Effects of chronic treatment with corticosterone and imipramine on fos immunoreactivity and adult hippocampal neurogenesis.

In a previous study we showed that rats chronically treated with corticosterone (CORT) display anxiogenic behavior, evidenced by facilitatio … We also evaluated the number of cells expressing the neurogenesis marker doublecortin (DCX) in the hippocampus and measured plasma CORT levels on the 21st day of treatment. Results showed that CORT increased fos-ir in the ventrolateral septum, medial amygdala and paraventricular hypothalamic nucleus and decreased fos-ir in the lateral periaqueductal gray. Im …

IL-6 and PACAP receptor expression and localization after global brain ischemia in mice.

Pituitary adenylate cyclase-activating polypeptide (PACAP) exerts a neuroprotective action against ischemic damage. This action is mediated by the interleukin-6 (IL-6) pathway. However, as the expression patterns of PACAP receptors and IL-6 following ischemia are not understood, we evaluated them in the mouse hippocampus in response to ischemia induced by bilateral common carotid artery occlusion. Real-time PCR determination of PAC1R mRNA expression in the hippocampus was significantly elevated on day 7 after ischemia. VPAC1R mRNA expression was significantly decreased 3 days after the ischemi…

The effects of bilateral common carotid artery occlusion on expression of peripherin and choline acetyltransferase activity in C57BL/6 mice.

Peripherin is a 57-kDa type III neuronal intermediate filament protein that appears to play a role in neurite elongation during development … and Western Blot assay of brain sections. Peripherin expression increased in neurons of the cortex, hippocampus, and thalamus after BCCAO. By double immunofluorescence staining, neurons showed a cytoplasmic co-localization of peripherin and MAP-2, but not of peripherin and GFAP. ChAT activity was determined spectrophotometrically using the assay kit. There wa …

Multisynaptic inputs from the medial temporal lobe to V4 in macaques.

Retrograde transsynaptic transport of rabies virus was employed to undertake the top-down projections from the medial temporal lobe (MTL) to … mber of neurons were labeled in other regions, such as the subicular complex and CA1 and CA3 of the hippocampus proper. The present results indicate that V4 receives major input from the hippocampus proper via the entorhinal cortex, as well as "short-cut" pathways that bypass the entorhinal cortex. These multisynaptic pathways may define an anatomical basis f …

Expression of NR2B in different brain regions and effect of NR2B antagonism on learning deficits after experimental subarachnoid hemorrhage.

Approximately 50% of patients who survived after aneurysmal subarachnoid hemorrhage (SAH) have cognitive or neurobehavioral dysfunction. The mechanisms are not known. NR2B, one of the subunits of N-methyl-d-aspartate (NMDA) receptors, has been proved to be an important factor for synapse function and behavior cognition. Experiment 1 aimed to investigate the timecourse of the NR2B expression in the cortex, hippocampus, and cerebellum after SAH in rats. In experiment 2, we assessed the effect of Ro 25-6981 (a specific NR2B antagonist) on regulation of learning deficits and behavioral activity fo…

Imipramine treatment reverses depressive-like behavior in alloxan-diabetic rats.

BACKGROUND: A growing body of evidence has shown an association between diabetes and depression, as well a role of brain-derived neurotrophic factor (BDNF) in diabetes and depression. The present study was designed to evaluate the behavioural and molecular effects of the anti-depressant imipramine in diabetic rats. METHODS: To this aim, after induction of diabetes by alloxan (150 mg/kg), Wistar rats were treated with imipramine (30 mg/kg) once a day for 14 days and then subjected to behavioural tests. BDNF was then assessed in the prefrontal cortex, hippocampus and amygdala. RESULTS: In diab…

Distribution of pH changes in mouse neonatal hypoxic-ischaemic insult.

