YAP Signaling in Glia: Pivotal Roles in Neurological Development, Regeneration and Diseases.

Yes-associated protein (YAP), the key transcriptional co-factor and downstream effector of the Hippo pathway, has emerged as one of the primary regulators of neural as well as glial cells. It has been detected in various glial cell types, including Schwann cells and olfactory ensheathing cells in the peripheral nervous system, as well as radial glial cells, ependymal cells, Bergmann glia, retinal Müller cells, astrocytes, oligodendrocytes, and microglia in the central nervous system. With the development of neuroscience, understanding the functions of YAP in the physiological or pathological p…

Interstitial-fluid shear stresses induced by vertically oscillating head motion lower blood pressure in hypertensive rats and humans.

The mechanisms by which physical exercise benefits brain functions are not fully understood. Here, we show that vertically oscillating head motions mimicking mechanical accelerations experienced during fast walking, light jogging or treadmill running at a moderate velocity reduce the blood pressure of rats and human adults with hypertension. In hypertensive rats, shear stresses of less than 1 Pa resulting from interstitial-fluid flow induced by such passive head motions reduced the expression of the angiotensin II type-1 receptor in astrocytes in the rostral ventrolateral medulla, and the resu…

Tracers progress for positron emission tomography imaging of glial-related disease.

Glial cells play an essential part in the neuron system. They can not only serve as structural blocks in the human brain but also participate in many biological processes. Extensive studies have shown that astrocytes and microglia play an important role in neurodegenerative diseases, such as Alzheimer's disease, Parkinson's disease, Huntington's disease, as well as glioma, epilepsy, ischemic stroke, and infections. Positron emission tomography is a functional imaging technique providing molecular-level information before anatomic changes are visible and has been widely used in many above-menti…

Glial roles in sterile inflammation after ischemic stroke.

Stroke is a leading cause of death and disability worldwide, but there are a limited number of therapies that improve patients' functional recovery. The complicated mechanisms of post-stroke neuroinflammation, which is responsible for secondary ischemic neuronal damage, have been clarified by extensive research. Activation of microglia and astrocytes due to ischemic insults is implicated in the production of pro-inflammatory factors, formation of the glial scar, and breakdown of the blood-brain barrier. This leads to the infiltration of leukocytes, which are activated by damage-associated mole…

The brain development of infants with intrauterine growth restriction: role of glucocorticoids.

Brain injury is a serious complication of intrauterine growth restriction (IUGR), but the exact mechanism remains unclear. While glucocorticoids (GCs) play an important role in intrauterine growth and development, GCs also have a damaging effect on microvascular endothelial cells. Moreover, intrauterine adverse environments lead to fetal growth restriction and the hypothalamus-pituitary-adrenal (HPA) axis resetting. In addition, chronic stress can cause a decrease in the number and volume of astrocytes in the hippocampus and glial cells play an important role in neuronal differentiation. There…

Experimental models of neuromyelitis optica.

For a long time, the most important inflammatory demyelinating diseases of the central nervous system (CNS), for example, multiple sclerosis (MS) and neuromyelitis optica (NMO), were extremely hard to differentiate, often with severe consequences for affected patients. This changed with the discovery of NMO-immunoglobulin G (IgG), a specific autoantibody which was detected in the vast majority of NMO patients, and with the demonstration that this autoantibody targets aquaporin 4 (AQP4), a water channel found on astrocytes in the CNS. These findings paved the way for the generation of experimen…

Expression of the interleukin 17 in cortical tubers of the tuberous sclerosis complex.

The role of interleukin 17 (IL-17) to epilepsy-associated cortical tubers of tuberous sclerosis complex (TSC) is unknown. We investigated the expression patterns of the IL-17 and IL-17 receptor (IL-17R) in cortical tubers of TSC compared with normal control cortex (CTX). We found that IL-17 and IL-17R were clearly upregulated in cortical tubers at the protein levels. Immunostaining indicated that IL-17 was specifically distributed in the innate immunity cells (DNs, GCs, astrocytes, and microglia) and adaptive immunity cells (T-lymphocytes) as well as the endothelial cells of blood vessels. The…

Multinucleated giant cells in AIDS encephalopathy: an immunohistochemical study.

