Evidence for high activity of xylem parenchyma and ray cells in the interface of host stem and Agrobacterium tumefaciens-induced tumours of Ricinus communis.

Rapidly developing tumours at hypocotyls of Ricinus communis, induced by Agrobacterium tumefaciens strain C58, were characterized by strong differentiation of vascular bundles and their functional connection to the host bundles. The stem/tumour interface showed increased xylem, with numerous vessels accompanied by multiseriate unlignified rays. To know how nutrients efficiently accumulate in the tumour sink tissue, cell electropotentials (E(m)) in cross-sections were mapped. The measured cells were identified by injected Lucifer Yellow. Xylem and phloem parenchyma cells and stem/tumour-located

No evidence for an association between the Glu298Asp polymorphism of the NOS3 gene and Alzheimer's disease.

Recently a significant association of a missense mutation (Glu298Asp) of the endothelial nitric oxide synthase (NOS3) gene with late-onset Alzheimer's disease (LOAD) was reported. We tried to replicate this finding in a Japanese sample of 121 patients with LOAD, 51 with early-onset AD (EOAD), and 165 medical controls. However, the genotype and allelic distributions for the Glu298Asp polymorphism were similar for these three groups, suggesting that the Glu298Asp polymorphism of the NOS3 gene has no relevance to the development of AD in Japanese.

Identification of three novel mutations (E196K, V203I, E211Q) in the prion protein gene (PRNP) in inherited prion diseases with Creutzfeldt-Jakob disease phenotype.

Inherited prion diseases are characterized by mutations in the PRNP gene encoding the prion protein (PrP). As the other sporadic or infectious prion disease forms, they are almost all characterized by the accumulation in the brain of an abnormal misfolded form of the patient's PrP. Brain extracts can often transmit the disease once inoculated in a recipient animal. Inherited prion diseases with Creutzfeldt-Jakob disease (CJD) phenotype are autosomal forms, although sporadic cases have been reported. We report three novel mutations of the PRNP gene in unrelated patients with clinical and histop

Phrenic nerve responses to chemical stimulation of the subregions of ventral medullary respiratory neuronal group in the rat.

Phrenic nerve (PN) responses to unilateral microinjections of L-glutamate (L-Glu, 5 mM) or N-methyl-D-aspartic acid (NMDA, 1 mM) into different subregions of ventral respiratory neuronal group (VRG) were studied in urethane-anesthetized, immobilized, and artificially ventilated, adult male Wistar rats. A 50-nl volume of microinjection was used in all the subregions of VRG except in Pre-Bötzinger complex (Pre-BötC) where a 20-nl volume was used. Unilateral microinjections of L-Glu or NMDA into the Bötzinger complex (BötC) and caudal VRG (cVRG), caused a transient cessation of phrenic nerve (PN)

Two regions in the isolated brainstem of the frog that modulate respiratory-related activity.

Using microinjection techniques, we have explored the isolated, complete midline sectioned brainstem of the frog (Rana catesbeiana) to identify regions that influence the endogenous respiratory-related motor activity. Ten-nanoliter injections of lidocaine (1%), GABA (100mM) and glutamate (10 and 100 mM) into discrete regions of the rostral and the caudal brainstem produced different effects on the phasic neural discharge. In the rostral site lidocaine, GABA and glutamate injections altered neural burst frequency with little or no effect on burst amplitude. In the caudal site, responses to lido

Investigation of in-vivo GABA+ and Glutamate levels in patients with drug resistant unilateral temporal lobe epilepsy using MEGA-PRESS and correlates with clinical semiology.

BACKGROUND: In this study, we used in vivo MEGA-PRESS spectroscopy to investigate GABA and Glu levels in patients with drug-resistant unilateral temporal lobe epilepsy (TLE) and also look for correlation between neurometabolites and clinical semiology assessed by other modalities. MATERIALS AND METHODS: Twenty-five patients with TLE underwent MRS data acquisition on a 3.0 T MRI. Bilateral single-voxel MEGA-PRESS data was acquired from mesial temporal lobes, and data was post-processed using MRSpa and LC-Model for computing neurometabolites concentration. Tissue segmentation from spectroscopic

Kainic acid-induced status epilepticus decreases mGlu5 receptor and phase-specifically downregulates Homer1b/c expression.

