A Two-Component-System-Governed Regulon That Includes a β-Lactamase Gene is Responsive to Cell Envelope Disturbance.

β-Lactamase production facilitates bacterial survival in nature and affects many infection therapies. However, much of its regulation remains unexplored. We used a genetics-based approach to identify a two-component system (TCS) present in a strain of Burkholderia thailandensis essential for the regulated expression of a class A β-lactamase gene, penL, by sensing subtle envelope disturbance caused by β-lactams, polymyxin B, or other chemical agents. The genes encoding stress responses and resistance to various antibiotics were coregulated, as were the catabolic genes that enabled the B. thaila

Disulfidptosis in osteoarthritis: Role of SLC2A3 downregulation and potential therapeutic implications.

OBJECTIVE: Disulfidptosis is a newly recognized form of regulated cell death characterized by actin cytoskeleton collapse under disulfide stress. Recent studies suggest it may significantly contribute to osteoarthritis (OA), though its exact role in OA development remains unclear. This study aims to analyze the expression of disulfidptosis-related genes in OA, identifying potential biomarkers and candidate therapeutic leads. METHODS: Transcriptomic data from six datasets related to OA and single-cell RNA sequencing data were sourced from the GEO database. To identify differentially expressed

Germline mutational profile of Chinese patients under 70 years old with colorectal cancer.

BACKGROUND: Inherited susceptibility accounts for nearly one-third of colorectal cancer (CRC) predispositions and has an 80%-100% lifetime risk of this disease. However, there are few data about germline mutations of hereditary CRC-related genes in Chinese patients with CRC. This study aimed to assess the prevalence of gene mutations related to cancer susceptibility among Chinese patients with CRC, differences between Chinese and Western patients, and the phenotype-genotype correlation. METHODS: We retrospectively collected tumor samples from 526 patients with CRC under 70 years old who under

Effects of second-generation antipsychotics on human subcutaneous adipose tissue metabolism.

OBJECTIVE: Metabolic syndrome is prevalent in up to 50% of schizophrenia patients, which reduces their quality of life and their compliance with the treatment. It is unclear whether metabolic adverse effects of these agents are due to their direct effect on insulin-sensitive tissues or are secondary to increased adiposity. The study aimed to investigate the direct effects of the second-generation antipsychotics olanzapine and aripiprazole on human subcutaneous adipose tissue and isolated adipocyte metabolism. METHODS: Abdominal subcutaneous adipose tissue needle biopsies were taken from 72 he

[Effects of Epstein-Barr virus on host gene expression in Burkitt's lymphoma cell lines].

BACKGROUND AND OBJECTIVE: Epstein-Barr virus (EBV) is present in Burkitt's lymphoma (BL) cells in a latent form, showing a highly restricted pattern of gene expression with few tumor cells undergoing viral lytic replication. BL cell lines can be induced to enter the viral lytic cycle and initiate replication by stimulating surface immunoglobulin molecules. During this process many EBV genes are expressed that have the potential to influence host gene expression. We aimed to identify host genes that are regulated by EBV in BL cells and those that are regulated following ligation of surface IgG.

Analysis of Colon Transcriptomes in a Porcine Model of Dextran Sodium Sulfate (DSS)-Induced Ulcerative Colitis.

Dextran sodium sulfate (DSS) was used to induce ulcerative colitis in a porcine model to investigate the transcriptional responses in inflamed colonic tissue. Eleven pigs were divided into two groups, of which five pigs were administered an oral dose of DSS daily for five days, whereas six non-treated pigs served as a control group. Differences in transcript expression between treated and control pigs identified 425 down-regulated and 780 up-regulated mRNAs in response to DSS treatment. Fifty-nine differentially expressed miRNAs were also identified, comprising 20 up-regulated and 39 down-regu

A pilot study on identifying gene signatures as markers for predicting patient response to antiseizure medications.

The majority of the biomarkers were associated with the diagnosis of epilepsy and few of them can be applied to predict the response to antiseizure medications (ASMs). In this study, we identified 26 significantly up-regulated genes and 32 down-regulated genes by comparing the gene expression profiles of patients with epilepsy that responded to valproate with those without applying any ASM. The results of gene set enrichment analysis indicated that the ferroptosis pathway was significantly impacted (p = 0.0087) in patients who responded to valproate. Interestingly, the gene NCOA4 in this pathw

Retinoic acid signaling pathway perturbation impacts mesodermal-tissue development in the zebrafish embryo: Biomarker candidate identification using transcriptomics.

