Exome-Wide Association Analysis Identifies Rare Germline Susceptibility Variants in Early-Onset Breast Cancer Among Saudi Women.

Early-onset breast cancer (EOBC) is disproportionately common in Saudi Arabia, where women present nearly a decade earlier than in Western countries, suggesting unique inherited susceptibility. While BRCA1/2 explain part of the hereditary risk, the contribution of rare coding variants in Arab EOBC remains unclear. Whole-exome sequencing was performed on germline DNA from 102 unrelated Saudi EOBC patients and 1395 cancer-free controls recruited from the same national Saudi cohort. Rare variants were defined by stringent frequency and quality thresholds and classified as rare loss-of-function (R

Comprehensive Evaluation of Safety and Probiotic Potential of Lactiplantibacillus plantarum HY7715 Isolated from Korean kimchi.

Lactiplantibacillus plantarum HY7715, isolated from Korean kimchi, has been previously reported to overproduce riboflavin in vitro and in vivo, indicating its functional potential as a probiotic candidate. Given this characteristic, a comprehensive safety and genomic evaluation is required prior to application. In this study, we assessed the safety, genomic stability, and probiotic-related properties of HY7715 through whole-genome sequencing, comparative genomics, and phenotypic analyses. The genome (3,348,486 bp; 44.29% GC content) was assembled into seven contigs and encoded 3,224 protein-co

The Evolutionary Maintenance of Amino Acid Prototrophy in Escherichia coli.

Escherichia coli is a prototroph and can synthesize all twenty proteinogenic amino acids when required to grow in minimal medium. There are approximately sixty protein-coding genes individually essential for amino acid synthesis. This is a large mutational target for the accumulation of detrimental mutations. E. coli can rewire biosynthetic pathways in response to mutational damage, but the limits of this capacity are poorly understood. Here, to address evolutionary robustness, we asked whether and how the phenotypes of irreversible mutations causing auxotrophy could be suppressed or bypassed

Genetic analyses enabled by the fourth chromosome resource project reveal unexpected mutant phenotypes and suggest new disease models.

To date, the Fourth Chromosome Resource Project (FCRP) has deposited over 850 stocks for the genetic analysis of its 79 protein-coding genes. Here, we employ those stocks to reveal unexpected phenotypes for multiple exemplar genes. Expression studies of the transcript and protein provide clues to an adult function for maverick, a gene that, despite prior efforts, has remained inscrutable. Loss-of-function studies reveal an adult brain phenotype for the well-studied signal transducer legless and the first phenotype for datilographo, a gene with no prior mutations. Marked clones with a new eyele

Gestational Diabetes Mellitus Increases the Risk of Atopic Dermatitis in Children: A National Retrospective Study with Analysis by Treatment Modality.

INTRODUCTION: Atopic dermatitis (AD) is a common childhood inflammatory disease influenced by genetic and environmental factors. Maternal metabolic conditions, including gestational diabetes mellitus (GDM), may affect offspring risk, but large population-based studies are limited. We aimed to examine the association between maternal GDM and offspring AD, and whether risk varies by maternal diabetes treatment type. METHODS: We conducted a retrospective cohort study of 326,611 mother-child dyads from Clalit Health Services, Israel, between 2010 and 2023. GDM was classified as diet-controlled (A

Riluzole Restores Circuit and Behavioral Function Altered by Allele-Specific Expression-Mediated LINC02449-CPLX1 Dysregulation.

BACKGROUND AND HYPOTHESIS: Allele-specific expression (ASE) of long non-coding RNAs (lncRNAs) links genetic variation to synaptic dysfunction in psychiatric disorders. We previously identified a disease-associated ASE shift in LINC02449 at rs149707223 (C/G), with preferential expression of the alternative G allele in bipolar disorder (BD) and schizophrenia (SZ). Overexpression of LINC02449-G induced social deficits, repetitive behaviors, and enhanced excitatory synaptic transmission in the medial prefrontal cortex-nucleus accumbens (mPFC-NAc) circuit via upregulation of CPLX1. We hypothesized

Genomic characterisation and lytic potential of phage SF01 against multidrug-resistant Salmonella enterica subsp. enterica, a key agent of infection in poultry.

1. Salmonella enterica remains the key cause of salmonellosis in poultry, causing high morbidity and mortality. Due to the unprecedented resistance of S. enterica to antibiotics, bacteriophages have emerged as a powerful alternative to conventional antibiotics treatment for salmonellosis.2. In this study, a strain was isolated from infected broiler chickens and whole-genome sequencing (WGS) identified the strain SFD-01 as S. enterica subsp. enterica. Bioinformatics analyses revealed that the genome was 4.6 Mb in size with 4559 coding sequences (CDS), 77 tRNAs and 4 rRNAs. Additionally, 119 vir

Exploring biomarkers for treatment response in psoriatic arthritis: a focus on multi-omics technologies.

