A highly sensitive genetically encoded red cAMP sensor for multiplex imaging in vivo.

Circularly permuted green fluorescent protein (cpGFP)-based high-performance cAMP sensors have enabled real-time monitoring of cAMP dynamics with high spatiotemporal resolution in living animals. However, their utility is hampered by significant spectral overlap with other green/yellow fluorescent indicators and blue/cyan light-activated optogenetic actuators, limiting their compatibility in multiplexed imaging applications. While existing red cAMP sensors offer great spectral separation, they often suffer from a limited dynamic range ( < 1.5-fold in HEK293T cells), low cellular brightness, ag

Curcumin as a ceramide metabolism-targeting agent: Molecular mechanisms and therapeutic implications in cancer.

Curcumin, a lipophilic polyphenol widely used in traditional medicine, is the active ingredient in turmeric (Curcuma longa L.). It has various therapeutic effects, such as antitumor potential, due to its interaction with intracellular and extracellular molecules associated with different malignancies. Ceramides are fundamental constituents of cellular membranes that maintain the stability and integrity of cells. Additionally, they have become key signaling agents that govern a spectrum of cellular functions, such as differentiation, proliferation, apoptosis, and cellular senescence. This revie

The Potential Role of Vitamin D in BRCA1 Pathogenic Variant Carriers: A Narrative Review.

Vitamin D is a fat-soluble secosteroid essential for skeletal development and calcium homeostasis, but it also exerts pleiotropic effects on numerous biological processes via its active metabolites. Vitamin D metabolites act as steroid hormones that regulate cell-cycle progression, proliferation, differentiation, apoptosis, immune responses, and multiple intracellular signaling pathways. Moreover, they modulate the expression of genes involved in carcinogenesis. As circulating vitamin D levels are influenced by diet, fortified foods, and supplementation, they represent a potentially modifiable

Temporal immunomodulation of CD4+ T cells by magnesium regulates osteoimmune responses in osteoporotic fracture healing.

Osteoporotic fracture healing is impaired by dysregulated immune responses characterized by a TH1/M1-biased inflammatory microenvironment. In this study, we show that extracellular magnesium ions (Mg2+) reshape this osteo-immune niche in a dose- and time-dependent manner. Briefly, Mg2+ suppresses TRPM7-mediated Ca2+ spikes and the NFATc1-driven proinflammatory axis, thereby promoting TH2/M2 responses. However, sustained excess Mg2+ attenuates TH2/M2 responses by inhibiting Orai1/CaV-dependent Ca2+ influx and reactivating TH1/M1 responses through JAK-STAT1 signaling under low-calcium stimulatio

Intercellular Mitochondrial Transfer in the Nervous System: Roles of Tunneling Nanotubes, Mitochondrial Extracellular Vesicles, and Neuroimmune Bioenergetic Interactions.

Mitochondria are essential organelles that maintain neuronal bioenergetics, redox homeostasis, calcium signaling, and immune regulation. Traditionally, mitochondrial dysfunction has been primarily considered as an intracellular event associated with neuronal injury and neurodegeneration. However, accumulating evidence indicates that mitochondria and mitochondrial components can be transferred between cells, forming an intercellular communication network that dynamically regulates tissue homeostasis and disease progression. Intercellular mitochondrial transfer occurs through contact-dependent p…

4,4'-Dimethoxychalcone ameliorates estrogen-deficient osteoporosis by targeting PTP1B to inhibit the c-Src/NFATc1 signaling axis.

Postmenopausal osteoporosis (PMOP) is a skeletal disease linked to immune dysregulation, characterized by the overactivation of myeloid-lineage osteoclasts. To address the limitations of current therapies, this study investigated the immunomodulatory potential and molecular mechanism of the natural flavonoid 4,4'-dimethoxychalcone (DMC). Using in vitro and in vivo models, we found that DMC exhibited a desirable dual-action profile, potently inhibiting osteoclast differentiation while also promoting osteogenesis. Mechanistically, we identified Protein Tyrosine Phosphatase 1B (PTP1B) as a novel,

Glycation-Driven Mitochondrial and ER Stress Underlies Iodoacetic Acid-Induced Apoptosis in Porcine Uterus and Oviduct Epithelial Cells.

Iodoacetic acid (IAA), a highly cytotoxic disinfection byproduct commonly detected in drinking water, poses a potential risk to female reproductive health. The direct molecular mechanisms underlying its effects on the reproductive system epithelium remain unclear. This study demonstrates that IAA induces glycational stress in primary porcine uterine (UECs) and oviduct epithelial cells (OECs), representing an early event contributing to extensive cellular toxicity. IAA exposure inhibited Glyceraldehyde-3-Phosphate Dehydrogenase (GAPDH) enzymatic activity and promoted the accumulation of advance

Non-invasive electrical stimulation restores corneal nerve density and sensory function in diabetic neuropathy via KCNN-dependent mechanism.

Diabetic neuropathy (DN) is the most common complication of diabetes mellitus (DM) and often involves the cornea, where progressive loss of nerve fibers contributes to impaired corneal sensitivity and wound healing defects. Current treatments are limited, underscoring the need for a regenerative therapy. Here we show that a non-invasive neural modulation therapy, transcutaneous electrical stimulation (ES), significantly restore the nerve density and sensory function in both streptozotocin-induced DM mice and in vitro isolated trigeminal ganglia (TG) neurons. New pharmacologic and interventional horizons in hypertension and nondialysis chronic kidney disease: bridging innovation and clinical translation.

