ROS-responsive nanomicelles encapsulating celastrol ameliorate pressure overload-induced cardiac hypertrophy by regulating the NF-κB signaling pathway.

Celastrol (CEL) belongs to the group of non-steroidal immunosuppressants with the potential to improve cardiac hypertrophy (CH). However, the poor biocompatibility and low bioavailability of CEL limit its in vivo application. This study was aimed to develop a targeted drug delivery system that can efficiently and safely deliver CEL to target tissues, providing a research basis for the application of CEL in CH therapy. A novel ROS-sensitive drug-loaded nanomicelle, dodecanoic acid (DA)-phenylboronic acid pinacol ester-dextran polymer encapsulating CEL (DBD@CEL), was synthesized using chemical s

Unlocking the potential of lipid-based nanoparticles for intranasal drug delivery in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by dopaminergic neuronal loss, α-synuclein aggregation, mitochondrial dysfunction, and persistent neuroinflammation. Despite symptomatic advances, the blood-brain barrier (BBB) continues to restrict the delivery of many potentially disease-modifying agents. Intranasal (IN) administration, by exploiting direct olfactory and trigeminal pathways, offers a non-invasive means to bypass the BBB. When combined with lipid-based nanoparticles (LNPs), this route has shown promise in enhancing central nervous system target

Key Tumor Responsive ZIF-8 Nanocarriers for Effective Anti-Cancer Therapeutics.

The intricate pathological features of the tumor microenvironment (TME) collectively contribute to therapeutic resistance, thereby substantially constraining the antitumor efficacy of conventional treatments. Zeolitic imidazolate framework-8 (ZIF-8), a pH-responsive metal-organic framework material, serves not only as an efficient drug delivery system but also actively participates in TME remodeling through the release of Zn2+ upon its degradation. This dual function as both a "carrier" and an "effector" offers a synergistic mechanism to mitigate therapeutic resistance. The article reviews rec