Psoriatic arthritis (PsA) is a complex inflammatory disease characterised by a combination of cutaneous and musculoskeletal symptoms, which complicates both diagnosis and management. Despite advances in targeted therapy, the treatment of PsA remains challenging, with up to 40% of patients experiencing inadequate response. Predictive biomarkers may serve as valuable tools to help clinicians select the most appropriate therapy at the optimal time. However, despite substantial research activity, the current evidence remains inconclusive. Moreover, biomarkers that predict treatment response may di

Development of a recombinant single-cycle influenza viral vector as an intranasal vaccine against SARS-CoV-2.

The COVID-19 pandemic has demonstrated the detrimental potential of zoonotic coronavirus transmission to human populations. Effective vaccines capable of eliciting immunity to SARS-CoV-2 have been pivotal in mitigating the spread of the virus. In this study, we describe the generation of a non-replicating pseudotyped influenza A virus (S-FLU), where the native haemagglutinin (HA) sequence is replaced with the coding sequence of either a membrane-anchored form (TM) or secretory form (Sec) of the receptor-binding domain (RBD) of the ancestral SARS-CoV-2 Wuhan (S-RBD Wuhan). We showed that both S

A highly penetrant LMNA R541C variant associated with dilated cardiomyopathy leads to dysregulation in metabolism and proliferation pathways in stem cell-derived cardiomyocytes.

BACKGROUND: LMNA codes a widely expressed nuclear cytoskeletal protein (lamin A/C) with multiple important functions. Pathogenic LMNA genetic variation may lead to autosomal dominant cardiomyopathy, though the severity and rate of progression can vary with the specific nucleotide change and location. Prior studies showed that induced pluripotent stem cells (iPSC)-derived cardiomyocytes (iCMs) with LMNA R541C exhibited reduced LMNA protein abundance, increased sarcomere disorganization, and abnormal electrophysiology. METHODS: We investigated the LMNA-R541C variant that exhibits a highly penet

Transcriptomic plasticity in hybrid schistosomes can contribute to their zoonotic potential.

Hybrids between Schistosoma haematobium and Schistosoma bovis contribute to human and animal infections, highlighting complex interspecies interactions that facilitate schistosomiasis transmission. Schistosoma bovis infects multiple ruminant hosts, promoting cross-species transmission and increasing zoonotic risk. This study explores transcriptomic plasticity as a mechanism enabling hybrid schistosomes to adapt to different definitive hosts. We analysed two contexts: (1) introgressed S. haematobium × S. bovis hybrids, which exhibited higher virulence in sheep than parental S. bovis; and (2) S.

Field-isolate recombinant tick-borne encephalitis viruses define reporter-stability guidelines for antiviral testing in flaviviruses.

As arthropod-borne viruses continue to threaten populations globally, there is a pressing need for experimental systems that enable rapid antiviral discovery. Reverse-genetics platforms producing recombinant reporter orthoflaviviruses have been developed to address this gap. Here, we present two new recombinant tick-borne encephalitis viruses (TBEVrec) generated on a European-subtype Haselmühl Tiho1 isolate backbone. A reporter gene, either eGFP or Nluc, was inserted in the capsid-coding region of the genome downstream of the capsid RNA regulatory signal and separated from the complete viral p

Chromosome-level genome assembly of the Endangered scaly-sided merganser Mergus squamatus with insight into its demographic history.

The scaly-sided merganser, Mergus squamatus, is an Endangered piscivorous duck which has been declining since the late 1900s due to habitat loss, over-hunting, and climate change. Despite being a species of global conservation concern and subject to ex- and in-situ conservation efforts, genomic research has been limited, hindering our understanding of its population genetic status and evolutionary history. In this study, we present the first fully annotated, chromosome-level genome for the scaly-sided merganser, generated using Oxford Nanopore long reads, Illumina short reads, and Hi-C sequenc

Regulation and Interaction Among SOCS1 and SOCS3 by MicroRNAs in Multiple Sclerosis: A Review and In Silico Analysis.

Multiple sclerosis (MS) is a chronic demyelinating disease and a leading cause of disability in young adults. Its pathophysiology involves complex genetic and immunological mechanisms, among which the suppressors of cytokine signaling (SOCS) proteins play a critical role. These intracellular regulators control cytokine-mediated JAK/STAT signaling, thereby modulating immune activation. In particular, SOCS1 and SOCS3 inhibit key cytokines such as IL-2, IL-12, IL-6, and IL-23, preventing excessive Th1/Th17 responses and neuroinflammation. Dysregulation of SOCS expression or function can favor per

RNA-based therapies for neurodegenerative disease: Targeting molecular mechanisms for disease modification.

Neurodegenerative diseases such as Alzheimer's disease (AD), Parkinson's disease (PD), and Huntington's disease (HD) are characterized by progressive neuronal damage, protein aggregation, and chronic inflammation, leading to cognitive and motor impairments. Despite symptomatic relief from current therapies, disease-modifying treatments targeting the core molecular mechanism are still lacking. RNA-based therapies offer a promising approach to treating neurodegenerative disease by targeting molecular mechanisms such as gene expression, protein synthesis, and neuroinflammation. Therapeutic strate
