A highly conserved two-gene operon is crucial for lipoarabinomannan localization, pathogenesis, and cell envelope function in Mycobacterium abscessus.

Mycobacterium abscessus is an emerging threat, causing infections that are difficult to treat due to intrinsic resistance to most antibiotics. Determinants of M. abscessus physiology and pathogenesis remain poorly understood, hampering therapeutic development. Here, we show that in M. abscessus, the lprg-mfs operon is essential for virulence in macrophages and in mice. Loss of lprg-mfs in M. abscessus causes accumulation of the glycolipid lipoarabinomannan (LAM) on the cell surface and in culture supernatant suggesting that this system participates in LAM import. This contrasts with its propos…

First insight of characteristics and prediction of Mycobacterium tuberculosis drug resistance by whole genome sequencing in Fujian Province, China.

To delineate the molecular characteristics and assess the drug resistance prediction value of whole-genome sequencing (WGS) for Mycobacterium tuberculosis (MTB) in Fujian Province, China. Representative MTB surveillance isolates underwent phenotypic drug susceptibility testing (pDST), followed by WGS(gDST), with resistance mutation analysis conducted on the SAM-TB platform. The rate of mono-resistance, poly-resistance, and multidrug-resistance (MDR) identified by pDST were 38.67%, 12.00% and 10.00% respectively. Lineage analysis revealed that L2 (60.0%) and L4 (38.7%) predominated. No statisti…

Detection of Anti-Mycobacterium leprae Antibodies in Leprosy Contacts Using a Label-Free Approach with the Heat-Transfer Method.

Early detection of Mycobacterium leprae infection is essential to interrupt leprosy transmission and guide prophylactic interventions. Current diagnostic strategies rely largely on clinical signs, and laboratory tools lack sensitivity to detect asymptomatic individuals, who can play a critical role in disease transmission and evolve to disease as well. In this study, we report the development of a heat-transfer method (HTM)-based biosensor employing a newly designed multiepitope protein receptor, capable of detecting anti-M. leprae antibodies in human serum, with sensitivity for leprosy contac…

Molecular Construction and Expression Analysis of Rv3875 and Rv2873 from Mycobacterium tuberculosis as Novel Protein Biomarkers for Tuberculosis Immunodiagnostics.

BACKGROUND: Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis (TB), continues to become a significant risk to world health despite the availability of Bacille Calmette-Guérin (BCG) vaccination. The limited efficacy of BCG in adults and the low sensitivity of conventional diagnostic methods highlight the urgent need for novel antigens to improve TB immunodiagnostics. METHODS: This study constructs, clones, and expresses Rv3875 and Rv2873, encoding ESAT-6 and MPT83 proteins from Mtb H37Rv as potential subunit proteins. Genomic DNA was extracted, and target genes were amplifi…

Physiologic recovery of Mycobacterium tuberculosis from drug injury: A molecular study of post antibiotic effect in vitro and in vivo.

Post-antibiotic effect (PAE) describes the delay in bacterial growth that continues after antibiotics are cleared. The physiologic basis of PAE in Mycobacterium tuberculosis (Mtb) remains poorly understood. Here, we evaluated the long-standing hypothesis that PAE reflects the time required for bacteria to recover from drug-induced physiologic damage by comparing Mtb after varying durations of treatment with the four-drug isoniazid, rifampin, pyrazinamide, ethambutol combination in vitro and in BALB/c mice using two novel molecular readouts of bacterial health. In aerobic axenic culture and in …

High prevalence of depressive symptoms and its impact on quality of life in patients with Mycobacterium avium complex lung disease.

BACKGROUND: Depression is a known complication of various chronic diseases; however, its prevalence in patients with Mycobacterium avium complex lung disease (MAC-LD) remains unclear. Although patients with MAC-LD have an impaired health-related quality of life (HrQoL), previous studies on HrQoL have lacked an assessment of depressive symptoms, which could be associated with HrQoL. OBJECTIVE: We investigated the prevalence of depressive symptoms in patients with MAC-LD to assess whether depressive symptoms are related to HrQoL and examined factors associated with the presence of depressive sy…

A Simultaneous Assessment of Efficacy and Safety of Amikacin Liposome Inhalation Suspension for Refractory Mycobacterium avium Complex Lung Disease.

