Integrative and conjugative elements (ICEs) of the SXT/R391 family are mobile genetic elements that integrate into the bacterial host chromosome and can be transferred horizontally, spreading antimicrobial resistance genes. Our study aimed to evaluate aspects of the relationship between ICEPmiJpn1, one of the most widespread SXT/R391 variants, with its natural host Proteus mirabilis. For this investigation, we used isogenic strains (containing or not the ICEPmiJpn1) that enabled us to evaluate the influence of this element on several physiological aspects of P. mirabilis as well as the effect

BACKGROUND: The aims of this study were to evaluate the host-tick-pathogen interface of Babesia spp. and Anaplasma phagocytophilum in restored areas in both questing and host-attached Ixodes ricinus and Dermacentor reticulatus and their small mammalian hosts.
METHODS: Questing ticks were collected from 5 sites within the city of Leipzig, Germany, in 2009. Small mammals were trapped at 3 of the 5 sites during 2010 and 2011. DNA extracts of questing and host-attached I. ricinus and D. reticulatus and of several tissue types of small mammals (the majority bank voles and yellow-necked mice), were

UNLABELLED: A rising theme among intracellular microbes is the delivery of ankyrin repeat-containing effectors (Anks) that interact with target proteins to co-opt host cell functions. Orientia tsutsugamushi, an obligate intracellular bacterium and the etiologic agent of scrub typhus, encodes one of the largest Ank repertoires of any sequenced microorganism. They have been previously identified as type 1 secretion system substrates. Here, in silico and manual sequence analyses revealed that a large proportion of O. tsutsugamushi strain Ikeda Anks bear a eukaryotic/poxvirus-like F-box motif, whi

Zika virus (ZIKV) targets the neural progenitor cells (NPCs) in brain during intrauterine infections and consequently causes severe neurological disorders, such as microcephaly in neonates. Although replicating in the cytoplasm, ZIKV dysregulates the expression of thousands of host genes, yet the detailed mechanism remains elusive. Herein, we report that ZIKV encodes a unique DNA-binding protein to regulate host gene transcription in the nucleus. We found that ZIKV NS5, the viral RNA polymerase, associates tightly with host chromatin DNA through its methyltransferase domain and this interactio

This narrative review seeks to examine the relationships between bacterial microbiomes and infectious disease. This is achieved by detailing how different human host microbiomes develop and function, from the earliest infant acquisitions of maternal and environmental species through to the full development of microbiomes by adulthood. Communication between bacterial species or communities of species within and outside of the microbiome is a factor in both maintenance of homeostasis and management of threats from the external environment. Dysbiosis of this homeostasis is key to understanding th

Internal ribosome entry sites (IRES) are structured RNA elements that facilitate cap-independent translation initiation. IRES activity is regulated by its interactions with host and viral RNA-binding proteins (RBPs), collectively known as IRES trans-acting factors (ITAFs), which modulate RNA folding, ribosome recruitment, and translation efficiency. Characterizing these RNA--protein interactions provides valuable insights into the mechanisms that regulate IRES activity. Hepatitis E virus (HEV), is a positive-sense, single-stranded RNA virus. Our earlier study discovered an IRES-like (IRESl) el

As an obligate biotrophic pathogen, Puccinia triticina (Pt) subverts wheat immunity via effector delivery. However, the molecular mechanisms underlying Pt effector-mediated wheat susceptibility remain largely unexplored. In this study, an effector Pt_20 that is induced expression at the early stage of Pt infection to wheat was identified. Interestingly, the Pt_20 gene exhibits two distinct transcriptional isoforms (Pt_20636, Pt_20807) among Pt races and both are localized in the nucleus, cytoplasm, and plasma membrane. Pt_20636 was able to suppress BAX-mediated cell death in Nicotiana benthami

MYB transcription factors play crucial roles in orchestrating plant immunity against various pathogens. However, their regulation during virus infection and how viruses counteract these defenses remain poorly understood. In this study, we demonstrated that turnip mosaic virus (TuMV) viral genome-linked protein (VPg) interacts with Nicotiana benthamiana MYB330 (NbMYB330), an R2R3-MYB transcription factor, in the nucleus. NbMYB330 expression was activated at the early stage of TuMV infection, and NbMYB330 positively regulated host antiviral defense. NbMYB330 directly bound to the promoter region

The hepatitis B virus (HBV) X protein (HBx) functions as a versatile regulatory molecule that is indispensable for both efficient HBV replication and the pathogenesis of HBV-related hepatocellular carcinoma (HCC). Accumulating studies indicate that a broad spectrum of post-translational modifications (PTMs), including ubiquitination, phosphorylation, NEDDylation and ISGylation, critically influence HBx stability, intracellular trafficking, and its capacity to engage with diverse host factors. Collectively, these PTMs constitute a complex modulatory network that governs viral persistence and co

Respiratory infections during critical periods of lung development can shape immune responses and cause lasting alterations in pulmonary structure and function. Lung development begins in utero, with airway branching, alveolarisation and epithelial differentiation continuing postnatally, making the lungs particularly vulnerable to early-life insults. Early-life respiratory syncytial virus infections are a major cause of severe respiratory diseases. Furthermore, risks are amplified by preterm birth and specific maternal environmental exposures. In this review, we discuss how early-life respirat