Globally, over 50 million people are affected by epilepsy, which is characterized by the occurrence of spontaneous recurrent seizures. Almost one-third of the patients show resistance to current anti-epileptic drugs, making the exploration of new molecular targets necessary. An interesting target may be Homer1, due to its diverse roles in epileptogenesis and synaptic plasticity. Indeed, Homer1 regulates group I metabotropic glutamate (mGlu) receptors (i.e. mGlu1 and mGlu5) scaffolding and signaling in neurons. In the present work, using the systemic kainic acid (KA)-induced status epilepticus

Brain glutamate in anorexia nervosa: a magnetic resonance spectroscopy case control study at 7 Tesla.

RATIONALE: Anorexia nervosa (AN) is a serious psychiatric disorder with high morbidity and mortality. There are no established pharmacological treatments and the neurobiology of the condition is poorly understood. Previous studies using magnetic resonance spectroscopy (MRS) have shown that AN may be associated with reductions in indices of brain glutamate; however, at conventional field strengths (≤3 T), it is difficult to separate glutamate from its precursor and metabolite, glutamine. OBJECTIVES: The objective of the present study was to use high field (7 T) MRS to measure concentrations of

Interleukin-1beta promotes oligodendrocyte death through glutamate excitotoxicity.

Glutamate excitotoxicity is implicated in the progressive loss of oligodendrocytes in multiple sclerosis, but how glutamate metabolism is dysregulated in the disease remains unclear. Because there is microglia activation in all stages of multiple sclerosis, we determined whether a microglia product, interleukin-1beta, could provide the mechanism for glutamate excitotoxicity. We found that whereas interleukin-1beta did not kill oligodendrocytes in pure culture, it produced apoptosis of oligodendrocytes in coculture with astrocytes and microglia. This requirement for a mixed glia environment sug

Astrocytes from Na(+)-K(+)-Cl(-) cotransporter-null mice exhibit absence of swelling and decrease in EAA release.

We reported previously that inhibition of Na(+)-K(+)-Cl(-) cotransporter isoform 1 (NKCC1) by bumetanide abolishes high extracellular K(+) concentration ([K(+)](o))-induced swelling and intracellular Cl(-) accumulation in rat cortical astrocytes. In this report, we extended our study by using cortical astrocytes from NKCC1-deficient (NKCC1(-/-)) mice. NKCC1 protein and activity were absent in NKCC1(-/-) astrocytes. [K(+)](o) of 75 mM increased NKCC1 activity approximately fourfold in NKCC1(+/+) cells (P < 0.05) but had no effect in NKCC1(-/-) astrocytes. Intracellular Cl(-) was increased by 70

Memantine suppresses the glutamatergic neurotransmission of mammalian inner hair cells.

The glutamatergic synapses between inner hair cells and afferent neurons seem to be involved in pathophysiological conditions of the cochlea. The excessive release of glutamate from inner hair cells during noise trauma and ischemia affects the afferent neurons. It is possible that in tinnitus outer hair cell or inner hair cell dysfunction or damage leads to an altered spontaneous release of glutamate from inner hair cells. Thus, the pharmacological modulation of glutamatergic neurotransmission could be of great value in the therapy of certain inner ear diseases. Recently, it has been discovere

A study of the nephrotoxicity of three cephalosporins in rabbits using 1H NMR spectroscopy.

Male New Zealand White rabbits received a single intravenous injection of 125 mg/kg cephaloridine, 500 mg/kg cefoperazone or 1000 mg/kg cephalothin. Histological examination of kidneys at 48 h post-dose confirmed the presence of bilateral necrosis of the proximal convoluted tubules in the cephaloridine-treated animals. 1H-NMR urinalysis of cephaloridine-treated rabbits detected drug-related resonances, decreased hippurate and increased glucose at 0-24 h post-dose accompanied by elevated levels of lactate, glycine, citrate, glutamine/glutamate and alanine at 24-48 h post-dose. No histopathologi

Spiny dogfish, Squalus suckleyi, shows a good tolerance for hypoxia but need long recovery times.

