Loss of the Asn333 high-mannose N-glycan impairs ACP3 folding and drives disease-associated dysfunction.

Missense mutations disrupting N-glycosylation motifs can perturb protein folding and trafficking, yet the site-specific contributions of individual N-glycans to proteostasis remain poorly defined. Here, we delineated the functional hierarchy of three N-glycosylation sites in human prostatic acid phosphatase (ACP3). Glycoproteomic analysis showed that Asn94 and Asn220 carry predominantly complex-type N-glycans, whereas Asn333 is enriched in high-mannose structures. Glycan-deficient N94Q and N220Q variants were efficiently secreted and retained phosphatase activity, whereas N333Q and T335M accum

Molecular mechanism of GTPase-activating protein-assisted hydrolysis of GTP catalyzed by Cdc42.

Cell division control protein 42 homolog (Cdc42), a key regulator of cell proliferation and migration implicated in cancer progression, cycles between active GTP-bound and inactive GDP-bound states. GTPase-activating proteins (GAPs) promote Cdc42 inactivation by accelerating GTP hydrolysis, but the underlying catalytic mechanism remains incompletely understood. Here, integrated molecular dynamics (MD) simulations and density functional theory (DFT) calculations demonstrate that GAP binding promotes hydrolysis through a combination of active-site preorganization, enhanced electrostatic interact

Acquisition of amantadine resistance via M gene reassortment in canine H3N2 influenza virus and elucidation of the resistance mechanism.

A novel variant of the canine influenza virus H3N2 (cH3N2), designated as the M variant, was identified as containing a matrix (M) gene segment derived from the 2009 pandemic H1N1 virus (pdmH1N1), raising concerns regarding potential changes in antiviral drug sensitivity. In vitro and in vivo antiviral susceptibility assays demonstrated that while the parental cH3N2 strain was sensitive to amantadine, the M variant strain had acquired resistance to this drug. In addition, both strains remained susceptible to neuraminidase (NA) inhibitors such as oseltamivir and zanamivir. Comparative amino aci

PreMemMoRF: A pretraining-fine-tuning framework for predicting membrane molecular recognition features.

Membrane molecular recognition features (MemMoRFs) are lipid-binding intrinsically disordered regions (IDRs) that undergo disorder-to-order transitions to mediate critical membrane dynamics. Consequently, their dysregulation is closely linked to severe human pathologies, including neurodegenerative diseases and viral infections. Despite their biological significance, annotations for MemMoRFs are scarce, limiting the accuracy of computational predictors. We introduce PreMemMoRF, a deep learning framework that leverages transfer learning to alleviate data scarcity. The model is pre-trained on li

Potassium channels in cancer: from hallmarks to therapeutic targeting.

Potassium channels constitute important signaling nodes in the progression of human cancers. Through canonical ion conduction-dependent functions, including intracellular K⁺ homeostasis, membrane potential and associated Ca²⁺ signaling, and cell volume, they influence proliferation, cell death, phenotypic plasticity, invasion and metastasis, metabolic reprogramming, and angiogenesis. Beyond ion conduction, potassium channels may affect malignant phenotypes through non-canonical mechanisms, including conformational signaling, protein interactions, transcriptional regulation, and auxiliary-subun

The inhibitory effect of mannoproteins on bitter aftertaste: From the salivary layer to the tongue surface to the receptor surface.

To investigate the mechanisms by which mannoproteins (MPs, CECA, and 2082) with different chain conformations and concentrations inhibit the bitter aftertaste of monomeric and oligomeric proanthocyanidins (MOPC), a multilayer penetration approach was employed to reveal their mechanisms of action across different levels. At the salivary level, MPs formed a dense network with artificial saliva (AS) through key hydrophobic interactions. Fluorescence spectroscopy results indicated that the primary interaction force between CECA and AS were electrostatic interaction. However, as the concentration o

Antimicrobial Efficient Biological Adhesive for Infected Wound Repair: Robust Tissue Adhesion, Broad-Spectrum Antimicrobial Activity, and Scar Minimization.

