[The role of hyperprolactinaemia in sterility where the cycle is anovulatory and where there is deficit of progestogen. (Investigations of the pituitary reserves of FSH, LH and prolactin) (author's transl)].

We have measured by radio-immunological assays the levels of prolactin (PRL), of FSH and of LH in the plasma in 7 cases where sterility was due to persistent anovulatory cycles and in 6 cases where there was a progesterone deficiency as the sole aetiological factor. Then we stimulated the release of these three hormones by giving 300 micrograms of TRF, PRL, and 100 micrograms of LH-RH, of FSH and of LH. The results were compared with those obtained in 7 cases of normal young women with clear-cut biphasic cycles used as controls. These results show that a rise in the reserves of prolactin in th

Leukocyte migration enhancement as an indicator of immunologic enhancement. II. Malignancy.

Leukocyte migration studies were performed on 19 cancer patients and 24 control subjects. Amniotic fluid (AF) as well as autologous plasma (PL) were investigated. No difference in the migration pattern between patients and control subjects was observed with AF, but cancer patients showed significant leukocyte migration enhancement (LME) with PL when compared to control subjects (p less than 0.04). Among 18 cancer patients with persistent disease only three did not show significant LME. All three represented specifically immunodepressed states. None of the untreated group of cancer patients fai

Quantitation and characterization of soluble immune complexes precipitated from sera by polyethylene glycol (PEG).

Polyethylene glycol (PEG) was used to isolate immune complexes from sera. Complexes were then quantified and partially characterized by a variety of immunological techniques. Complexes were detected in rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), Felty's syndrome and vasculitis, but not in polymyositis, scleroderma or Behçets syndrome. The level of immune complexes correlated with disease activity in SLE and RA patients. Rheumatoid factors and anti-DNA antibodies were enriched by PEG precipitation of RA and SLE sera, respectively, thus these antibodies appeared to be present

Selective immunological evaluation of mycosis fungoides.

Tests of specific aspects of immune function were conducted on 15 patients with clinically and histologically evident mycosis fungoides of various stages. These tests included peripheral blood lymphocyte counts, T and B cell counts, mitogen stimulation tests to phytohemagglutinin (PHA), concanavalin A (con A), and pokeweed mitogen, as well as determinations of IgE levels and antinuclear factor (ANF). We found that mycosis fungoides patients, as a group, had normal peripheral blood lymphocyte, T cell, and B cell counts, but had decreased stimulation to PHA and con A. When evaluated by stage of

Liver disease and vasculitis in a patient taking cromolyn.

Hypersensitivity reactions to cromolyn sodium occur rarely. On several occasions they have been associated with peripheral eosinophilia and granulomatous inflammation. Liver disease has not been reported previously as a complication of inhaled cromolyn. We describe here a woman in whom marked peripheral eosinophilia, liver disease, and systemic vasculitis developed while taking cromolyn and resolved or improved on discontinuation of the drug and treatment with corticosteroids. The liver disease was similar to primary biliary cirrhosis except that marked eosinophilic infiltration and granulomas

Exposure to dust-borne bacteria in agriculture. II. Immunological survey.

In order to investigate whether high exposure to Erwinia herbicola causes sensitization in grain handlers, immunological tests (agargel precipitation, complement fixation, passive cutaneous anaphylaxis, and intradermal) with the extracts of these bacteria were performed in different groups of grain workers and in other groups of population. Tests with extracts of grain dust and other microorganisms were also performed. Grain workers showed in all the tests high incidence of positive reactions to E. herbicola, being significantly higher, as compared with unexposed subjects. Different actinomyce

X-linked immunodeficiency with increased IgM: clinical, ethnic, and immunologic heterogeneity.

Two cases of immunodeficiency with increased IgM are reported. Patient 1 was a black male 3.5 years old who had recurrent pyogenic infections, failure to thrive, oral thrush, and systemic cryptococcal infection. Patient 2 was a 9-year-old white female who had recurrent cervical abscesses. Serum immunoglobulin determinations by radial immunodiffusion in both patients showed marked depression of IgG and IgA and marked elevation of IgM. A low molecular weight circulating monomeric IgM was demonstrated by immunoelectrophoresis and gel filtration in the second patient; this was not present in the f

Analysis of group B streptococcal types associated with disease in human infants and adults.

It is important to resolve existing differences of opinion regarding group B streptococcal type distribution in human disease because of the relevance of type prevalence to future programs of prevention. This report compares data obtained from typing 392 group B streptococci isolated from systemic infections in both infants and adults in the United States from 1972 through 1975. The data showed a substantial predominance of type III among strains isolated from cases of infant meningitis and from "late-onset" septicemia but did not confirm a prior report that type Ia causes most cases of "early

Reversal reactions in lepromatous leprosy following transfer factor therapy.

Five patients with active leprosy, four with polar lepromatous (LL) and one with borderline lepromatous (BL) disease, were each treated with transfer factor (TF) from approximately 7.4 x 10(9) lymphocytes given in 36 divided doses over a 12-week period. The TF was prepared from blood donated by normal, healthy, lepromin skin test-positive individuals. During treatment all four of the LL patients, but not the BL patient, developed clinical reversal reactions. Histopathologically, skin biopsies in these four LL patients showed evidence of transformation of collections of multibacillary macrophag

Immunologic reconstitution in man--present status.

Recent advances in immunobiology have shed new light on our understanding of the essential role of immunity as it relates to body economy. Immunity, once thought to be the primary defense mechanism against microbial infection, appears to have a much broader function--recognition and elimination of foreign bodies and preservation of the integrity of the individual. As our understanding of the pathophysiology of the immune system became more clear, immunologic reconstitution emerged as a new, promising mode of treatment for a variety of diseases with immunodeficiency. Bone marrow transplantation

Degradation of ACSL3 by influenza A virus shifts unfolded protein response from antiviral defense to viral evasion.