Hypertension is highly prevalent among patients with chronic kidney disease (CKD), where it functions both as a cause and a consequence of disease progression. Renin-angiotensin-aldosterone system inhibitors remain foundational for nephroprotection, with calcium channel blockers, thiazide diuretics and mineralocorticoid receptor antagonists (MRAs) commonly added to improve blood pressure (BP) control. However, the limited efficacy of thiazides at low estimated glomerular filtration rate and the increased risk of hyperkalemia with MRAs, together with polypharmacy, and suboptimal adherence, furt

Mitochondrial-centric regulatory networks in MASLD: Integrating quality control, organelle interactions, and gut-liver communication.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is closely related to liver mitochondrial dysfunction, which is driven not as an isolated event but by a self-amplifying injury loop involving impaired intrinsic quality control, aberrant organelle crosstalk, and dysregulated gut-liver signaling. This review summarizes findings in three interconnected regulatory layers: (1) intrinsic mitochondrial quality control (MQC) (PINK1/Parkin- and BNIP3/NIX-mediated mitophagy, Drp1/Mfn-driven dynamics, and chaperone/protease-maintained proteostasis); (2) organelle interactions (ER-mitochon

Environmental Factors Drive Neurodegenerative Diseases Through Glutamate Excitotoxicity: A Convergent Mechanistic Pathway.

This review illustrates how environmental stressors disrupt glutamate homeostasis via specific mechanisms: lead-induced thiol modification, manganese mediated yin yang 1 (YY1)-histone deacetylases (HDAC) repression, PM2.5-triggered microglia-astrocyte crosstalk, and advanced glycation end products (AGEs)-receptor for advanced glycation end products (RAGE)-nuclear factor kappa-B (NF-κB) signaling from high-sugar diets. Together with genetic susceptibility and pigment epithelium-derived factor (PEDF), these factors impair astrocytic glutamate uptake, promoting synaptic glutamate accumulation. Su

Proteomic profiling reveals novel insights into hypercortisolism adrenal adenomas.

Adrenal cortisol-producing adenomas (CPAs) represent the most common endocrine disorder among adrenal incidentalomas and are classified as either subclinical Cushing's syndrome (SCS) or Cushing's syndrome (CS) based on the degree of cortisol hypersecretion. The molecular mechanisms driving hypercortisolism remain poorly understood. To address this, we perform comprehensive proteomic profiling of 9 CS, 10 SCS, and 10 non-functioning adenomas (NFAs). Our analysis reveals a central role for cholesterol metabolic reprogramming in the pathogenesis of CPAs. We observe a progressive gradient of dysre

Dynamic imbalance of mitochondria-associated ER membranes in cardiovascular disease: mechanisms, context dependence and therapeutic potential.

Mitochondria-associated endoplasmic reticulum membranes (MAMs) are dynamic contact sites that coordinate structural and functional communication between the endoplasmic reticulum (ER) and mitochondria. By organizing tethering complexes, calcium channels, lipid-transfer machinery, and stress-responsive signaling modules, MAMs regulate Ca2+ homeostasis, lipid metabolism, mitochondrial dynamics, mitophagy, oxidative stress, endoplasmic reticulum stress, and cell fate determination. Increasing evidence indicates that MAMs dysfunction is critically involved in the initiation and progression of card

Role of mitochondria in neuronal function and survival in the enteric and central nervous systems.

Mitochondria are indispensable organelles that not only generate cellular energy through oxidative phosphorylation but also regulate calcium homeostasis, redox balance, and apoptotic signaling. Given the high metabolic demands of neurons, mitochondrial function and resilience mechanisms are essential for neuronal development, maturation, and survival; when these systems fail, pathological outcomes can arise. This review highlights the critical role of mitochondria in maintaining neuronal function, with discussion related to both the central (CNS) and enteric (ENS) nervous systems. We present h

From Envelope to Encephalopathy: How HIV-1 gp120 Drives Neurocognitive Decline.

Although neurons are not productively infected by HIV-1, the envelope glycoprotein gp120, detectable in cerebrospinal fluid independently of active viral replication, gains intraneuronal access via lipid raft-mediated endocytosis, macropinocytosis, and retrograde axonal transport, contributing to persistent neurobiological dysfunction within the central nervous system. Once internalized, gp120 is associated with neuronal dysfunction involving convergent pathways, including excitotoxic calcium dysregulation, mitochondrial and metabolic failure, and inflammatory and senescence-associated amplifi

Neurobiology of TRPM channels in pain processing: Current advances and future directions.

Chronic pain affects 7-10 % of the global population and remains a major clinical and socioeconomic burden. Transient receptor potential melastatin (TRPM) channels are emerging as key modulators of somatosensory processing and pain pathophysiology. This review examines the roles of major TRPM subtypes including TRPM2, TRPM3, TRPM7, and TRPM8 in diverse chronic pain states. TRPM2 is strongly associated with oxidative stress-driven neuroinflammation and migraine-related mechanisms. TRPM3 contributes to heat sensitivity, inflammatory pain, and spontaneous nociception. TRPM7 regulates calcium and

Transcriptional characteristics of human brain alterations in major depressive disorder: A systematic review.

Many patients with major depressive disorder (MDD) experience limited treatment effectiveness due to an incomplete understanding of its neurobiological underpinnings. This review integrates neuroimaging and genetic data to examine structural and functional brain changes in MDD, alongside their genetic bases. A PRISMA-guided systematic review of imaging transcriptomics over the past decade was conducted using PubMed and Web of Science. Studies included MRI scans of both MDD patients and healthy controls, as well as brain-wide gene expression data, excluding those that were purely meta-analytica