We report the neuropathological findings of 5 post-mortem cases of AIDS. The most common causes of the death were multiple opportunistic infections associated with cutaneous and/or visceral Kaposi sarcoma in two cases. Cerebral edema, demyelination and spongiosis of the white matter were present but the most remarkable finding was the presence of multinucleated giant cell (MGC) within and nearby microglial nodules. On immunohistochemical investigation MGC and microglial cells exhibit positive stain only for RCA I and Ferritin, while immunohistochemical markers for astrocytes, neurons, macropha…

Synaptic endfeet in the 'acoustic nerve nucleus' of the rat. An electron microscopic study.

The medial portion of the cochlear nerve of the rat contains astrocytes, oligodendrocytes and neurons. These neurons form what has been called the 'acoustic nerve nucleus'. This nucleus has been studied here at the electron microscopic level. Its neurons are large and round, showing an eccentric nucleus, fibrillary bodies and rough endoplasmic reticulum which is not arranged in stacks. The somata and dendrites receive synaptic endfeet which can be classified into three groups according to vesicle size and shape. In general, the ultrastructural characteristics of these cells are similar to thos…

Ultrastructural findings in experimental Creutzfeldt-Jakob disease in guinea pigs.

Brains from six clinically ill guinea pigs from various serial passages infected with brain homogenate from a patient with Creutzfeldt-Jakob disease were studied by light and electron microscopy. There were no appreciable morphologic differences among the animals at different passages. Light microscopy revealed clearing, swelling, and vacuolar changes in neurons and astrocytes. These changes occurred in both perikarya and cell processes. Large isolated vacuoles in the neuropil appeared to develop by progressive fusion between swollen cellular processes. By electron microscopy many neuronal pro…

Transmission of chronic spongiform encephalopathy with kuru plaques from humans to small rodents.

An unusual case of spongiform encephalopathy was transmitted directly from a human to rats and mice. After serial passages, incubation periods were shortened to about six months in rats and four months in mice. Clinical symptoms were similar in rats and mice, including ruffled fur, arched back, bradykinesia, and hind limb paralysis. Pathologically, a spongy state, proliferation of astrocytes, and neuronal changes were observed. Electron microscopic observation of the parietal cortex of rats and mice disclosed many membrane-bound vacuoles in the neuropil, predominantly in dendrites. In the pons…

[Neuromorphological and immunofluorescence study of experimental infection caused by the attenuated strain of tick-borne encephalitis virus].

In Syrian hamsters inoculated subcutaneously with an attenuated strain of tick-borne encephalitis virus the infection runs an asymptomatic course in all the animals, some of which, however, develop within 4 weeks some changes in nerve cells and hypertrophy of astrocytes. The viral antigen is detected by Coons' method in nerve cells and spleen cells. An intensive treatment of the hamsters with cyclophosphane 106 days after virus inoculation provoked no morphological lesions in the brain which could have been associated with virus activation. No virus antigen could be found in brain impression s…

The neurotoxic effects of triethyltin (TET) sulfate on myelinating cultures of mouse spinal cord.

Well myelinated cultures of mouse spinal cord, exposed to varying concentrations of triethyltin sulfate, were examined at different time intervals by light and electron microscopy. At a high concentration of TET (10(-6)M) for 1-2 hours, the myelin sheaths and cells showed marked degenerative changes. At lower concentrations of TET (10(-7)-10(-9) M) intramyelinic and other types of vacuole formation were seen; however, these lesions were always accompanied by changes in the perikarya, dendrites and axons of neurons and in the astrocytes. Also, there was an apparent increased number of neurofila…

The inflammatory nexus: Mechanisms linking sleep disorders to neural damage and therapeutic strategies.

Inflammation is a significant contributor to neural damage. Sleep disorders, particularly sleep deprivation, have been shown to induce neurological dysfunction and contribute to multiple sleep disorder comorbidities by activating microglia and astrocytes, disrupting the integrity of the blood-brain barrier and gut barrier, promoting peripheral immune cell infiltration, and triggering systemic inflammation. In recent years, intervention strategies targeting inflammatory pathways have demonstrated therapeutic potential, suggesting that regulating neuroinflammatory responses may be an important d…
