of Supplemented Lactobacillus During the Formation of Goose Fatty Liver.

Intestinal bacteria play an important role in the formation of fatty liver in animals by participating in the digestion and degradation of nutrients, producing various metabolites, and altering the barrier effect of the intestine. However, changes in the gut microbiota during the formation of goose fatty liver are unclear. In this study, 80 healthy Landes geese with similar body weights at 70 days of age were randomly divided into two groups: the control group (n = 48; fed ad libitum) and the overfeeding group (n = 32; overfed). The intestinal contents were collected at 0, 12, and 24 days of o

Spastin binds to lipid droplets and affects lipid metabolism.

Mutations in SPAST, encoding spastin, are the most common cause of autosomal dominant hereditary spastic paraplegia (HSP). HSP is characterized by weakness and spasticity of the lower limbs, owing to progressive retrograde degeneration of the long corticospinal axons. Spastin is a conserved microtubule (MT)-severing protein, involved in processes requiring rearrangement of the cytoskeleton in concert to membrane remodeling, such as neurite branching, axonal growth, midbody abscission, and endosome tubulation. Two isoforms of spastin are synthesized from alternative initiation codons (M1 and M8

Preterm birth and later insulin resistance: effects of birth weight and postnatal growth in a population based longitudinal study from birth into adult life.

AIMS/HYPOTHESIS: An increased risk of type 2 diabetes mellitus is associated with low birthweight after full-term gestation, including amplification of this risk by weight gain during infancy and adult body composition. Premature birth is also associated with insulin resistance, but studies conducted so far have not provided follow-up into adulthood. We studied the effects of (1) lower birthweight (as standard deviation score [SDS]) and infancy weight gain on insulin resistance in 19-year-olds born before 32 weeks of gestation, and (2) the interaction between lower birthweight SDS and infancy

Adult mice are unresponsive to AAV8-Gremlin1 gene therapy targeting the liver.

OBJECTIVE: Gremlin 1 (GREM1) is a secreted BMP2/4 inhibitor which regulates commitment and differentiation of human adipose precursor cells and prevents the browning effect of BMP4. GREM1 is an insulin antagonist and serum levels are high in type 2 diabetes (T2D). We here examined in vivo effects of AAV8 (Adeno-Associated Viral vectors of serotype eight) GREM 1 targeting the liver in mature mice to increase its systemic secretion and also, in a separate study, injected recombinant GREM 1 intraperitoneally. The objective was to characterize systemic effects of GREM 1 on insulin sensitivity, glu

Establishment of type 2 diabetes mellitus models using streptozotocin after 3 months high-fat diet in Bama minipigs.

In the past twenty years, the number of adults with diabetes has tripled. Most studies have been conducted using rodent models of type 2 diabetes mellitus (T2DM), and the developed drugs have low clinical conversion efficiency. Therefore, it is urgent to establish a more human-like large animal model to explore T2DM pathogenesis and formulate new disease prevention and control strategies. This study was designed to establish and validate a T2DM model using minipigs fed a high-fat or high-cholesterol/high-fat diet and injected with low-dose streptozotocin (STZ). We examined the influence of the

Combined exercise for body composition and cardiometabolic health in obese young people meta analysis.

Combined exercise (CE) may offer benefits beyond aerobic exercise (AE) alone for individuals with overweight or obesity, but comparative evidence remains uncertain. We performed a systematic review and meta-analysis of randomized trials comparing CE with AE in adolescents and young adults aged 6-45 years. Twenty-nine studies involving 1,383 participants were included. Compared with AE, CE was associated with greater improvements in lean body mass and insulin-resistance indicators, including insulin and HOMA-IR. No consistent between-group differences were observed for body weight, BMI, body fa

Weight bias internalization in treatment-seeking overweight adults: Psychometric validation and associations with self-esteem, body image, and mood symptoms.

