Social presence and its role in social interactions: Insights, challenges and a call for action.

Over the past decades, we have seen a fundamentally new form of social interaction become more and more common in our everyday lives: mediated social interactions, in which we do not speak to a person face-to-face, but the interaction takes place via a video call or some other form of technology. While most people would agree that a mediated interaction is somehow different from a face-to-face interaction, we currently lack a framework for conceptualizing what exactly is different. This is especially important when studying how mediated interactions shape our understanding of others' mental st

Targeting PSAT1 in diabetic kidney disease: a ferroptosis-driven strategy for precision therapy.

Diabetic kidney disease (DKD) involves mitochondrial dysfunction and ferroptosis, yet the convergence of these processes at the gene level remains unclear. We first analyzed human renal transcriptomic data (GSE30528) to identify differentially expressed genes (DEGs). DEGs associated with both mitophagy and ferroptosis (DEMFRGs) were extracted. Protein-protein interaction (PPI) networks, the MCC algorithm, and LASSO regression were used to screen hub genes. Immune infiltration was assessed via CIBERSORT. Diagnostic performance of hub genes was evaluated using ROC curves in GSE30528 and validate

Applicability of next-generation risk assessment generic PBK models and their performance within the chemical domain.

Physiologically based kinetic (PBK) models are a cornerstone of in vitro to in vivo extrapolation and are therefore central to next-generation risk assessment (NGRA). This study assesses the performance of a generic, high-throughput PBK model for 26 organic chemicals spanning a wide range of physicochemical properties. Model predictions based on standard in silico and in vitro input parameters were benchmarked against published, chemical-specific PBK models and in vivo kinetic data. Predicted plasma maximum concentrations (Cmax) were within tenfold of in vivo values for 50% of the chemicals an

Hofmeister-assisted electro-fabrication of Janus gelatin hydrogels with tunable structure and biofunctionality.

Natural vessel structures inspire Janus hydrogels with spatially separated functions. Here, we report a one-step electro-fabrication strategy to construct gelatin-based Janus hydrogels featuring a dense, hydrophobic inner layer and a porous, hydrophilic outer layer. Introducing sodium citrate generates a directional ion gradient under an electric field, triggering interface-directed gelation through the Hofmeister effect and enhancing anticoagulant performance. The layered architecture can be finely tuned by varying voltage, deposition time, and gelatin concentration, producing inner pores bel

Manipulation of nonclassical ubiquitin signaling by pathogenic bacteria.

Manipulation of the host ubiquitin signaling system is emerging as a common and critical mechanism of virulence among pathogenic bacteria. In some cases, such as the intracellular pathogens Legionella pneumophila and Shigella flexneri, ubiquitin regulation is the focus of a significant proportion of the bacterial repertoire of secreted effectors (10% and 30%, respectively). While many of these bacterial effectors hijack classical aspects of the ubiquitin system, such as proteasomal degradation and immune signaling, others target nonclassical ubiquitin signals that are less well understood. The

Hydrophilicity engineering of amphiphilic macrocyclic MRI contrast agents for enhanced hepatic targeting efficiency.

Effective hepatic enhancement and high kinetic inertness are pivotal driving forces for developing gadolinium-based hepatobiliary MRI contrast agents (CAs). Although lipophilic modifications have dominated efforts to promote hepatic uptake, the vital role of hydrophilicity has been overlooked. Herein, we investigated the structure-activity relationship by fine-tuning of an amphiphilic macrocyclic complex through hydrophilicity engineering. The water-soluble Gd-HE(BnOPh)-DO3A exhibited exceptional kinetic inertness with a dissociation half-life of 8.3 h at pH 1.2, nearly 5-fold longer than the

Enhanced Intramuscular mRNA Activity of Peptide-Lipid Nanoparticles with Hydrophobized Head Groups.

A library of xenopeptide-based ionizable lipids (XP-ILs) was evaluated in mRNA lipid nanoparticles (LNPs) to investigate the impact of modifications within the polar oligoamine headgroup. Modifications included hydrophobic or bulky side chains or a reduced number of protonatable amines. The resulting XP-ILs were characterized by their key physicochemical properties (logD, logP, HLB, CMC), which were correlated with LNP transfection efficiency and cytotoxicity. The inverse relationship between computational logP and experimental logD5.5 is consistent with the HLB, indicating a lower tendency fo

Live imaging captures the events immediately following fertilization.