Chronic infected wounds face dual challenges of inflammatory dysregulation and biofilm persistence, resulting in delayed healing and pathological scarring. Current bioadhesives lack microenvironmental adaptability and multifunctionality, often aggravating tissue damage. Here, a protein-based bioadhesive is engineered by integrating a Lanmodulin (LanM) module with an arginine-rich elastin-like (Rprotein) module and incorporating lanthanide ions (Ln3⁺), which induced a conformational transition in LanM, enhancing its hydrogen bonding and molecular packing. The fabricated materials achieve high a

Mutations in Hsp40 co-chaperone change the unique canonical inter-domain interactions stimulating LGMDD1 myopathy.

UNLABELLED: Limb-girdle muscular dystrophy D1 (LGMDD1) is a rare, dominantly inherited neuromuscular disorder caused by mutations in the HSP40 co-chaperone DNAJB6, primarily in the GF or J-domains. Currently, no treatments are available, and a challenge in understanding the disease is identifying a specific client protein for DNAJB6 in skeletal muscle. Our previous research indicated that LGMDD1 GF domain mutants in Sis1 exhibit substrate-specific effects, influenced by HSP70 activity. Herein, we found that novel mutations in the J-domain similarly affected chaperone function. The J-domain mut

Novel Compound Heterozygous Missense Variants in RPL3L Gene Associated With Neonatal Dilated Cardiomyopathy.

Dilated cardiomyopathy type 2D (CMD2D) is a rare autosomal recessive disorder characterized by neonatal-onset severe cardiomyopathy, rapid progression to cardiac decompensation, and high mortality, with heart transplantation being the only life-saving intervention. Although mutations in RPL3L, a muscle-specific ribosomal protein gene critical for cardiac and skeletal muscle function, are known to cause CMD2D, this disorder remains genetically and clinically undercharacterized. To date, only 13 patients with RPL3L-related CMD2D have been reported, with nearly all of whom are attributed to compo

Pulling Forces Dampen Torsional Fluctuations of Actin Filaments and Reduce Cooperative Cofilin Binding.

A variety of potential biological roles of mechanical forces have been proposed in the field of cell biology. In particular, mechanical forces alter the mechanical conditions within cells and their environment, exerting a strong effect on the reorganization of the actin cytoskeleton. Single-molecule imaging studies have provided evidence that an actin filament may act as a mechanosensor. An increase in the tension within actin filaments causes changes in their conformation and affinity to their regulatory proteins. However, our current understanding of the molecular mechanisms of the tension s

Discovery and mechanism of novel anticoagulant peptides from Larimichthys crocea based on in silico, molecular docking, molecular dynamics, and in vitro analysis.

Fish is rich in protein and is an ideal raw material for the preparation of bioactive peptides. This study effectively identified 21 bioactive peptides from Larimichthys crocea protein using a combination of bioinformatics and in vitro evaluation. Molecular docking and molecular dynamics simulations (MD) showed that six of the peptides could strongly bind to the active center of thrombin through hydrogen bonding and hydrophobic interactions (binding energies are -7.2 to -8.5 kcal/mol), thereby displaying significant anticoagulant potential. These peptides were further demonstrated to have sign

The Role of α-Synuclein in Etiology of Neurodegenerative Diseases.

A presynaptic protein called α-synuclein plays a crucial role in synaptic function and neurotransmitter release. However, its misfolding and aggregation have been implicated in a variety of neurodegenerative diseases, particularly Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. Emerging evidence suggests that α-synuclein interacts with various cellular pathways, including mitochondrial dysfunction, oxidative stress, and neuroinflammation, which contributes to neuronal cell death. Moreover, α-synuclein has been involved in the propagation of neurodegenerative proces

Supervised screening of Tecovirimat-like compounds as potential inhibitors for the monkeypox virus E8L protein.

