We deleted the two C-terminal residues of the scorpion toxin BmTx3, a peptidyl inhibitor of a transient A-type K(+) current in striatum neurons in culture, to assess their contribution to receptor recognition. The sBmTX3-delYP analog was shown to have a native-like structure in one-dimensional 1H-nuclear magnetic resonance (NMR) spectroscopy. We found that sBmTX3-delYP bound to its receptor less efficiently than the wild-type molecule (by a factor of about 10(5)) in binding assays with rat brain membranes, and that this molecule did not block the A-type K(+) current (at a concentration of 35 m

A new glycoside of salicylic aldehyde, paeonicluside, was isolated from the roots of the Greek endemic species Paeonia clusii subsp. clusii and identified as alpha-L-arabinopyranosyl-(1-->6)-O-beta-D-glucopyranoside of salicylic aldehyde (1). In addition, one characteristic monoterpene and two monoterpene glycosides were identified as paeoniflorigenone, paeoniflorin and benzoyl paeoniflorin, respectively. The structure of 1 was elucidated on the basis of its spectroscopic data and chemical correlation. It is the first time that a derivative of salicylic aldehyde is isolated from the well-studi

Mammalian lung surfactant is a mixture of phospholipids and four surfactant-associated proteins (SP-A, SP-B, SP-C, and SP-D). Its major function is to reduce surface tension at the air-water interface in the terminal airways by the formation of a surface-active film highly enriched in dipalmitoyl phosphatidylcholine (DPPC), thereby preventing alveolar collapse during expiration. SP-A and SP-D are large hydrophilic proteins, which play an important role in host defense, whereas the small hydrophobic peptides SP-B and SP-C interact with DPPC to generate and maintain a surface-active film. Surfac

The crystal structure of neurotoxin Ts1, a major component of the venom of the Brazilian scorpion Tityus serrulatus, has been determined at 1.7 A resolution. It is the first X-ray structure of a highly toxic anti-mammalian beta-toxin. The folding of the polypeptide chain of Ts1 is similar to that of other scorpion toxins. A cysteine-stabilised alpha-helix/beta-sheet motif forms the core of the flattened molecule. All residues identified as functionally important by chemical modification and site-directed mutagenesis are located on one side of the molecule, which is therefore considered as the

Very recently we have proposed [Hemrika et al. (1997) Proc. Natl. Acad. Sci. USA 94, 2145-2149] that the active site of the vanadate-containing chloroperoxidase from the fungus Curvularia inaequalis, of which the tertiary structure is known, is structurally very similar to that of the membrane-bound mammalian glucose-6-phosphatases for which no structural data are available. The proposed active site of glucose-6-phosphatase, however, is incompatible with the six transmembrane-helix topology model that is currently used. Here we present a new topology model for glucose-6-phosphatase which is in

The crystal structure of toxin II from the scorpion Androctonus australis Hector has been refined at 1.3 A resolution using restrained least-squares methods. The final R-factor is 0.148 for the 13,619 reflections between 7.0 A and 1.3 A resolution with F > 2.5 sigma (F) and the bond length standard deviation from ideality is 0.017 A. Although minor changes have been introduced relative to the model previously refined at 1.8 A resolution, the use of higher-resolution data has allowed the modelling of some discrete disorder. Thus, three residues (including a disulphide bridge) have been built wi

The venom of the scorpion Androctonus mauretanicus mauretanicus contains a toxin--P05--which is structurally and functionally similar to scorpion leiurotoxin I (87% sequence identity), a blocker of the apamin-sensitive Ca(2+)-activated K+ channels. P05, a 31-residue polypeptide cross-linked by three disulfide bridges, also possesses binding and physiological properties similar to those of the bee venom toxin apamin (18 residues, two disulfides). However, the amino acid sequences of these two polypeptides are dissimilar, except for a common Arg-Arg-Cys-Gln motif which is located on an alpha-hel

Nephrogenic diabetes insipidus (DIR) is an X-linked disorder characterized by insensitivity of the distal nephron for the pituitary hormone, vasopressin. The genetic map location of the DIR gene on chromosome Xq28 coincides with the physical map location of the functional vasopressin renal V2-type receptor. Recently, the human and rat cDNAs for the vasopressin V2 receptor (AVPR2) have been identified. We show here that the structural AVPR2 gene is localized between DXS52 and G6PD, which is within the genetic map location of DIR. We also tested eight X-linked DIR probands and their families for