Host restriction factors and viral evasion strategies involved in the virus-host arms races remain to be discovered. Through an initial membrane protein-targeted CRISPR-Cas9 screening, we identified Acyl-CoA synthetase long-chain family member 3 (ACSL3) as a restriction factor against influenza A virus (IAV). Clinical transcriptomic analysis shows that ACSL3 is upregulated in peripheral blood mononuclear cells from mildly ill patients with influenza. Ectopic expression and loss-of-function validation demonstrate the physiological role of ACSL3 in restricting viral proliferation of diverse IAV

Reversing enhancer RNA-mediated IKBKE gene repression enables synthetic anticancer immunity in prostate cancer models.

Immunotherapy has been effective in many cancer types but has failed in multiple clinical trials in prostate cancers, with the underlying mechanisms remaining largely unclear. Here, we demonstrate that androgen receptor pathway inhibitor (ARPI) plus irradiation (IR) triggered robust anticancer immunity in prostate cancers in both patients and mice. We show that androgen-activated AR suppressed innate immune signaling by inducing inhibitor of nuclear factor kappa-B kinase subunit epsilon (IKBKE) gene repression through HDAC2 interaction with an IKBKE enhancer RNA (IKBKE eRNA, or IKBKE-e). ARPI

Unraveling the role of human endogenous retroviruses in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a systemic autoimmune disease characterized by chronic and painful joint inflammation that can lead to bone erosion and significant disability if not treated promptly. The etiology of this disease is not fully defined. Multiple genetic, epigenetic, environmental, and immunological factors have been associated with the development and perpetuation of this autoimmune and inflammatory phenomenon. Human endogenous retroviruses (HERVs), retroelements that comprise approximately 8% of the human genome, have been proposed as potential contributors to the loss of immune to

An overview of commercially available polymers for colon-targeted drug delivery systems.

The colon consists of the cecum, transverse colon, ascending colon, descending colon, and sigmoid colon. Colonic diseases such as inflammatory bowel disease (IBD), irritable bowel syndrome (IBS), colon cancer, and various other conditions may occur due to alterations in pathophysiology or factors such as environmental influences, dietary habits, and immunological variations. Conventional oral dosage forms require higher doses and pose the risk of exposing non-colonic tissues to the drug, potentially leading to adverse reactions. Successful colon targeting can be accomplished achieved by levera

Autoantibodies in COVID-19: implications for disease severity and clinical outcomes.

Few pathogens have historically been subjected to as intense scientific and clinical scrutiny as SARS-CoV-2. The genetic, immunological, and environmental factors influencing disease severity and post-infection clinical outcomes, known as correlates of immunity, remain largely undefined. Clinical outcomes of SARS-CoV-2 infection vary widely, ranging from asymptomatic cases to those with life-threatening COVID-19 symptoms. While most infected individuals return to their former health and fitness within a few weeks, some develop debilitating chronic symptoms, referred to as long-COVID. Autoimmun

Chronic Hyperplastic Candidiasis-An Adverse Event of Secukinumab in the Oral Cavity: A Case Report and Literature Review.

Secukinumab (SEC) is a recombinant, fully human monoclonal antibody that is selective for interleukin-17A (IL-17A). SEC may increase the risk of developing infections such as oral herpes and oral candidiasis. The aim of this case report and literature review was to describe chronic hyperplastic candidiasis (CHC) in a patient with psoriasis (PsO) and psoriatic arthritis (PsA) treated with SEC. CHC is a rare and atypical clinical entity. A definitive diagnosis requires biopsy of the oral mucosa for histopathological diagnosis (PHD). The differential diagnosis includes hairy tongue, hairy leukopl

Gastrointestinal and Hepatobiliary Manifestations Associated with Untreated Celiac Disease in Adults and Children: A Narrative Overview.

Celiac disease (CeD) is a chronic inflammatory disease of the small intestine, produced by ingesting dietary gluten products in susceptible people. Gluten causes an impairment of the mucosal surface and, consequently, an abnormal absorption of nutrients. Although malabsorption of essential nutrients is a major risk factor for various CeD-associated morbidities, genetic, immunological, and environmental factors also play an important role. The clinical presentation of CeD widely varies and can range from asymptomatic to full-blown symptoms due to the multi-system nature of CeD. The typical gast

Tumor cell metabolic reprogramming and hypoxic immunosuppression: driving carcinogenesis to metastatic colonization.

A significant factor in the antitumor immune response is the increased metabolic reprogramming of immunological and malignant cells. Increasing data points to the fact that cancer metabolism affects not just cancer signaling, which is essential for maintaining carcinogenesis and survival, but also the expression of immune cells and immune-related factors such as lactate, PGE2, arginine, IDO, which regulate the antitumor immune signaling mechanism. In reality, this energetic interaction between the immune system and the tumor results in metabolic competition in the tumor ecosystem, limiting the

Helicobacter pylori in Inflammatory Bowel Diseases: Active Protagonist or Innocent Bystander?

Helicobacter pylori (H. pylori) infection is a prominent entity within human infectious diseases which cause chronic gastritis, peptic ulcers, gastric malignancies, and extragastric disorders. Its persistent colonization can lead to a systemic inflammatory cascade, potentially instigating autoimmune responses and contributing to the pathogenesis of autoimmune diseases. While the specific etiopathogenesis of inflammatory bowel diseases (IBDs) is still unknown, it is widely recognized that immunological, genetic, and environmental factors are implicated. Various bacterial and viral pathogens hav