We assessed the distribution in brain pH after neonatal hypoxic-ischaemic insult and its correlation with local injury. Postnatal day 7 mice were injected with neutral red and underwent left carotid occlusion and exposure to 8% oxygen. Images captured from the cut surface of snap-frozen brain were used to calculate the pH from the blue-green absorbance ratios. Carotid occlusion alone had no effect, but combined with hypoxia caused rapid, biphasic pH decline, with the first plateau at 15-30 min, and the second at 60-90 min. The ipsilateral dorsal cortex, hippocampus, striatum and thalamus were …

Effects of high-speed railway noise on the synaptic ultrastructure and phosphorylated-CaMKII expression in the central nervous system of SD rats.

To investigate the toxic effects of high-speed railway noise on learning and memory function, Sprague-Dawley (SD) rats were exposed to high-speed railway noise for 90 days. The noise was recorded from an actual environment and adjusted to a day-night equivalent continuous A-weighted sound pressure level (L(dn)) of 70dB(A). Transmission electron microscopy (TEM) and Western blot analysis were used to observe the synaptic ultrastructure and detect the level of phosphorylated-Ca(2+)/calmodulin-dependent protein kinase II (p-CaMKII), respectively, in the hippocampus, temporal lobe and amygdala. Co…

Profound retrograde but absence of anterograde amnesia for cued place learning in rats with hippocampal lesions.

Previous studies in our lab have shown that slight modifications in the spatial reference memory procedure can overcome the deficit in spati … amnesia. Thus, although the absence of anterograde amnesia suggests that structures other that the hippocampus can take charge of the acquisition, the presence of retrograde amnesia indicates the critical role of the normal hippocampus in the long-term formation of allocentric information.

Neuropathological correlates of late-life depression.

The relationship between depression and both dementia and cerebrovascular pathology has, for good reason, received much attention from resea … erebrovascular disease. There were associations with Lewy body pathology and loss of neurons in the hippocampus and other subcortical areas. Although the authors are cautious with regard to drawing firm conclusions, the results suggest that, in the community, depression is not an important etiological factor for the development of the neuropathology of Alzhei …

Acoustic startle hypersensitivity in Mceph mice and its effect on hippocampal excitability.

Current therapies and research for epilepsy concentrate mainly on controlling the disease, but not on prevention of its development and prog … strocytes, as indicated by glial fibrillary acidic protein, occurred in the inferior colliculus and hippocampus. Both the hypersensitivity of acoustic startle as well as activation of astrocytes could be maintained at WT levels by pre-treating the Mceph mice with the anti-epileptic drug valproate. Down-regulation of hippocampal BDNF and Arc associated with improvement in aversive spatial memory performance in socially isolated rats.

Rats deprived of social contact with other rats at a young age experience a form of prolonged stress that leads to long-lasting changes in b … ived neurotrophic factor (BDNF) and activity-regulated cytoskeletal associated protein (Arc) in the hippocampus. In the Morris water maze, isolated rats showed a reduced latency to reach the hidden platform during training, indicative of an improved learning performance, compared with group-housed rats. Altered expression of neuropeptides in the primary somatosensory cortex of the Down syndrome model Ts65Dn.

Down syndrome is the most common genetic disorder associated with mental retardation. Subjects and mice models for Down syndrome (such as Ts65Dn) show defects in the formation of neuronal networks in both the hippocampus and the cerebral cortex. The principal neurons display alterations in the morphology, density and distribution of dendritic spines in the cortex as well as in the hippocampus. Several evidences point to the possibility that the atrophy observed in principal neurons could be mediated by changes in their inhibitory inputs and, in fact, an imbalance between excitation and inhibit…

Neural stem cells reduce hippocampal tau and reelin accumulation in aged Ts65Dn Down syndrome mice.

Tau accumulation, in the form of neurofibrillary tangles (NFT), is an early neuropathological characteristic of Alzheimer's disease (AD) and early onset AD frequently seen in Down syndrome (DS). We investigated the presence of tau accumulation in the brains of aging DS mice using the Ts65Dn mouse model. All aged mice appeared to have substantial clusters of extracellular granules that were positive for tau and reelin, but not for amyloid-β or APP. These clusters were found primarily in CA1 of the hippocampus. In addition, the aged trisomic DS mice had a significantly greater accumulation of ex…
