Global regulation of nucleotide biosynthetic genes by c-Myc.

BACKGROUND: The c-Myc transcription factor is a master regulator and integrates cell proliferation, cell growth and metabolism through activating thousands of target genes. Our identification of direct c-Myc target genes by chromatin immunoprecipitation (ChIP) coupled with pair-end ditag sequencing analysis (ChIP-PET) revealed that nucleotide metabolic genes are enriched among c-Myc targets, but the role of Myc in regulating nucleotide metabolic genes has not been comprehensively delineated. METHODOLOGY/PRINCIPAL FINDINGS: Here, we report that the majority of genes in human purine and pyrimid

Spectroscopic characterization of heme iron-nitrosyl species and their role in NO reductase mechanisms in diiron proteins.

Nitric oxide (NO) plays an important role in cell signalling and in the mammalian immune response to infection. On its own, NO is a relatively inert radical, and when it is used as a signalling molecule, its concentration remains within the picomolar range. However, at infection sites, the NO concentration can reach the micromolar range, and reactions with other radical species and transition metals lead to a broad toxicity. Under aerobic conditions, microorganisms cope with this nitrosative stress by oxidizing NO to nitrate (NO3−). Microbial hemoglobins play an essential role in this NO-detox

Brewing controversies: Darwinian perspective on the adaptive and maladaptive effects of caffeine and ethanol as dietary autonomic modulators.

Ethanol and caffeine are two of the oldest human drugs. Their pervasive integration into the modern human diet may reflect behavioral attempts to correct maladaptations induced by evolutionary displacement of the autonomic system. The dietary adoption of caffeine may parallel the emergence of cognition as an independent basis of competition. Enhancement of the cognitive ability to gather and process information likely evolved as a valuable adjunct to physical behavior in prehistoric fight-or-flight encounters. Caffeine effectively exploits this pre-existing association between adrenergic activ

Loss of Brca2 and p53 synergistically promotes genomic instability and deregulation of T-cell apoptosis.

BRCA2 is a breast cancer susceptibility gene of which the product is thought to be involved in monitoring genome integrity and cell cycle progression. Brca2-null mice have a defect in embryonic cellular proliferation and die in utero. Here we report the generation of T-cell lineage-specific Brca2-deficient (tBrca2(-/-)) mice using the Cre-loxP system. Mice with a flanked by loxP allele of Brca2 were crossed to transgenic mice bearing Cre recombinase driven by the T cell-specific promoter Lck. Thymic cellularity and distribution of subset populations were normal in tBrca2(-/-) mutants. Thymocyt

Mechanical, translucency, tribological resistance, and biocompatibility of lithium disilicate glass-ceramics produced by ZrO2 doping and digital light processing for dental prosthesis.

BACKGROUND: Lithium disilicate glass-ceramics (LDGCs) have become one of the most widely used dental prosthesis materials in clinical practice due to their excellent esthetics, biocompatibility, and enamel-like wear resistance. However, their inherent brittleness, limited mechanical strength, and progressive wear under long-term service have restricted their application in high-stress posterior regions. Herein, this study aims to develop zirconia (ZrO2)-reinforced LDGCs and systematically investigate the effects of ZrO2 content on microstructure evolution and mechanical, translucency, tribolog

Mitochondrial Protection by Trifolirhizin Alleviates Primary Sjögren's Syndrome and Liver Injury via Coordinated Suppression of the ROS/cGAS-STING Pathway.

BACKGROUND: Autoimmune diseases such as primary Sjögren's syndrome and type 1 diabetes are frequently complicated by hepatic injury, yet therapies that simultaneously target inflammation and parenchymal damage remain limited. Mitochondrial dysfunction with excessive reactive oxygen species (ROS) production drives a self-amplifying pathogenic loop by activating the cGAS-STING innate immune pathway. We previously observed that a Chinese herbal formula preserved mitochondrial ultrastructure in autoimmune NOD mice, and computational screening identified trifolirhizin-a natural pterocarpan flavonoi

Prehabilitation Program for Lung and Esophageal Cancers (Boosting Recovery and Activity Through Early Wellness): Protocol for a Nonrandomized Trial.

