Natural tick-borne encephalitis in 2 Huacaya alpacas (Vicugna pacos).

Although rare, tick-borne encephalitis (TBE) is one of the most important and commonly fatal viral diseases affecting the central nervous system (CNS). This arboviral disease is transmitted by ticks and prevalent in widespread parts of Eurasia. Besides humans, several domestic animals such as dogs, horses, and ruminants can also be infected. To our knowledge, there have been no reports of TBE in South American camelids, so far. Here, we present 2 cases of Huacaya alpacas with progressive, therapy-resistant neurologic signs, which were euthanized and submitted for necropsy. Histologic examinati…

Molecular Detection of Coxiella burnetii in Vaginal Swab Samples from Sheep That Aborted.

Background: Coxiella burnetii, an obligate intracellular bacterium, is the etiological agent of Q fever in humans and one of the causes of abortion in small ruminants. Although coxiellosis is considered an exotic disease, there are a few reports in Mexico. Methods: The objective of this work was to determine the presence of C. burnetii DNA in vaginal samples from sheep that presented abortion and ram semen. A total of 180 vaginal exudate samples and 20 semen samples were obtained from five Central and Southern States of Mexico. Total DNA was extracted from vaginal swabs and C. burnetii was ide…

Comparison of the efficacy of macrocyclic lactone anthelmintics, either singly or in combination with other anthelmintic(s), in nine beef herds in southern NSW.

Anthelmintic resistance (AR) is a well-recognized challenge in farmed ruminants. The use of anthelmintics in combination is one of the strategies recommended to slow the rate of AR development. Two studies were undertaken in 2017 and 2019 to assess the efficacy of single-dose macrocyclic lactone (ML) anthelmintic and ML combination drenches. In total, 11 Faecal Egg Count Reduction Trials (FECRTs) were set up in 10 different beef herds, with results available from 10 of those FECRTs (9 herds). AR to a single ML anthelmintic was detected in all 9 herds, with resistance to Cooperia and Haemonchus…

Vaccination of White-Tailed Deer with Mycobacterium bovis Bacillus Calmette-Guérin (BCG): Effect of Mycobacterium avium ssp. paratuberculosis Infection.

In many parts of the world, bovine tuberculosis eradication efforts are hampered by wildlife reservoirs of Mycobacterium bovis, which serve … he M. tuberculosis complex. M. avium subspecies paratuberculosis (Map) infection is not uncommon in ruminants such as deer. Using natural exposure to Map and experimental infection with M. bovis, we demonstrate that Map infection increased BCG vaccine efficacy as measured by lesion severity scores.

Advancements in Rift Valley fever vaccines: a historical overview and prospects for next generation candidates.

Rift Valley fever (RVF) is a zoonotic viral disease transmitted by mosquitoes and causes abortion storms, fetal malformations, and newborn animal deaths in livestock ruminants. In humans, RVF can manifest as hemorrhagic fever, encephalitis, or retinitis. Outbreaks of RVF have been occurring in Africa since the early 20th century and continue to pose a threat to both humans and animals in various regions such as Africa, Madagascar, the Comoros, Saudi Arabia, and Yemen. The development of RVF vaccines is crucial in preventing mortality and morbidity and reducing the spread of the virus. While se…

Prion Disease in Dromedary Camels, Algeria.

Prions cause fatal and transmissible neurodegenerative diseases, including Creutzfeldt-Jakob disease in humans, scrapie in small ruminants, and bovine spongiform encephalopathy (BSE). After the BSE epidemic, and the associated human infections, began in 1996 in the United Kingdom, general concerns have been raised about animal prions. We detected a prion disease in dromedary camels (Camelus dromedarius) in Algeria. Symptoms suggesting prion disease occurred in 3.1% of dromedaries brought for slaughter to the Ouargla abattoir in 2015-2016. We confirmed diagnosis by detecting pathognomonic neuro…

The role of signal transducer and activator of transcription 3 in Rift Valley fever virus infection.

Rift Valley fever (RVF) is a zoonotic disease that can cause severe illness in humans and livestock, triggering spontaneous abortion in almost 100% of pregnant ruminants. In this study, we demonstrate that signal transducer and activator of transcription 3 (STAT3) is phosphorylated on its conserved tyrosine residue (Y705) following RVFV infection. This phosphorylation was dependent on a major virulence factor, the viral nonstructural protein NSs. Loss of STAT3 had little effect on viral replication, but rather resulted in cells being more susceptible to RVFV-induced cell death. Phosphorylated …

Genetic resistance: tolerance to vector-borne diseases and the prospects and challenges of genomics.

Vector-borne diseases in cattle and small ruminants (e.g. trypanosomosis, Rift Valley fever and East Coast fever) are associated with major economic losses in tropical countries, and particularly on the African continent. A variety of control strategies (e.g. management, vaccination and/or acaricide treatments) are used to minimise their negative impacts. These strategies are often associated with environmental, technical and/or economic drawbacks. However, several indigenous livestock populations have been reported to show a level of genetic tolerance or resistance to such disease challenges …

Detection of rift valley Fever virus interepidemic activity in some hotspot areas of kenya by sentinel animal surveillance, 2009-2012.