Pacific spiny dogfish, Squalus suckleyi, move to shallow coastal waters during critical reproductive life stages and are thus at risk of encountering hypoxic events which occur more frequently in these areas. For effective conservation management, we need to fully understand the consequences of hypoxia on marine key species such as elasmobranchs. Because of their benthic life style, we hypothesized that S. suckleyi are hypoxia tolerant and able to efficiently regulate oxygen consumption, and that anaerobic metabolism is supported by a broad range of metabolites including ketones, fatty acids a

Anticonvulsant potential of Grewia tiliaefolia in pentylenetetrazole induced epilepsy: insights from in vivo and in silico studies.

Epilepsy, a chronic neurological condition, impacts millions of individuals globally and remains a significant contributor to both illness and mortality. Available antiepileptic drugs have serious side effects which warrants to explore different medicinal plants used for the management of epilepsy reported in Traditional Indian Medicinal System (TIMS). Therefore, we explored the antiepileptic potential of the Grewia tiliaefolia (Tiliaeceae) which is known for its neuroprotective properties. Aerial parts of G. tiliaefolia were subjected to extraction with increasing order of polarity viz. hexan

Glutamate binding perturbs structure-function integrity of carbonic anhydrase and promotes its aggregation: Implications of dysregulated metabolite levels in neurodegeneration.

Metabolite dysregulation is increasingly recognized as a crucial factor in protein misfolding and aggregation underlying various neurodegenerative diseases. Among these, glutamate, the most abundant excitatory neurotransmitter, is strongly regulated under physiological conditions but reaches pathological concentrations in most of the neurodegenerative disorders. In this context, the present study investigated how glutamate modulates the structure-function integrity of carbonic anhydrase (CA), a neuronal enzyme implicated in brain homeostasis, pH regulation, and cognitive processes. Pharmacological and Physiological Characteristics of Synaptic Transmissions From the Medial Prefrontal Cortex Onto Noradrenergic Neurons and Inhibitory Interneurons in the Mouse Locus Coeruleus.

The locus coeruleus (LC) is the brain's primary source of norepinephrine. It modulates the brain's overall state of arousal. Recent evidence suggests that the LC also regulates immediate attentional responses by resetting related cortical networks to optimize behavioral outcomes. Cortical regions associated with high cognitive function, such as the medial prefrontal cortex (mPFC), are theorized to influence LC output directly to regulate behavior. However, the available evidence is insufficient for a comprehensive understanding of the underlying mechanisms and properties. To address this gap i

Hippocampal Glutamatergic Hyperactivation Mediates High-Loading Intensity of Exercise-Induced Cognitive Deficits Via HPC-mPFC Circuit Dysfunction.

AIMS: Excessive exercise impairs cognition and elevates late-life cognitive risk. However, the underlying mechanisms remain unclear. This study investigates the neural mechanisms through which high-intensity endurance exercise induces cognitive deficits. METHODS: Mice underwent high-loading intensity exercise (HLIE) with a 7-day treadmill procedure (25 m/min, 90 min/day). A battery of behavioral tests was conducted to assess cognitive performance, including the Morris Water Maze, Novel Object Recognition, and Y Maze. fMRI and c-Fos activity mapping were employed to identify key brain regions

Layer-specific variation in intracellular GABA immunoreactivity in the neuronal organization of the mouse main olfactory bulb.

The main olfactory bulb (MOB) hosts diverse γ-aminobutyric acid (GABA)ergic interneurons that play critical roles in sensory processing. Previous studies have reported variability in GABA immunoreactivity among cells in the granule cell layer (GCL) of the mouse MOB. However, the degree of intracellular GABA content across MOB layers and its relationship with molecular subtypes remain unclear. In this study, we conducted a quantitative analysis of GABA immunoreactivity in individual cells across all MOB layers using confocal laser scanning and electron microscopy. We found striking variability

Cell Type-Specific Encoding of Cocaine-Conditioned Responses in the Lateral Preoptic Area.

The lateral preoptic area (LPO) is a functionally heterogeneous hypothalamic structure that is increasingly recognised for its role in motivated and drug-seeking behaviours. Although prior studies have shown that LPO neurons exhibit diverse activity patterns during cocaine self-administration, the specific contributions of glutamatergic and GABAergic populations to conditioned responses to cocaine remain unclear. In this study, we recorded the activity of LPO glutamatergic (vglut2-expressing) and GABAergic (vgat-expressing) neuronal subpopulations during cocaine conditioning to identify how th