BACKGROUND: Cancer is the leading cause of death in Canada, responsible for 28.2% of all deaths. Based on surgical candidacy and disease status, both lung and esophageal cancer are treated through surgical resection by a thoracic surgeon. Although surgery contributes to improved outcomes, the 30-day postoperative mortality risks are as high as 10% and 2.8%, respectively. Evidence has shown that prehabilitation is a way in which patients can have improved postoperative outcomes. Prehabilitation is multimodal, often including some form of movement, nutrition, stress management, and smoking cessa

Enhancing Distress Tolerance Skills in Adolescents With Anorexia Nervosa Through the BALANCE Mobile App: Feasibility and Acceptability Study.

BACKGROUND: Anorexia nervosa is a severe psychiatric disorder with high morbidity and mortality, particularly among adolescents. Family-based treatment (FBT) is the leading evidence-based intervention for adolescent anorexia nervosa, involving parents in renourishment and behavior interruption. Despite its effectiveness, challenges in distress tolerance and emotion regulation during high-stress situations, such as mealtimes, contribute to suboptimal treatment outcomes, with only 35% to 50% of adolescents achieving full recovery. Enhancing distress tolerance skills during FBT may improve treatm

Therapy-Induced Cellular Senescence in Non-Hodgkin Lymphomas, with Emphasis on Aggressive B-Cell Subtypes: Molecular Mechanisms and Emerging Drug Targets.

INTRODUCTION/BACKGROUND: Therapy-Induced Senescence (TIS) is increasingly recognized as a clinically relevant outcome of anticancer therapy, particularly after sublethal genotoxic stress. In Non-Hodgkin Lymphomas (NHL), TIS may coexist with apoptosis and contribute to treatment heterogeneity by enabling the long-term survival of dam-aged but viable tumor cells. The aim of this article is to synthesize the core molecular mechanisms of TIS in NHL, and to highlight druggable senolytic and senomorphic strate gies that may improve disease control and reduce the risk of relapse. MATERIAL AND METHOD

Inflammatory mechanisms contribute to long-term cognitive deficits induced by perinatal asphyxia via interleukin-1.

Perinatal asphyxia remains a leading cause of neonatal mortality and a major contributor to permanent neurological deficits. Even mild cases can result in long-term neurodevelopmental, cognitive, behavioural and psychiatric disorders. However, the mechanisms underlying asphyxia-induced hypoxic-ischaemic brain injury remain poorly understood, limiting the development of targeted interventions during the critical early plastic period. To explore the behavioural and molecular outcomes of perinatal asphyxia that may model important aspects of neuropsychiatric disorders observed in humans, we utili

Corticotropin-releasing factor receptors: physiology, pharmacology, biochemistry and role in central nervous system and immune disorders.

Corticotropin-releasing factor (CRF) plays a major role in coordinating the endocrine, autonomic, behavioral and immune responses to stress through actions in the brain and the periphery. CRF receptors identified in brain, pituitary and spleen have comparable kinetic and pharmacological characteristics, guanine nucleotide sensitivity and adenylate cyclase-stimulating activity. Differences were observed in the molecular mass of the CRF receptor complex between the brain (58,000 Da) and the pituitary and spleen (75,000 Da), which appeared to be due to differential glycosylation of the receptor p

Primary Blast-Induced Traumatic Brain Injury as a Risk Factor for (Cerebro)vascular Disorder: Clinical Manifestations, Blast Physics, Biomechanics, Pathobiology, and Critical Gaps.

Exposure to blast waves without kinetic, penetrating, thermal, or toxic components causes a distinct form of traumatic brain injury, termed primary blast-induced TBI (pbTBI). Clinical manifestations of pbTBI span a wide spectrum, ranging from life-threatening intracranial hemorrhage, hyperemia, and delayed cerebral edema to mild and transient neurological symptoms without detectable structural abnormalities on routine imaging. At the mild end of the spectrum, symptoms after a single exposure may resolve quickly, yet repeated exposures-even at very low levels, termed "subconcussive"-can develop

Can the presence of a human or a conspecific reduce stress-related behaviours in goats?