Monkeypox virus (MPXV) is a budding public health threat worldwide, and there lacks a personalized drug availability to treat MPXV infections. Tecovirimat, an antiviral drug against pox viruses, is recently confirmed to be effective against the MPXV in vitro using nanomolar concentrations. Therefore, the current study considers Tecovirimat as a reference compound for a machine learning-based guided screening to scan bioactive compounds from the DrugBank with similar chemical features or moieties as the Tecovirimat to inhibit the MPXV E8L surface binding protein. We used AlphaFold2 to model the

Volatile Anesthetic Sevoflurane Precursor 1,1,1,3,3,3-Hexafluoro-2-Propanol (HFIP) Exerts an Anti-Prion Activity in Prion-Infected Culture Cells.

Prion disease is a neurodegenerative disorder with progressive neurologic symptoms and accelerated cognitive decline. The causative protein of prion disease is the prion protein (PrP), and structural transition of PrP from the normal helix rich form (PrPC) to the abnormal β-sheet rich form (PrPSc) occurs in prion disease. While so far numerous therapeutic agents for prion diseases have been developed, none of them are still useful. A fluorinated alcohol, hexafluoro isopropanol (HFIP), is a precursor to the inhalational anesthetic sevoflurane and its metabolites. HFIP is also known as a robust

Tauopathy: A common mechanism for neurodegeneration and brain aging.

Tau, a microtubule-associated protein promotes assembly and stability of microtubules which is related to axoplasmic flow and critical neuronal activities upon physiological conditions. Under neurodegenerative condition such as in Alzheimer's Disease (AD), tau-microtubule binding dynamics and equilibrium are severely affected due to its aberrant post-translational modifications including acetylation and hyperphosphorylation. This event results in its conformational changes to form neurofibrillary tangles (NFT) after aggregation in the cytosol. The formation of NFT is more strongly correlated w

Crystal structure of the C-terminal domain of the primosomal DnaT protein: Insights into a new oligomerization mechanism.

DnaT is a replication restart primosomal protein required for re-initiating chromosomal DNA replication in bacteria. DnaT can be a monomer, dimer, trimer, tetramer, or pentamer. The oligomerization and disassembly of DnaT oligomers are critical in primosome assembly. Prior to this work, only the ssDNA-bound structure of the pentameric DnaT truncated protein (aa 84-153; DnaT84-153) was available. The mechanism by which DnaT oligomerizes as different states is unclear. In this paper, we report the crystal structure of the C-terminal domain of DnaT (aa 84-179; DnaTc) at 2.30 Å resolution (PDB ent

Crystal structure reveals vaccine elicited bactericidal human antibody targeting a conserved epitope on meningococcal fHbp.

Data obtained recently in the United Kingdom following a nationwide infant immunization program against serogroup B Neisseria meningitidis (MenB) reported >80% 4CMenB vaccine-mediated protection. Factor H-binding protein (fHbp) is a meningococcal virulence factor and a component of two new MenB vaccines. Here, we investigated the structural bases underlying the fHbp-dependent protective antibody response in humans, which might inform future antigen design efforts. We present the co-crystal structure of a human antibody Fab targeting fHbp. The vaccine-elicited Fab 1A12 is cross-reactive and tar

X-ray structure of human aquaporin 2 and its implications for nephrogenic diabetes insipidus and trafficking.

Human aquaporin 2 (AQP2) is a water channel found in the kidney collecting duct, where it plays a key role in concentrating urine. Water reabsorption is regulated by AQP2 trafficking between intracellular storage vesicles and the apical membrane. This process is tightly controlled by the pituitary hormone arginine vasopressin and defective trafficking results in nephrogenic diabetes insipidus (NDI). Here we present the X-ray structure of human AQP2 at 2.75 Å resolution. The C terminus of AQP2 displays multiple conformations with the C-terminal α-helix of one protomer interacting with the cytop
