Polycystin, the polycystic kidney disease 1 protein, is expressed by epithelial cells in fetal, adult, and polycystic kidney.

Polycystic kidney disease 1 (PKD1) is the major locus of the common genetic disorder autosomal dominant polycystic kidney disease. We have studied PKD1 mRNA, with an RNase protection assay, and found widespread expression in adult tissue, with high levels in brain and moderate signal in kidney. Expression of the PKD1 protein, polycystin, was assessed in kidney using monoclonal antibodies to a recombinant protein containing the C terminus of the molecule. In fetal and adult kidney, staining is restricted to epithelial cells. Expression in the developing nephron is most prominent in mature tubul

Isolation and analysis of the breakpoint sequences of chromosome inversion In(3L)Payne in Drosophila melanogaster.

Chromosomal rearrangements constitute a significant feature of genome evolution, and inversion polymorphisms in Drosophila have been studied intensely for decades. Population geneticists have long recognized that the sequence features associated with inversion breakpoints would reveal much about the mutational origin, uniqueness, and genealogical history of individual inversion polymorphisms, but the cloning of breakpoint sequences is not trivial. With the aid of a method for rapid recovery of DNA clones spanning rearrangement breakpoints, we recover and examine the DNA sequences spanning the

Conserved, extended MHC haplotypes.

Extended major histocompatibility complex (MHC) haplotypes are fixed conserved regions of the short arm of the sixth human chromosome defined by their HLA-B, complotype (BF, C2, C4A, C4B), HLA-DR alleles. The regions of conservation may extend further. At least a third of normal MHC haplotypes in Caucasians are extended, and they are largely responsible for previously observed linkage disequilibrium in the region. They are relatively population-specific. Their presence has major implications for tissue transplantation, and particularly for the identification of unrelated donor-recipient pairs,

Location of breakpoints within the major breakpoint cluster region (bcr) in 33 patients with bcr rearrangement-positive chronic myeloid leukemia (CML) with complex or absent Philadelphia chromosomes.

We report the sublocalization of the breakpoint in chromosome 22 in 33 patients with chronic myeloid leukemia (CML) who also had unusual marrow cytogenetics. In 23 patients, the leukemic clones were characterized by Philadelphia (Ph1) chromosomes that arose through complex translocations that involved three or more chromosomes. In the remaining ten patients, there were no detectable Ph1 chromosomes despite molecular evidence for the presence of rearrangements in the major breakpoint cluster region (bcr) of chromosome 22 in all cases. There was no significant difference between the two groups w

Viral determinants of virulence for Rift Valley fever (RVF) in rats.

Rift Valley fever viral strains or variants (RVFV) were compared with respect to (a) virulence for Wistar-Furth rats; (b) in vitro sensitivity to rat and human interferon; (c) ability to form plaques in primary hepatocyte cultures from genetically resistant or susceptible rat strains, and (d) replicative potential in continuous rat cell lines. Egyptian strains were highly virulent for Wistar-Furth rats; relatively resistant to rat interferon-alpha/beta; capable of producing plaques in primary hepatocyte monolayers; and, in general, replicated more rapidly than the low-virulent, sub-Saharan str

Molecular studies on the nature of the repair defect in ataxia-telangiectasia and their implications for cellular radiobiology.

We have utilized DNA transfer and recombinant DNA techniques to probe DNA double-strand break repair in the human ionizing radiation-sensitive genetic syndrome ataxia-telangiectasia (A-T). Using restriction enzyme-generated double-strand breaks in the coding sequence of a selectable gene we have detected a significantly greater frequency of mis-repair of such breaks in a permanent A-T cell line compared with cell lines of normal radiosensitivity. This mis-repair in A-T can plausibly explain many of the clinical features of the disease but was insufficiently detailed to address the broad proble

The chromosome-level genome of Mesobuthus martensii provides insights into its evolution and the diversity of venom peptides.