The present research uses integrative computational analysis to assess Streptomyces koyangensis L-asparaginase as a therapeutic against acute lymphoblastic leukemia (ALL), overcoming immunogenicity and cross-reactivity issues with E. coli and Erwinia carotovora enzymes. We characterized enzyme-oncoprotein interactions using six in silico methods: homology modeling (SWISS-MODEL, AlphaFold2), molecular docking (ClusPro, HADDOCK, AutoDock Vina), 100 ns molecular dynamics (MD) (GROMACS), and pharmacophore modeling (LigandScout). Exceptional stability of the S. koyangensis-BCL-2 complex was reveale

Poly (ADP-ribose) polymerase-1 (PARP-1) is a key enzyme in the base excision repair pathway, crucial for maintaining genomic stability by repairing DNA breaks. In cancers with mutations in DNA repair genes, such as BRCA1 and BRCA2, PARP-1 activity becomes essential for tumor cell survival, making it a promising target for therapeutic intervention. This study employs QSAR modeling, virtual screening, and molecular dynamics (MD) simulations to identify potential PARP-1 inhibitors. A dataset of inhibitors was analyzed using 12 molecular fingerprint descriptors to develop robust QSAR models, with

New antibiotics are needed to battle growing antibiotic resistance, but the development process from hit, to lead, and ultimately to a useful drug takes decades. Although progress in molecular property prediction using machine-learning methods has opened up new pathways for aiding the antibiotics development process, many existing solutions rely on large data sets and finding structural similarities to existing antibiotics. Challenges remain in modeling unconventional antibiotic classes that are drawing increasing research attention. In response, we developed an antimicrobial activity predicti

The hematopoietic cell malignancy is one of the most prevalent type of cancer and the disease has multiple pathologic molecular signatures. Research on the origin of hematopoietic cancer stem cells and the mode of subsequent maintenance and differentiation needs robust animal models that can reproduce the transformation and differentiation event in vivo. Here, we show that co-transduction of MYC and PIM2 proto-oncogenes into mouse bone marrow cells readily establishes permanent cell lines that can induce lethal myeloid sarcoma in vivo. Unlike the previous doubly transgenic mouse model in which

Since the discovery of the clustered regularly interspaced short palindromic repeat (CRISPR)-associated (Cas) system, genome-editing technologies have advanced rapidly, and the field has expanded substantially. In parallel, artificial intelligence (AI) has progressed at an unprecedented pace and is transforming genetic engineering. Early AI applications primarily addressed predictive tasks, including guide RNA (gRNA) activity, off-target effects, and editing outcomes for CRISPR-Cas nucleases, base editors, and prime editors. Recently, transformer-based protein language models (PLMs) and genera

Myotonic dystrophy type 1 (DM1) is caused by expanded CTG repeats, d(CTG)exp, transcribed into toxic r(CUG)exp RNA repeats that sequester splicing regulator MBNL1, leading to its loss-of-function. An emerging therapeutic strategy toward DM1 treatment relies on the inhibition of MBNL1 sequestration by using small molecules, oligomers, peptides, engineered proteins, or synthetic oligonucleotides that interact with CUG repeats at the RNA level and/or CTG repeats at the DNA level. This review covers ∼18 years of research in the field of CUG and CTG ligands that were identified or rationally design

The longevity research field is experiencing unprecedented momentum, driven by advances in molecular biomarkers, digital health technologies, and artificial intelligence. However, established physiological measures-VO2max, heart rate variability, body composition, muscle strength, among others-with decades of validation are increasingly presented as novel "longevity biomarkers" without acknowledging their foundational research. This perspective proposes integration as the constructive path forward, creating multidimensional aging models that combine functional measures, molecular biomarkers, m

Over a decade ago, in an Editorial, we proposed that beyond the foundational discovery of the DNA double helix, modern molecular biology has undergone a second revolution: a shift from static, single-structure models to dynamic, ensemble-based energy landscapes that define biomolecular function. Recently, Quarterly Reviews of Biophysics revisited this question, arguing that this shift is not merely conceptual but represents a revolution that explains functional regulation and enables the design of next-generation allosteric therapies. These ideas-driven by physics and chemistry-have transforme

Mutations in the chromatin remodeler CHD8 are associated with autism and macrocephaly. While mouse models of Chd8 haploinsufficiency recapitulate brain overgrowth, the specific cellular mechanisms and developmental timing that lead to these anatomical abnormalities remain poorly understood. Here, we conducted 3D imaging of Chd8 V986*/+ mouse brains using magnetic resonance imaging followed by tissue clearing and cellular resolution light-sheet microscopy across embryonic and postnatal developmental stages. We found that brain overgrowth occurs postnatally, driven by an expansion of oligodendro