The zebrafish embryo (ZE) model provides a developmental model well conserved throughout vertebrate embryogenesis, with relevance for early human embryo development. It was employed to search for gene expression biomarkers of compound-induced disruption of mesodermal development. We were particularly interested in the expression of genes related to the retinoic acid signaling pathway (RA-SP), as a major morphogenetic regulating mechanism. We exposed ZE to teratogenic concentrations of valproic acid (VPA) and all-trans retinoic acid (ATRA), using folic acid (FA) as a non-teratogenic control com

Comprehensive and computational analysis of genes in human umbilical vein endothelial cells responsive to X-irradiation.

Radiation exposure such as A-bomb or radiation therapy is considered a major health-risk factor for cardiovascular disease. In order to understand the molecular mechanisms underlying the inflammatory reaction frequently encountered in the vascular system after exposure to ionizing radiation, we carried out a global scale microarray and computational gene expression analyses on human umbilical endothelial cells (HUVECs) exposed to X-ray (2.5 Gy). The gene ontology analysis revealed that the down-regulated genes were associated with cell cycle regulation, whereas the up-regulated genes were asso

Application of a functional genomics approach to identify differentially expressed genes in human myometrium during pregnancy and labour.

The molecular mechanisms regulating uterine relaxation and contraction during pregnancy are poorly understood. In the present study, we used for the first time a functional genomics approach applying gene array technology to identify novel candidate genes involved in the regulation of uterine quiescence and contractility during pregnancy. The purpose of this approach was to obtain a molecular snapshot of the expression profile of gene transcripts as a function of the time dependent process regulating myometrial quiescence. Using this approach, we found several genes whose expression in human m

Expression profiling of endometrium from women with endometriosis reveals candidate genes for disease-based implantation failure and infertility.

Endometriosis is clinically associated with pelvic pain and infertility, with implantation failure strongly suggested as an underlying cause for the observed infertility. Eutopic endometrium of women with endometriosis provides a unique experimental paradigm for investigation into molecular mechanisms of reproductive dysfunction and an opportunity to identify specific markers for this disease. We applied paralleled gene expression profiling using high-density oligonucleotide microarrays to investigate differentially regulated genes in endometrium from women with vs. without endometriosis. Transcriptional regulation of artemin is related to neurite outgrowth and actin polymerization in mature DRG neurons.

Artemin is a member of the glial cell line-derived neurotrophic factor (GDNF) family of ligands that helps to ensure the survival of sensory neurons. We used an in vitro isolated dorsal root ganglia model to study the effects of artemin on the adult rat neuronal system and investigate differentially regulated genes. We found that 285 genes were differentially transcribed by artemin after 3 h of treatment, including genes related to cell adhesion and actin polymerization. A series of genes involved in the regulation of actin dynamics, including coronin, Myr 5, Wiskott-Aldrich syndrome protein i

Identification of a novel gene in ROD9 island of Salmonella Enteritidis involved in the alteration of virulence-associated genes expression.

Salmonella enterica subsp. I serovar Enteritidis (S. Enteritidis), one of the causative agents for non-typhoidal gastrointestinal diseases in humans is an intracellular bacterium and mechanism for its invasion into host cells is critical to cause infection. The virulence of the pathogen is explained by the expression of genes located on its pathogenicity islands, mostly encoded under SPI-1 and SPI-2. However, S. Typhimurium SL1344, despite sharing ∼98% of its genome with S. Enteritidis P125109, lacks few regions of differences (ROD) that are hypothesized to impart virulence potential to S. Ent

Molecular Mechanism of Sterilization by Quinestrol, a Rodent Sterility Control Agent.

Sterility control is one of the key tools for regulating pest rodent population density. An in-depth analysis of the molecular mechanism of sterility caused by control agents is of great significance for further exploration of novel sterility controls and the development of alternative drugs. In this study, male plateau zokors (Eospalax baileyi) in the breeding period were tested to explore the molecular mechanism of quinestrol-induced sterility. We used RNA-seq technology to investigate key genes and signaling pathways associated with the inhibition of testicular development and spermatogenes

Identification of Genes and microRNAs Associated with Midfacial Hypoplasia in Mice.

Midfacial hypoplasia is a developmental defect caused by insufficient growth of the nasal placodes and maxillary prominences. While genetic studies in mice have identified key genes involved in midfacial development, the regulation of these genes during craniofacial development remains poorly understood. In this study, we demonstrate that microRNAs, short non-coding RNAs, play a crucial role in regulating genes involved in midfacial development. We identified 224 genes associated with midfacial malformations in mice. Through bioinformatics analyses, we predicted that several microRNAs, specifi