Mesobuthus martensii, the source species of the traditional Chinese medicine "Quanxie", has long been utilized for its therapeutic properties. Venom peptides are recognized as the major active molecular basis of these pharmacological activities. Despite this, their development as therapeutic agents remains poorly explored. In this study, we performed an integrated multi-omics investigation to systematically explore the venom peptides of Mesobuthus martensii. We generated a chromosome-level genome assembly of Mesobuthus martensii using third-generation sequencing technologies, yielding a genome

Quorum-Sensing-Mediated Extracellular Electron Transfer Enables Hydrogel Morphogenesis.

Engineered living materials seek to capture the sensitivity, responsiveness, and programmable characteristics of biological systems. One emergent property of living systems is genetically driven spatial patterning, which controls cell differentiation and the development of complex multicellular organisms. Inspired by this capability, we use bacteria to spatially control material assembly. In our system, extracellular electron transfer (EET) flux from Shewanella oneidensis drives hydrogel synthesis via copper-catalyzed radical polymerization. We first construct a recombinant quorum-sensing syst

Variant-divergent death: Omicron intensifies bystander T-cell apoptosis via GDF15-BCL2L13.

BACKGROUND: Severe Omicron cases present profound lymphocytopenia, suggesting variant-specific immune injury. RESULTS: We identify CD63 as a conserved T-cell host factor supporting ACE2-independent SARS-CoV-2 entry. Despite lower intracellular viral loads than the ancestral strain, Omicron elicits enhanced T-cell apoptosis largely through a bystander mechanism. Omicron-stimulated epithelial cells secrete GDF15, which upregulates the pro-apoptotic protein BCL2L13 in T cells and thereby remotely accelerates apoptosis in uninfected bystanders. Functionally, recombinant GDF15 increases BCL2L13 an

The potential antitumor effects of combining intravesical therapy with recombinant bacillus calmette-guérin and an anti-PD-1 inhibitor in bladder cancer.

BACKGROUND: Recombinant Bacillus Calmette-Guérin (rBCG) is a genetically modified BCG that enhances therapeutic efficacy and safety in patients with bladder cancer. Pembrolizumab is an immune checkpoint inhibitor (ICI) that is used to treat bladder cancer. We aimed to evaluate the effectiveness of the combination of rBCG and pembrolizumab in vitro and in vivo bladder cancer models. METHODS: We investigated the antitumor effects of rBCG-dltA and pembrolizumab in a bladder cancer-on-a-chip (BCOC) model, an organoid three-dimensional cell culture tool. An orthotopic mouse model was employed to as

Rapid Detection of N9 Subtype Avian Influenza Viruses Using an NA-Targeting Lateral Flow Immunoassay.

The emergence of highly pathogenic avian influenza viruses (AIVs) poses significant threats to global public health and the poultry economy. Rapid detection methods targeting the hemagglutinin (HA) protein have become increasingly unreliable due to ongoing antigenic drift and genetic mutations. In this study, we screened two monoclonal antibodies (mAbs) against the highly conserved neuraminidase (NA) protein, which were characterized by high affinity and specificity for the N9 subtype. Using the paired mAbs 12A2 and 12F12, an NA-targeting lateral flow immunoassay (NA-LFIA) was developed for th

Rapid and potent induction of protective mucosal immunity against SARS-CoV-2 by an adenovirus-vectored nanoparticle vaccine.

Effective vaccine design ideally elicits potent, durable, and broad-spectrum immunity against rapidly evolving pathogens. However, current platforms, such as recombinant adenovirus vectors and protein nanoparticles, are developed and administered separately, and each has distinct immunological and practical limitations. Here, we introduce a synergistic vaccine framework that intrinsically combines genetic delivery with rational antigen design. We engineer an adenovirus type 5 vector to co-express a prefusion-stabilized SARS-CoV-2 spike and ferritin subunit (Ad5-SpFN), which drives in vivo asse

Targeting Corin and Furin in Hypertension and Heart Failure: A New Therapeutic Frontier in Cardiovascular Therapeutics.

Corin and furin are protetic enzymes central to the activation of natriuretic peptides (NPs), which regulate cardiovascular homeostasis. Recent insights suggest that disruptions in the Corin-Furin axis-via genetic polymorphisms, aberrant post-translational modifications, or disease-associated downregulation-contribute to the pathogenesis of hypertension, heart failure, and myocardial fibrosis. This study examines current challenges in enzymatic stability, pharmacodynamics, and delivery of corin- and furin-based therapies, emphasizing translational barriers and the need for precision medicine.