Internalized weight bias has been previously associated with impairments in eating behaviors, body image, and psychological functioning. The present study explored the psychological correlates and psychometric properties of the Weight Bias Internalization Scale (WBIS) among overweight adults enrolled in a behavioral weight loss program. Questionnaires assessing internalized weight bias, anti-fat attitudes, self-esteem, body image concern, and mood symptoms were administered to 90 obese or overweight men and women between the ages of 21 and 73. Reliability statistics suggested revisions to the

Dose adjustment of anaesthetics in the morbidly obese.

Anaesthesiologists must be prepared to deal with pharmacokinetic and pharmacodynamic (PD) differences in morbidly obese individuals. As drug administration based on total body weight can result in overdose, weight-based dosing scalars must be considered. Conversely, administration of drugs based on ideal body weight can result in a sub-therapeutic dose. Changes in cardiac output and alterations in body composition affect the distribution of numerous anaesthetic drugs. With the exception of neuromuscular antagonists, lean body weight is the optimal dosing scalar for most drugs used in anaesthes

Apolipoprotein D modulates obesity-linked inflammation and metabolic syndrome development in a sex dependent manner.

Obesity, or the excessive and often deleterious accumulation of body fat, is a disease that has gained in prevalence over time. This fat accumulation is often the starting point of other obesity-related pathology, such as low-grade systemic inflammation, insulin resistance, glucose intolerance, hepatic steatosis, behavioral changes including increased anxiety and depression, renal dysfunction, and cardiovascular problems, all of which are costly on a personal and societal level. Apolipoprotein D (ApoD), a circulating lipocalin that accumulates in largely in the liver, has been implicated in a

Nonpharmacological Childhood Obesity Management in Denmark Reduces Steatotic Liver Disease and Obesity.

Background: Steatotic liver disease (SLD) represents a multisystem disease and is a common complication of childhood obesity. We studied fat content at the abdominal level (liver, subcutaneous, and visceral) and the response to childhood obesity management. Methods: In this retrospective longitudinal study, 8-18-year-olds with a body mass index (BMI) z-score above 1.28 (corresponding to a BMI above the 90th percentile), as a proxy for obesity, were offered person-centered, family-oriented obesity management in a hospital setting and in a magnetic resonance (MR) scan. Liver fat content (LFC) wa

Understanding Metabolic Alterations in Cancer Cachexia through the Lens of Exercise Physiology.

Cancer cachexia is one of the leading causes of mortality for late-stage cancer patients. One of its key characteristics is abnormal metabolism and loss of metabolic flexibility, i.e., loss of ability to switch between use of fats and carbohydrates as needed. Here, it is hypothesized that late-stage systemic cancer creates a chronic resource drain on the body that may result in the same metabolic adaptations that occur during intense endurance exercise, activating some of the same mechanisms of nutrient consumption that are supposed to be transient during strenuous physical activity. This hypo

Erythritol Ameliorates Small Intestinal Inflammation Induced by High-Fat Diets and Improves Glucose Tolerance.

BACKGROUND: Erythritol, a sugar alcohol, is widely used as a substitute for sugar in diets for patients with diabetes or obesity. METHODS: In this study, we aimed to investigate the effects of erythritol on metabolic disorders induced by a high-fat diet in C57BL/6J mice, while focusing on changes in innate immunity. RESULTS: Mice that were fed a high-fat diet and administered water containing 5% erythritol (Ery group) had markedly lower body weight, improved glucose tolerance, and markedly higher energy expenditure than the control mice (Ctrl group) (n = 6). Furthermore, compared with the Ct

Association of 29C>T polymorphism in the transforming growth factor-β1 gene with lean body mass in community-dwelling Japanese population.

AIM: Sarcopenia is the significant degenerative loss of skeletal muscle mass and strength associated with aging, and it is one of the components of frailty. We previously reported an association between the 29C>T polymorphism in the transforming growth factor-β1 gene (rs1800470) and the prevalence of vertebral fractures in subjects with postmenopausal osteoporosis. The association was not attributable to bone mineral density, which suggests that polymorphism influences some aspects of bone quality that affects strength and/or frailty rather than bone strength itself. Thus, we examined the rela