Comparative transcriptomics offers a powerful approach to elucidate host-virus interactions across related pathogens, yet systematic evaluations across species-matched cellular systems remain limited. We performed a cross-species RNA sequencing analysis of respective species' cells infected with three alphaherpesviruses-herpes simplex virus 1 (HSV-1), bovine alphaherpesvirus 1 (BHV-1), and equid alphaherpesvirus 1 (EHV-1)-to dissect conserved and virus-specific transcriptional responses. We show that certain orthologous genes and orthologous pathways are differentially regulated upon infection

Gpi7 gene, encoding the catalytic subunit of GPI ethanolamine-phosphate (Etn-P) transferase II, is primarily involved in the synthesis, maturation, and sorting of GPI-anchored proteins (GPI-APs), thereby playing a crucial part in cell wall functions and host-pathogen interactions. This study aimed to investigate the role of Gpi7 (here designated CcGpi7) in Corynespora cassiicola, a devastating fungal pathogen causing leaf spot in cucumber and many other cash crops. We systematically identified and inventoried 138 GPI-APs in C. cassiicola, followed by a detailed structural characterization of t…

Porcine deltacoronavirus (PDCoV) is an emerging enteric pathogen that poses significant economic threats to the swine industry and carries potential zoonotic risk. However, the interactions between PDCoV and host innate immunity, particularly those involving interferon-stimulated genes (ISGs), remain poorly understood. Porcine myxovirus resistance protein 1 (pMx1), a well-characterized ISG with broad-spectrum antiviral activity, has not been investigated in the context of PDCoV infection. This study demonstrates that PDCoV infection upregulates endogenous pMx1 expression in porcine intestinal

Microparticles (MPs) serve as critical mediators of intercellular communication by shuttling bioactive molecules, holding significant potential for clinical diagnostics and next-generation vaccine development. Brucella, an intracellular pathogen responsible for severe zoonosis, evades host immunity through persistent infection; however, the role of Brucella-induced MPs in modulating adaptive immunity remains poorly defined. In this study, we demonstrate that the infection of RAW264.7 macrophages with the attenuated Brucella melitensis strain M5 triggers robust MP release. These MPs enhance hos

Zika virus (ZIKV) can persist in mosquito populations through both horizontal and vertical transmission, and successful ovarian infection is a prerequisite for transgenerational spread. However, the mosquito host factors that regulate ovarian permissiveness and vertical transmission remain poorly characterized. Here, we investigated the role of cytochrome P450 4G15 (CYP4G15) in ZIKV infection and vertical transmission in Aedes albopictus and identified an associated downstream pathway involving SORD-like, a predicted sorbitol dehydrogenase-like gene. Expression profiling revealed that CYP4G15

Viruses exploit host genetic machinery to establish chronic infections and contribute to chronic neurodegenerative and immune disorders. By manipulating DNA methylation, histone modifications, and non-coding RNA networks, viruses such as Epstein-Barr virus (EBV), Herpes Simplex virus (HSV), Zika virus (ZIKV), and Human immunodeficiency virus (HIV) induce epigenetic changes that silence or delay the host antiviral defenses and reprogram host gene expression. For instance, EBV recruits DNA methyltransferases (DNMTs) to hypermethylate genes such as SOCS1 while ZIKV disrupts neuronal DNA methylati

Antiretroviral therapy (ART) effectively suppresses HIV-1 replication but does not purge the latent HIV-1 reservoir. Strategies aimed at HIV-1 latency reversal and subsequent elimination of infected cells are being explored. Targeting the inhibitor of apoptosis proteins (IAP) and DEAD-box polypeptide 3 (DDX3) RNA helicase reduces the HIV-1 reservoir ex vivo. However, the mechanisms driving apoptosis of HIV-1 infected cells remain unclear. Here, we uncovered the mechanism regarding HIV-1 transcriptional activation and induction of apoptosis specific for HIV-1 infected cells using an acute in vi

Do viral infections accelerate aging, and does this acceleration scale with pathogenicity? Using transcriptomic aging clocks, we measured biological age in Drosophila melanogaster infected with four enteric RNA viruses spanning a broad pathogenicity range (i.e., reduction of host lifespan). All pathogenic infections accelerated aging and the magnitude of acceleration tracked pathogenicity. This pattern held across oral and systemic infection routes and was conserved in Caenorhabditis elegans where the nonpathogenic Orsay virus produced negligible aging acceleration. Pathway analysis indicated

Inflammatory microenvironments regulate immune cell differentiation, activation, and programming. Although traditionally considered terminally differentiated effectors, neutrophils can acquire distinct functional states in response to environmental cues. To examine context-dependent neutrophil programming, cells from healthy donors were cultured in vitro under cytokine regimens mimicking opposing inflammatory environments (GM-CSF/IFN-γ or IL-4/IL-13/TGF-β), alone or combined with PMA and Mycobacterium tuberculosis, to assess functional and transcriptional responses. Cryptosporidium parvum is a water and foodborne apicomplexan pathogen that vigorously disrupts host signaling networks to enable intracellular colonization. However, the parasite-encoded effectors responsible for this modulation and the specific host signaling cascades they involve is still unexplored. In this study, we identify CpCML, a calmodulin-like protein encoded by C. parvum, as a perilous virulence effector that modulates host PI3K-NF-κB signaling and autophagic flux through direct interaction with the host protein SUGT1. Biochemical and cellular analyses reveal that CpCML binds SUGT1