Nanobody-mediated targeting of Plasmodium falciparum PfPIMMS43 can block malaria transmission in mosquitoes.

The transition from ookinete to oocyst is a critical step in the Plasmodium falciparum lifecycle and an important target for malaria transmission-blocking strategies. PfPIMMS43, a surface protein of P. falciparum ookinetes and sporozoites, is critical for this transition and aids the parasite in evading mosquito immune responses. Previous studies demonstrated that polyclonal PfPIMMS43 antibodies reduced P. falciparum infection in Anopheles mosquitoes. Here, building on these findings, we have developed high-affinity single-domain VHH antibodies (nanobodies) derived from llama heavy-chain-only

Structural and immunogenic characteristics of goose parvovirus virus-like particles.

Goose parvovirus (GPV) is a highly lethal infectious pathogen primarily affecting goslings under one month old, with infection resulting in nearly 100 % mortality. However, no recombinant vaccine has been proven effective in preventing GPV infection. This study aims to structurally characterize GPV virus-like particles (VLPs) produced from Saccharomyces cerevisiae using cryo-electron microscopy (Cryo-EM) and three-dimensional reconstruction, to explore potential binding receptors, and to preliminarily verify the immunogenicity of GPV VLPs. The results show that GPV VLPs produced from Saccharom

A targeted vector for brain endothelial cell gene delivery and cerebrovascular malformation modelling.

Defects in brain endothelial cells (brainECs) can cause severe cerebrovascular malformations, including arteriovenous malformation (AVM) and cerebral cavernous malformation. The lack of appropriate tools for cerebrovascular disease modelling and local genetic manipulation of the brain vasculature hinders research on cerebrovascular malformations. Here we develop a recombinant adeno-associated virus (rAAV) tool termed miniBEND (rAAV-based mini-system for brain endothelial cells, rAAV-miniBEND), which combines a minimal promoter and an optimized cis-acting element isolated from the mouse gene Te

Differing genetics of saline and cocaine self-administration in the hybrid mouse diversity panel.

To identify genes that regulate the response to the potentially addictive drug cocaine, we performed a control experiment using genome-wide association studies (GWASs) and RNA-Seq of a panel of inbred and recombinant inbred mice undergoing intravenous self-administration of saline. A linear mixed model increased statistical power for analysis of the longitudinal behavioral data, which was acquired over 10 days. A total of 145 loci were identified for saline compared to 17 for the corresponding cocaine GWAS. Only one locus overlapped. Transcriptome-wide association studies (TWASs) using RNA-Seq

S100B Mitigates Cytoskeletal and Mitochondrial Alterations in a Glial Cell Model of Autosomal Recessive Spastic Ataxia of Charlevoix-Saguenay.

Autosomal Recessive Spastic Ataxia of Charlevoix-Saguenay (ARSACS) is an early-onset neurological disorder caused by mutations in the SACS gene, resulting in the loss of sacsin function. Sacsin is a multidomain protein that plays key roles in chaperone regulation, protein quality control, and neurofilament dynamics. Sacsin deficiency leads to disruption of intermediate filament and mitochondrial networks. S100B, a multifunctional brain-enriched protein, exhibits protective neuroprotective functions that include chaperone activity and interactions with filament proteins and mitochondria. In thi

Progress in AAV-Mediated In Vivo Gene Therapy and Its Applications in Central Nervous System Diseases.

As the blood-brain barrier (BBB) prevents molecules from accessing the central nervous system (CNS), the traditional systemic delivery of chemical drugs limits the development of neurological drugs. However, in recent years, innovative therapeutic strategies have tried to bypass the restriction of traditional drug delivery methods. In vivo gene therapy refers to emerging biopharma vectors that carry the specific genes and target and infect specific tissues; these infected cells and tissues then undergo fundamental changes at the genetic level and produce therapeutic proteins or substances, thu