Fertilization involves dynamic sperm-egg interactions, yet has been primarily studied in static samples. Here, we use high-resolution live imaging to capture fertilization from the moment of sperm binding in zona-intact mouse oocytes. We identify two phases of sperm remodeling: a static phase, during which sperm remain beneath the oocyte cortex as DNA decondensation and histone loading occur, and a mobile phase characterized by stereotyped sperm movement. Initial displacement away from the spindle is driven by cytoplasmic streaming, with manipulations in mouse indicating that sperm movement re

Host-microbiome-immune disequilibrium in oral disease: mechanisms, dysbiosis, and precision therapeutics.

BACKGROUND: The oral cavity harbors a dynamic microbial ecosystem that interacts with epithelial barriers, host immunity, and local tissue environments. Disruption of this balance is increasingly recognized as a key driver of major oral diseases, including periodontitis, dental caries, and oral squamous cell carcinoma (OSCC). However, the biological links between microbial ecology, immune regulation, and disease progression are insufficiently integrated, limiting mechanistic understanding and translational progress. METHODS: This structured narrative review searched PubMed/MEDLINE, Web of Sci

Clathrins Are Involved in the Endocytosis of Host Cytosol in the Malaria Parasite.

In eukaryotic cells, clathrins interact with the adaptor protein (AP) complex-2 (AP-2) to facilitate endocytosis and AP-1 to mediate secretion and trafficking between the endosome and Golgi. In Plasmodium falciparum, recent studies revealed that the Kelch domain-containing protein 13 and AP-2 participate in hemoglobin uptake via cytostomes. However, clathrins appear not to be involved in this process because they primarily associate with AP-1. To investigate the roles of clathrins in P. falciparum, we characterized the clathrin heavy chain (PfCHC), the clathrin light chain (PfCLC), and the AP-

Pathogenicity and virulence of Candida auris.

Candida auris (Candidozyma auris) has emerged as a multidrug‑resistant human fungal pathogen that causes infections of high morbidity and mortality. Notably, it exhibits a unique ability to grow and persist on human skin, thus leading to efficient transmission through skin-to-skin contact. As a result, C. auris poses a significant risk of outbreaks in healthcare settings, especially in nursing homes that care for elderly patients. Most concerning, C. auris clinical isolates demonstrate widespread and, in some cases, untreatable resistance to all antifungal drug classes, including azoles, polye

Candida auris skin colonization: mechanisms, microecological interactions, and emerging decolonization strategies.

Candida auris, as an emerging multidrug-resistant pathogen, has become a major threat to global public health due to its high mortality rate, strong transmissibility, and propensity to cause outbreaks in healthcare settings. Persistent skin colonization is a key factor contributing to the high transmissibility of C. auris and represents one of its important distinguishing features from other Candida species. However, the mechanisms underlying C. auris skin colonization remain poorly understood, and relevant prevention and control strategies are still underdeveloped. This article aims to system

The skin microbiome as a key regulator of psoriasis: From dysbiotic drivers to host-directed therapeutics.

The skin harbors diverse microbial communities. Compared with gut microbiome dysbiosis, perturbations in the skin microbiome may be more directly implicated in the onset and progression of psoriasis (PSO). More than twenty studies reveal microbial dysbiosis within psoriatic lesions, which is manifested by elevated abundances of pro-inflammatory taxa such as Corynebacterium and Staphylococcus aureus, declined levels of commensal microbes including Cutibacterium, increased total bacterial load and decreased microbial community diversity. Such cutaneous microbial dysbiosis accelerates disease pro

Targeting MLKL-Driven Necroptosis: A Therapeutic Target in Inflammation and Host Defense.

