Highly Sensitive Detection of Chymotrypsin Using Gold Nanoclusters with Peptide Sensors.

Pancreatic function tests are used to determine the presence of chronic pancreatitis, particularly in the early stage of the disease. Chymotrypsin is an indicator of pancreatic function and is thus related to pancreatic diseases. However, these methods often require specific equipment and cannot always meet on-site analysis requirements. Consequently, a highly sensitive detection method needs to be developed. This research employed graphene oxide modified with NHS sensors and peptides (RRHFFGC: Arginine-Arginine-Histidine-Phenylalanine-Phenylalanine-Glycine-Cysteine) tagged with gold nanoclust

Electrosynthesis of C6 Chemicals by Propylene Oxidative Coupling on Au Surface.

Electrocatalytic oxidation of alkenes represents a promising pathway for sustainable fine chemical synthesis in future chemical manufacturing. However, anodic propylene transformations typically yield only oxygenation products, such as propylene glycol, propylene oxide, acrolein, or acetone, lacking the important electro-driven C-C coupling process. Here, we report a C-C coupling pathway for propylene oxidation on Au, producing two C6 compounds: 2,5-hexanedione and 3-hexene-2,5-dione. Under mild conditions, the total Faradaic efficiency for C6 products reaches 36%. Combining in situ infrared s

Preparation and Application of a Composite Formulation of Mint Essential Oil Nanoemulsion.

To develop an efficient and low-toxic green plant-based insecticide and address the issues of rapid volatilization and poor stability of mint essential oil, this study prepared a mint essential oil nanemulsion composite formulation using Tween 80 as the surfactant and propylene glycol as the cosurfactant. The insecticidal activity of the formulation was tested against fall armyworm, Spodoptera frugiperda and corn aphid, Rhopalosiphum maidis. Results showed that the optimal formulation consisted of a surfactant to cosurfactant mass ratio of 3:1 and a mixed surfactant to mint essential oil mass

Oriented Dipoles in Ordered Ensembles of Confined Lead Halide Perovskite Nanocrystals.

Here, we demonstrated directed self-assembly of quasi-2D cesium lead bromide perovskite nanoplates by liquid-air interfacial assembly. Due to their ionic crystal nature, perovskite nanocrystals are susceptible to degradation by most polar immiscible sublayer solvents (acetonitrile, diethylene glycol). We used glyceryl triacetate as a liquid substrate for nanoplate self-assembly. By tuning the interfacial energy and volume fraction of nanoplates, we achieved monolayer ensembles of nanoplates with face-down and edge-up ordering >100 μm2 in area. Controlled ordering of these confined structures a

Single-Nanoparticle Dynamics in Opto-Thermal Tweezers: Resolving the Temporal Resolution of Depletion Force Trapping.

Optothermal tweezers enable the manipulation of a wide range of nano-objects through optically induced depletion forces. Despite significant advances, the temporal dynamics of optothermal trapping remain elusive as existing methodologies rely almost exclusively on time and ensemble averaging. Consequently, stable trapping cannot be distinguished from local transient accumulation, where the time-averaged concentration increases, but particles exhibit rapid, dynamic motion in and out of the trap. Here, we investigate optothermal trapping with single-nanoparticle-level analysis and submillisecond

Revealing the bond exchange and network rearrangement mechanism in vitrimers.

Vitrimers combine thermosets stability with thermoplastic reprocessability, yet their rational design is hindered by an insufficient understanding of their viscoelasticity. We investigated the fundamental relationships among segmental dynamics, bond exchange kinetics, and network rearrangements in model poly(propylene glycol)-based vitrimers. Our results show that the bond rearrangement time (τr) and its temperature dependence remain nearly unchanged at high temperatures, where their segmental relaxation time (τα) differs by several orders. This indicates that rate-limited bond exchange kineti

Poly(sarcosine) Lipid Synthesis From CO2-Based Sarcosine-N-Carboxyanhydride.

Polymer lipids (PLs) are an essential component for the stability of lipid nanoparticles (LNPs) for gene delivery. Poly(ethylene glycol) (PEG)-based lipids are the current gold standard. However, they are suspected to be responsible for rare adverse reactions to the LNP-based COVID-19 vaccines. Therefore, alternative PLs are being intensively investigated. A particularly promising alternative are poly(sarcosine) (PSar)-based lipids. However, one significant bottleneck of pSar-based biomaterials is the synthesis of the monomer sarcosine-N-carboxyanhydride (Sar-NCA). Current methods rely on high

Design, Synthesis, and Evaluation of [68Ga/177Lu]-Labeled TSPO-Targeting Radioligands for PET/CT Diagnosis and Radionuclide Therapy of Brain Tumor.

The translocator protein (TSPO) has gained increasing attention in the diagnosis and treatment of brain cancers. Current TSPO-targeting radiotracers, predominantly labeled with 11C and 18F, limit the incorporation of emerging radionuclides into theranostic applications. In this study, we designed and synthesized four DOTA-conjugated TSPO-targeting precursors featuring various polyethylene glycol (PEG) linkers, which enabled straightforward and efficient labeling with 68Ga or 177Lu. In vitro and in vivo evaluations demonstrated that compared with [18F]FDG, all four 68Ga-labeled radioligands exh

Rational Design and Screening of Chemically Modified Anti-SARS-CoV-2 siRNA.

