GLUT1 expression, lymphocyte distribution and CD3+ T-cell metabolic subsets as predictive markers of response to immunotherapy in advanced melanoma.

BACKGROUND: Glycolysis, commonly used by malignant tumors for energy production, results in acidification of the tumor microenvironment (TME) through the secretion and accumulation of lactic acid. Acidosis is a potent inhibitor of immune cell function and may therefore affect T-cell infiltration and the efficacy of immunotherapy. This study aimed to characterize the metabolic tumor microenvironment and its association with lymphocyte distribution in patients with advanced melanoma treated with immune checkpoint blockade (ICB) therapies. METHODS: Pre-treatment formalin-fixed, paraffin-embedded …

Granzyme B+ B cells detected by single-cell sequencing are associated with prognosis in patients with intrahepatic cholangiocarcinoma following liver transplantation.

B cells possess anti-tumor functions mediated by granzyme B, in addition to their role in antigen presentation and antibody production. However, the variations in granzyme B+ B cells between tumor and non-tumor tissues have been largely unexplored. Therefore, we integrated 25 samples from the Gene Expression Omnibus database and analyzed the tumor immune microenvironment. The findings uncovered significant inter- and intra-tumoral heterogeneity. Notably, single-cell data showed higher proportions of granzyme B+ B cells in tumor samples compared to control samples, and these levels were positiv

In-situ nanoplatform with synergistic neutrophil intervention and chemotherapy to prevent postoperative tumor recurrence and metastasis.

In addition to residual tumor cells, surgery-induced inflammation significantly contributes to tumor recurrence and metastasis by recruiting polymorphonuclear neutrophils (PMNs) and promoting their involvement in tumor cell proliferation, invasion and immune evasion. Efficiently eliminating residual tumor cells while concurrently intervening in PMN function represents a promising approach for enhanced postoperative cancer treatment. Here, a chitosan/polyethylene oxide electrospun fibrous scaffold co-delivering celecoxib (CEL) and doxorubicin-loaded tumor cell-derived microparticles (DOX-MPs) i

Enhanced expression and shedding of receptor activator of NF-kappaB ligand during tumor-bone interaction potentiates mammary tumor-induced osteolysis.

The bone microenvironment plays a critical role in tumor-induced osteolysis and osteolytic metastasis through tumor-bone (TB)-interaction. Receptor activator of nuclear factor-kappaB (RANK) ligand (RANKL) is one of the critical signaling molecules involved in osteolysis and bone metastasis. However, the regulation and functional significance of RANKL at the TB-interface in tumor-induced osteolysis remains unclear. In this report, we examined the role of tumor-stromal interaction in the regulation of RANKL expression and its functional significance in tumor-induced osteolysis. Using a novel mam

A Cross-Sectional Study of CD163 Expression and CD8-Positive Tumor-Infiltrating Lymphocytes in Glial Tumors at a Tertiary Care Center.

Background and objective Glial tumors are increasingly recognized as immunologically active neoplasms in which macrophage-rich and lymphocyte-poor microenvironments may influence aggressiveness. Cluster of differentiation 163 (CD163) highlights tumor-associated macrophages, whereas CD8 marks cytotoxic tumor-infiltrating lymphocytes (TILs). This study aimed to evaluate CD163 expression and CD8-positive TILs in glial tumors and examine their association with tumor grade. Methods This cross-sectional study included 50 histopathologically confirmed glial tumors. Immunohistochemistry for CD163 and

Gegen Qinlian Decoction, a classic traditional Chinese medicine formula: a potential therapeutic strategy for ulcerative colitis and colorectal cancer.

Gegen Qinlian Decoction (GQD), a classic traditional Chinese medicine formula recorded in Shang Han Lun, has been used clinically for approximately two thousand years. In recent years, its potential roles in ulcerative colitis (UC) and colorectal cancer (CRC) have attracted increasing attention because of the multi-component, multi-target, and multi-pathway regulatory characteristics of traditional Chinese medicine (TCM). UC is a chronic relapsing inflammatory disease and an important risk factor for CRC. Although emerging immunomodulators, biologics, and targeted therapies have achieved certa

Spatially resolved multimodal hallmarks of response to neoadjuvant immunotherapies in the melanoma ecosystem in 2D and 3D.

Neoadjuvant immunotherapy has transformed cancer treatment, yet the spatial molecular architecture governing response and resistance across distinct immune checkpoint blockade (ICB) regimens remains poorly defined. We assembled the largest neoadjuvant ICB (NICB) spatial multi-omics cohort to date, profiling over 112 million cells at single-cell resolution across three melanoma ICB regimens using MERFISH spatial transcriptomics, multiplexed immunofluorescence, and scRNA-sequencing. These analyses revealed the full multicellular spatial architecture of the NICB tumor microenvironment, including …

Tenascin-C orchestrates radiotherapy-induced head and neck tumor regression.

Given that head and neck squamous cell carcinoma (HNSCC) patients have poor survival outcomes, a better understanding of the therapeutic benefits of ionizing irradiation (IR), the major treatment modality besides surgery, is needed. A confounding factor is the immunosuppressive tumor microenvironment determined by tenascin-C (TNC), a highly abundant extracellular matrix molecule upregulated by IR. We investigated the roles of TNC on radio-induced tumor regression in a murine oral HNSCC model expressing or lacking TNC. While tumors in a TNC-expressing host were radiosensitive, they were radiore…

Stromal Regulation of Tumor Perineural Invasion: A Multicellular and Neuro-Ecological Perspective.

Perineural invasion (PNI) is an aggressive driver of tumor progression, therapeutic resistance, and poor prognosis across diverse malignancies. Traditionally viewed as a tumor cell-autonomous process, PNI is increasingly recognized as a dynamic phenomenon shaped by the tumor-nerve-stromal ecosystem. This review synthesizes recent advances to reframe neural invasion as a coordinated, multicellular process governed by reciprocal interactions within this niche. We delineate how stromal and immune components, including cancer-associated fibroblasts, Schwann cells, immune cells, and the extracellul