Necroptosis is a regulated form of programmed cell death that helps the body defend itself against infections and cellular stress, especially when apoptosis is blocked. At the center of this process is mixed lineage kinase domain-like (MLKL) protein, the final effector of necroptosis, which is activated downstream of receptor-interacting protein kinase 3 (RIPK3). Once phosphorylated, MLKL changes shape, assembles into oligomers, moves to cellular membranes, and disrupts membrane integrity, ultimately causing cell death. While this RIPK3-MLKL pathway has been well described, it is becoming incr

Bacterial small RNAs and extracellular vesicles in inflammatory and osteoimmune diseases: regulatory mechanisms and host-microbe communication.

Bacterial small RNAs (sRNAs) functions as post-transcriptional regulators that influnce microbial adaptation, pathobiont behavior, and host-microbe communication. This review analyzes bacterial sRNAs and vesicle-associated RNA signals in inflammatory and osteoimmune diseases, with a focus on gastrointestinal and oral microbial communities. Major sRNA classes and mechanisms are summarized, followed by an evaluation of evidence in gastrointestinal inflammation, periodontitis, rheumatoid arthritis, osteoarthritis, and postmenopausal osteoporosis. Direct disease-related evidence is currently limit

The Liver Fluke Opisthorchis felineus Exosomal tRNA-Derived Small RNAs as Potential Mediators of Host Manipulation.

The role of extracellular vesicle non-coding RNAs in host-parasite interactions remains poorly understood, particularly for human liver flukes. Although tRNA-derived small RNAs (tsRNAs) are emerging as new regulatory molecules in parasite exosomes, they have not yet been characterized for the liver flukes. We performed small RNA sequencing to profile tsRNAs in the exosome-like vesicles derived from the liver fluke Opisthorchis felineus. Transcriptomic data from human cholangiocytes were analyzed to assess the enrichment of the predicted target genes among differentially expressed genes. We ide

Mechanisms of Immune Evasion, Dissemination, and Persistence in Leishmania amazonensis Infection.

Leishmania amazonensis is one of the etiological agents of cutaneous leishmaniasis in its localised form. Moreover, this parasite can cause more severe disease conditions, such as diffuse and disseminated cutaneous leishmaniasis. The development of more severe clinical manifestations is associated with the parasite's ability to establish a chronic infection and disseminate to cutaneous tissues distal to the site of primary infection. This ability is dependent on host immune factors but is also influenced by intrinsic factors of the parasite related to pathogenicity and evasion of the immune sy

Hepatocyte Autophagy in Malaria: Current Concepts, Emerging Mechanisms, and Future Therapeutic Directions.

The liver stage of Plasmodium infection represents a critical bottleneck in malaria pathogenesis and a unique interface between parasite development and hepatocyte-intrinsic immunity. Recent evidence suggests that hepatocytes do not eliminate liver-stage parasites through canonical xenophagy, as previously assumed, but instead employ a noncanonical autophagy response known as the conjugation of ATG8 to single membranes (CASM). CASM drives rapid lipidation of LC3 onto the parasitophorous vacuole membrane (PVM) via a V-ATPase-ATG16L1-dependent mechanism, thereby activating the Plasmodium-associa

Regulation and function of macrophages in Toxocara canis infection.

Macrophages are highly plastic and heterogeneous innate immune cells capable of polarizing into either classically activated or alternatively activated phenotypes, depending on the surrounding microenvironment. This review explores the cross-talk between macrophages and Toxocara canis larvae during infection, emphasizing the role of molecules present in Toxocara excretory-secretory products (TESPs) in regulating macrophage function, polarization and survival, as well as their interactions with macrophage proteins. This narrative review was conducted through a targeted search of peer-reviewed l

Beyond the parasite: reframing cutaneous leishmaniasis as a host-microbiome-vector ecosystem.

Disease outcomes of cutaneous leishmaniasis (CL) result from dynamic interactions between host immunity, Leishmania parasites and the host-sandfly microbiomes. Current treatments focus on parasite elimination but are limited by toxicity, drug resistance, side effects and poor wound healing. These challenges highlight the need for multi-targeted interventions by transforming CL treatment from a single-infection model to an integrated host-microbe-parasite model. This review features advances in host-microbe-parasite-vector-based strategies for CL treatment, including emerging engineering and sy