To realize the therapeutic potential of RNA interference against severe acute respiratory syndrome coronavirus 2, strategic chemical modifications are crucial for enhancing the siRNA stability and potency. In this study, 200 siRNAs were screened to identify effective candidates that targeted the highly conserved RNA-dependent RNA polymerase gene and identified VS-38m as a lead compound. Building on this scaffold, we systematically evaluated a series of modification patterns by introducing 2'-fluoro (2'-F) and 2'-O-methyl (2'-OMe) substitutions at specific positions on both the guide and sense

Rapid and improved surface passivation method for Single-Molecule experiments.

Single-molecule fluorescence experiments are a powerful tool for studying biomolecular interactions, including protein dynamics and oligomerization, protein-protein interactions, and protein-nucleic acid interactions. Biomolecules are commonly immobilized on the microscope surface to extend the observation time. However, non-specific interactions between biomolecules and the surface present a major challenge. The first critical step in these experiments is preparing the surface using polyethylene glycol (PEG) coated slides, which facilitate biomolecule immobilization while minimizing non-speci

IAPP surface-induced aggregation and corilagin's inhibitory effect.

Proteins and peptides spontaneously assemble into various structured supramolecular entities that perform vital physiological functions. In pathology and biopharmaceutics, protein assemblies can form cytotoxic oligomers or protofibrils. Most studies focus on aggregation and inhibition processes in bulk solutions. However, local surface interactions are largely responsible for protein structural changes leading to aggregation. To date, research on surface-induced protein aggregation remains insufficient to comprehend underlying mechanisms. This work reports a systematic process to produce islet

Selective mRNA Delivery to Activated Macrophages via Hyaluronic Acid-Functionalized Lipid Nanoparticles with Optimized PEGylation.

Activated proinflammatory macrophages are associated with various inflammatory diseases, and due to their overexpression of the CD44 receptor, they may be targeted for therapy by hyaluronic acid (HA), its natural ligand. This study aimed to develop lipid nanoparticles (LNPs) functionalized with HA and stabilized with an optimized amount of poly(ethylene glycol) (PEG) for targeted mRNA delivery to activated macrophages. Using microfluidic mixing, LNPs were produced with either 1.5% PEG (LNP1.5%PEG) or 0.5% PEG (LNP0.5%PEG). HA-coated LNPs (HA-LNPs) were prepared by postinsertion of an HA-DPPE c

Macrophage Responses to Silica Nanoparticles: Role of Physicochemical Properties and Surface Modification.

Silica nanoparticles are widely studied nanomaterials for biomedical applications owing to their tunable physicochemical properties, such as size, porosity, geometry, and surface modification. Despite their promising potential, concerns regarding their safety continue to limit clinical translation. In this study, we systematically investigated how key physicochemical parameters and surface attachment of poly(ethylene glycol) (PEG) affect the cytotoxicity and immune activation profiles of silica nanoparticles in macrophages. A structurally diverse set of silica nanoparticles (rod, spherical, po

Composite microspheres of bioactive glass and hydrophilic biodegradable polymers to accelerate infected wound healing.

Synthetic biodegradable microspheres hold great promise for complex wound repair. However, their clinical application is hindered by inflammatory responses triggered by acidic degradation byproducts. In this paper, we developed new composite microspheres as dressings to accelerate infected wound healing. Polyethylene glycol/poly(L-lactide)/poly(ε-caprolactone) multiblock copolymers were synthesized and engineered with Cu-doped bioactive glass to form composite microspheres. The Cu-doped bioactive glass not only improved the hydrophilicity of the composite microspheres but also neutralized the

N-halamine modified cotton dressing with thermosensitive antibacterial-antifouling switchable surface for wound healing.

Surface biofouling of cotton gauze dressings greatly promotes biofilm formation and delays wound healing. Herein a reversibly thermoresponsive switchable biointerface strategy is proposed to regulate the antibacterial and antifouling activity of cotton dressings for promoting wound healing. Poly(ethylene glycol) (PEG) brushes are covalently grafted onto cotton fibers to form a durable antifouling underlayer, followed by in situ polymerization of poly(N-isopropylacrylamide) (PNI) and subsequent chlorination to generate N-halamine antibacterial groups of N-Cl. In the initial antibacterial state,

Lurasidone hydrochloride-loaded microparticle-based long-acting injectable: formulation optimisation, characterisation, and payload release kinetics-a proof of concept study.

Lurasidone hydrochloride is a promising antipsychotic agent. The currently marketed formulation (Latuda®) of lurasidone hydrochloride exhibits extremely poor bioavailability and requires daily dosing. Current research focuses on the development and optimisation of a lurasidone hydrochloride-loaded poly(lactic-co-glycolic acid) microparticle-based long-acting depot formulation. The optimised batch of microparticles showed 32.81 ± 1.46 µm particle size, 95.74 ± 3.19% an entrapment efficiency, and a drug loading of 27.68 ± 0.95%. POSS as a versatile platform for developing multifunctional water-soluble photoinitiators.

Conventional photoinitiators act only as non-structural initiating species and do not regulate network topology after radical generation. In this work, a strategy for actively modulating hydrogel crosslinking behavior through photoinitiator design is presented. Polyhedral oligomeric silsesquioxane-based nanophotoinitiators (POSS-PIs) were synthesized by functionalizing POSS with polyethylene glycol (PEG) chains of varying lengths and coupling them with I2959. Unlike conventional initiators, POSS-PIs function as multifunctional junctions that participate in network formation. This dual function