From an ethological perspective, human presence may serve as a potential source of social support for animals facing challenging situations. This may be particularly relevant for animals involved in animal-assisted services, where interactions with different people are an integral part of their work. This study aimed to assess whether the presence of a human, both familiar and unfamiliar, can mitigate goats' behavioural responses to social isolation to a similar extent as the presence of a conspecific. It was also examined whether goats' responses to human presence during isolation depend on t

The effect of sex on the cardiopulmonary and neuromuscular response to high-intensity interval exercise.

Sex differences exist in the integrative response to exercise; however, these are typically researched during incremental and constant-load exercise. Interval exercise involves high-intensity efforts interspersed with recovery periods to repeatedly stress physiological systems, and it is currently unknown whether the response to this form of exercise differs between sexes. Ten males and 10 females (age: 25 ± 3 yr) completed two experimental visits. First, an incremental treadmill exercise test was performed to obtain submaximal (lactate threshold) and maximal (V̇o2peak) data. Thereafter, visit

Alignment Between Cardiologists and AI-Driven Diagnostic Systems: Mixed Methods Study.

BACKGROUND: The clinical value of artificial intelligence (AI)-based diagnostic systems depends not only on their accuracy but also on how well their outputs integrate with clinicians' judgments in practice. Critical knowledge gaps remain regarding diagnostic concordance between AI and clinicians in stress echocardiography interpretation, patient characteristics predicting discordance, and how cardiologists respond when AI recommendations conflict with their clinical diagnoses. OBJECTIVE: This study examined the diagnostic alignment between an AI-driven stress echocardiography system (EchoGo

CMA-associated cellular state remodeling defines prognostic heterogeneity and identifies ARRDC3 as a protective functional gene in small cell lung cancer.

BACKGROUND: Small cell lung cancer (SCLC) is a highly aggressive malignancy characterized by rapid progression, early metastasis, frequent relapse, and treatment resistance. Although chemoimmunotherapy has improved outcomes in a subset of patients, reliable molecular stratification tools reflecting tumor heterogeneity and survival risk remain limited. Chaperone-mediated autophagy (CMA) is involved in proteostasis, metabolic adaptation, and stress responses, but its cellular heterogeneity and prognostic relevance in SCLC remain unclear. METHODS: This study integrated single-cell RNA sequencing

Mechanisms of Diapause in Major Crustacean Taxa: Environmental Cues, Gene Regulation, and Adaptive Strategies.

Diapause serves as an adaptive dormancy strategy in freshwater, hypersaline, and marine crustaceans, allowing them to survive harsh conditions like temperature extremes, salinity fluctuations, hypoxia, and food scarcity. This review focuses on diapause across major crustacean taxa, with particular emphasis on branchiopods and copepods. However, detailed mechanistic evidence is more prevalent in well-studied species like Artemia, Daphnia, and calanoid copepods, leading to an uneven distribution of evidence across these taxa. Thus, the molecular and physiological mechanisms discussed in this rev

An O-glycosylation-based prognostic signature for biochemical recurrence in prostate cancer identifies GALNTL6 as a potential promoter of malignant phenotypes.

BACKGROUND: Prostate cancer (PCa) is frequently diagnosed in men, and biochemical recurrence (BCR) remains a persistent problem after clinical treatment. Commonly used indicators do not identify patients who are likely to relapse early. Protein O-linked glycosylation is an important post-translational modification involved in cancer progression, but its prognostic relevance and regulatory mechanisms in PCa remain insufficiently understood. GALNTL6, which encodes polypeptide N-acetylgalactosaminyltransferase-like 6, is putatively involved in mucin-type protein O-linked glycosylation and remains

Unraveling the complexity of Helicobacter pylori: Virulence factors, biofilm formation, and antibiotic resistance.

Helicobacter pylori infection remains one of the most common chronic bacterial infections worldwide and represents a major etiological factor in diseases of the upper gastrointestinal tract, including chronic gastritis, peptic ulcer disease, and gastric cancer. Despite continuous refinement of eradication regimens based on antibiotics and proton pump inhibitors, treatment efficacy has progressively declined, primarily due to increasing antimicrobial resistance and the ability of H. pylori to form biofilm structures. Accumulating evidence indicates that biofilm formation, bacterial virulence, a