PURPOSE: Patients with refractory Mycobacterium avium complex lung disease (MACLD) represent a difficult-to-treat population with limited treatment options. In the CONVERT trial in refractory MACLD, amikacin liposome inhalation suspension (ALIS) plus multidrug therapy (MDT) resulted in higher culture conversion (CC) than MDT alone in the first 6 months of treatment (29% vs 9%). Adverse events (AEs) were reported in 98% and 91% of patients in the ALIS plus MDT and MDT alone arms, respectively, mostly of mild to moderate severity. A simultaneous assessment of efficacy and safety could be informa…

Mycobacterium marinum Infection Misdiagnosed as Skin Lesions of Sjögren's Syndrome: A Case Report of Fishbone Injury and Literature Review.

Cutaneous infections caused by Mycobacterium marinum are rare. We report a case initially misdiagnosed as skin lesions associated with Sjögren's syndrome but clinically highly suspected of Mycobacterium marinum infection, and we review the relevant literature. The patient was a 39-year-old male with a prior history of Sjögren's syndrome. Following an accidental fishbone injury at work, he developed skin redness, swelling, and ulceration on the right middle and little fingers, which was subsequently followed by multiple nodules on the dorsum of the right hand and upper limb. After debridement, …

Comparative in vitro antimicrobial susceptibility profiles of clofazimine and pyrifazimine against clinical isolates of Mycobacterium tuberculosis in southwest China.

UNLABELLED: Clofazimine (CFZ) is a key drug used to treat drug-resistant tuberculosis (DR-TB), while pyrifazimine (TBI-166) is an improved riminophenazine derivative with better pharmacokinetics. However, there is a lack of data on its susceptibility and resistance in regions with a high disease burden, such as southwestern China. We compared the in vitro antimicrobial activities of CFZ and TBI-166 against 249 DR-TB clinical isolates (99 multidrug-resistant TB [MDR-TB] and 150 pre-extensively drug-resistant TB [pre-XDR-TB] isolates) from southwestern China. TBI-166 exhibited a concentration-de

Annual trends in Mycobacterium tuberculosis detection and rifampicin resistance at a regional referral hospital in Northern Ghana, 2019-2024.

Background Tuberculosis (TB) remains an important public health problem in Ghana, while rifampicin-resistant TB (RR-TB) poses an additional challenge to TB control. Routine molecular diagnostic data can provide useful information on temporal patterns of TB detection and drug resistance, particularly in regions where population-based surveillance data are limited. However, findings from referral facilities may be influenced by referral patterns and access to diagnostic services and should therefore be interpreted within their health-system context. We conducted a cross-sectional study using his

Expression of Toll-like Receptors and Chemokine Receptors in the Early and Advanced Stages of HIV Infection Aggravated by Mycobacterium tuberculosis.

This study examined Toll-like receptors (TLRs)/chemokine receptors (CCRs) gene expression levels in HIV-infected individuals at different stages of HIV infection. It has been demonstrated that in the advanced stages of HIV infection, aggravated by Mycobacterium tuberculosis (M. tuberculosis) infection, there was a significant increase in TLR2, TLR4, TLR7, and TLR9 expression levels (181-, 8.4-, 15-, and 3.4-fold, respectively), compared to the early stages of the disease, which also correlated with an increase in viral load. The advanced stages of HIV infection were also characterized by an in…

A genome-wide association study identified PRKCB as a causal gene and therapeutic target for Mycobacterium avium complex disease.

Non-tuberculous mycobacterial pulmonary disease (NTM-PD) is a chronic progressive lung disease that is increasing in incidence. Host genetic factors are associated with NTM-PD susceptibility. However, the heritability of NTM-PD is not well understood. Here, we perform a two-stage genome-wide association study (GWAS) and discover a susceptibility locus at 16p21 associated with NTM-PD, especially with pulmonary Mycobacterium avium complex (MAC) disease. As the lead variant, rs194800 C allele augments protein kinase C beta (PRKCB) gene expression and associates with severer NTM-PD. The functional…

Determining the Antimycobacterial Action of Rottlerin Against Mycobacterium Species and Toxicity, Antioxidant Properties, and Therapeutic Target Affinity of Rottlerin.

Infections by Mycobacterium spp. are responsible for thousands of deaths every year worldwide. Microbial resistance, toxic effects, and adverse consequences of conventional therapies bring forth the need to search for new therapeutic agents. The aim of this study was to determine the antimicrobial action of the molecule Rottlerin against Mycobacterium spp. The broth microdilution assay showed that Rottlerin inhibited the mycobacterial growth at concentrations ≤ 50 µg/mL (≤ 96.81 µM), and the lowest bactericidal concentration was observed against M. tuberculosis (25 µg/mL-48.40 µM). The cytotox…