Rift Valley fever virus causes an important zoonotic disease of humans and small ruminants in Eastern Africa and is spread primarily by a mosquito vector. In this region, it occurs as epizootics that typically occur at 5-15-year intervals associated with unusual rainfall events. It has hitherto been known that the virus is maintained between outbreaks in dormant eggs of the mosquito vector and this has formed the basis of understanding of the epidemiology and control strategies of the disease. We show here that seroconversion and sporadic acute disease do occur during the interepidemic periods…

Dual functions of Rift Valley fever virus NSs protein: inhibition of host mRNA transcription and post-transcriptional downregulation of protein kinase PKR.

Rift Valley fever virus (RVFV), which belongs to the genus Phlebovirus, family Bunyaviridae, is a negative-stranded RNA virus carrying a single-stranded, tripartite RNA genome. RVFV is an important zoonotic pathogen transmitted by mosquitoes and causes large outbreaks among ruminants and humans in Africa and the Arabian Peninsula. Human patients develop an acute febrile illness, followed by a fatal hemorrhagic fever, encephalitis, or ocular diseases. A viral nonstructural protein, NSs, is a major viral virulence factor. Past studies showed that NSs suppresses the transcription of host mRNAs, i…

Phase I biotransformation of albendazole in lancet fluke (Dicrocoelium dendriticum).

Dicroceliosis, a lancet fluke infection, is a frequent parasitosis of small ruminants and the anthelmintic drug albendazole (ABZ) is effective in control of this parasitosis. The aim of our project was to study the metabolism of ABZ and ABZ sulphoxide (ABZ.SO) in lancet fluke. Both invitro (subcellular fractions of fluke homogenates) and exvivo experiments (adult flukes cultivated in medium) were performed for this purpose. ABZ was metabolised invitro by lancet fluke NADPH-dependent enzymes by two oxidative steps (sulphoxidation and sulphonation). The apparent kinetic parameters of these react…

Serine protease activity demonstrated in the larval stage of the pentastomid Linguatula serrata.

Linguatula serrata is a cosmopolitan parasite whose intermediate hosts are cattle, goats, sheep, and other ruminants. The adult form is found in the nasal airways, frontal sinuses, and tympanic cavity of canines and felines, and it produces hemorrhages and breathing difficulties. To elucidate if L. serrata produces enzymes that are capable of degrading tissues from the intermediate host, proteolytic activities in larval products were studied. Using the zymography technique, one major protease was detected in parasite in vitro-released products with an approximate molecular weight of 75 kDa. Th…

Rift Valley fever.

Rift Valley fever (RVF) is an arthropod-borne viral disease of ruminants, camels and humans. It is also a significant zoonosis which may be encountered as an uncomplicated influenza-like illness, but may also present as a haemorrhagic disease with liver involvement; there may also be ocular or neurological lesions. In animals, RVF may be inapparent in non-pregnant adults, but outbreaks are characterised by the onset of abortions and high neonatal mortality. Jaundice hepatitis and death are seen in older animals. Outbreaks of RVF are associated with persistent heavy rainfall with sustained floo…

Spotted fever group rickettsiae from ticks captured in Sudan.

Ticks were collected from ruminants in various areas of Sudan in 1998 and 2000. Primer pairs of rickettsial citrate synthase gene (gltA) and a spotted fever group (SFG) rickettsial 190-kDa surface antigen gene (rompA), respectively, were used for identification. Polymerase chain reaction (PCR)-positive products were used for DNA sequencing. The gltA gene was detected in 55% of the ticks examined (57/104). Among the 57 ticks studied, 19 were positive for the rompA gene. Thus, 18% of the ticks examined were found to be infected with SFG rickettsiae. The nucleotide sequences of the amplified romp…

Q fever hazards from sheep and goats used in research.

Two recent outbreaks of Q fever in research institutions prompted the authors to review extant data, in an effort to delineate hazards to humans from small ruminants used in research. Prevalence of Q fever antibodies was 24% among 2097 sheep and 57% among 1475 goats from various sources. In a facility using sheep for prenatal research, in which an epidemic of human Q fever later occurred, antibody prevalence among sheep was 77%, with larger than usual proportions of high (greater than or equal to 128) titers. Antibody prevalence was 5% in a sample from the general human population and 11% amon…

Liquid feeding strategies to provide equal nutrient intake to dairy calves: increased liquid volume versus diet concentration.

The aim of this study was to compare the effects of feeding Holstein calves 750 g of daily milk replacer (MR) at either 12.5% of dry matter [(DM); 6 L/d] or 16.7% DM (4.5 L/d) on intake, growth performance, ruminal fermentation, apparent nutrient digestibility, urinary excretion, postprandial glucose and insulin dynamics, health, and behavior during the preweaning and immediate postweaning periods. Thirty calves were randomly assigned to one of 2 treatments delivering 750 g of MR powder per day but differing in concentration and volume: control (CON, 12.5% of DM, 6 L/d) or increased dry matter

Breed-specific microbiomes drive differential responses to 3-nitrooxypropanol and Acacia mearnsii in dairy cows.

Methane (CH4) inhibitor 3-nitrooxypropanol (3-NOP) shows variable efficacy in Brown Swiss (BS) and Holstein (HF) cows, but the underlying mechanisms remain unclear. Rumination bolus, a proxy for rumen microbiota, combined with exhalomics (analysis of exhaled volatiles), offered a noninvasive approach to differentiate breed responses to CH4 mitigation strategies. We investigated whether differences in rumination bolus microbiota and exhaled VFA (eVFA) between BS and HF cows contribute to their responses to 3-NOP. Sixteen multiparous cows (8 BS, 8 HF) were studied in a replicated 4 × 4 Latin squ
