Potent immunosuppression by secretory/excretory products of larvae from the sheep blowfly Lucilia cuprina.

Secretory/excretory products (sec/ex) of parasitic larvae of the sheep blowfly Lucilia cuprina potently inhibited proliferation of peripheral blood leucocytes stimulated by mitogens in vitro. Suppression of proliferation was not due to irreversible damage because cells cultured for 24 h in high concentrations of sec/ex appeared viable (assessed by Trypan blue exclusion) and did not show impaired proliferation after washing. Furthermore, suppression induced by sec/ex could be overcome by increasing concentrations of mitogen. The inhibitory activity could be demonstrated in cultures where sec/ex

Detection of cellular immunity with the soluble antigen of the fungus Sporothrix schenckii in the systemic form of the disease.

Sporothrix schenckii is the etiologic agent of sporotrichosis, a mycosis of world-wide distribution more commonly occurring in tropical regions. The immunological mechanisms involved in the prevention and control of sporotrichosis are not fully understood but apparently include both the humoral and cellular responses. In the present investigation, cellular immunity was evaluated by in vivo and in vitro tests in mice infected with yeast-like forms of S. schenckii. The disease developed systemically and cellular immunity was evaluated for a period of 10 weeks. The soluble antigen utilized in the

Cellular immunity in tuberculous pleural effusions: evidence of spontaneous lymphocyte proliferation and antigen-specific accelerated responses to purified protein derivative (PPD).

The kinetics of in vitro cellular proliferation against a PPD of Mycobacterium tuberculosis or streptococcal antigen (streptokinase-streptodornase) was evaluated in pleural fluid and peripheral blood mononuclear cells (PBMC) from patients with tuberculous and non-tuberculous pleuritis. The peak proliferative response to PPD by mononuclear cells from pleural fluid occurred earlier (day 3) in 65% of patients with tuberculosis, a finding not seen in non-tuberculous effusions. Spontaneous lymphocyte proliferation of both peripheral blood lymphocytes and pleural effusion lymphocytes was frequently

The Th1/Th2-like switch in syphilitic infection: is it detrimental?

Organisms that cause chronic diseases have evolved mechanisms to evade those immune defenses that resolve the acute stage of infection (10, 12-14, 21, 22, 32, 35, 37, 38, 40, 42, 45-49, 53). Much is to be learned by specifically identifying the mechanisms underlying these evasive strategies. Important new insights will emerge in terms of immunoregulatory pathways. This in turn will facilitate vaccine development. A good example is leishmania infection. The acute stage of this disease is resolved by DTH-macrophage activation. Leishmanial components preferentially activate Th2 lymphocytes. As a

The effector mechanism and vaccination against cutaneous leishmaniasis.

This paper summarises the recent findings that nitric oxide plays an important role in the elimination of intracellular and extracellular leishmania parasite in vitro and in vivo. Nitric oxide is produced by macrophages which are activated by IFN-gamma secreted from Th1 subsets of T cells. It is now believed that, at least in cutaneous leishmaniasis, Th2 subsets of CD4+ T cells are disease-promoting and they do so probably by producing IL-3 and IL-4 which inhibits the activation of macrophages by IFN-gamma. Thus, an effective anti-leishmania vaccine should aim at preferentially inducing Th1 su

Inhibition of transformation by levodopa-carbidopa in lymphocytes derived from patients with melanoma.

Levodopa-carbidopa has been widely used in the treatment of Parkinson's disease. We are currently investigating the effectiveness of this combination for the treatment of patients with advanced malignant melanoma. As part of this study, we have examined the effects of this combination on the phytohemagglutinin-induced transformation of human lymphocytes, isolated from patients with malignant melanoma prior to undergoing treatment. Levodopa, as well as carbidopa and 3 additional levodopa synthetic analogs, inhibited the transformation of phytohemagglutinin-stimulated DNA synthesis at pharmacolo

Development of cellular immunity in cutaneous leishmaniasis due to Leishmania tropica.

A laboratory worker was inadvertently inoculated with infectious material while passaging amastigotes of Leishmania tropica. The subsequent development of a cutaneous lesion was correlated with results of sequential in vitro bioassays of cell-mediated immunity. Induction of lymphocyte proliferation and interleukin 2 production by soluble L. tropica antigens appeared within five weeks of infection and reached maximal levels coincident with ulceration of the skin lesion. Thereafter interleukin 2 production rapidly decreased, whereas the lymphocyte proliferative response declined more slowly. Bio

[Immunoblastic lymphadenopathy occurring during treatment with carbamazepine. 2 cases].

Dysimmune lymphadenopathies during carbamazepine treatment. A report of two cases. We report on two cases of dysimmune lymphadenopathies with histological aspect of angio immunoblastic lymphadenopathy (AIL) developing after administration of carbamazepine. Clinical manifestations consisted of fever, erythroderma, generalized pruritus, facial edema, lymphadenopathy, liver enlargement. The two patients had anemia, hypogammaglobulinemia, impaired liver function and a negative Coomb's test. Lymphocyte stimulation test with carbamazepine in vitro was positive in both cases. Lymph node biopsy disclo

Cellular immune response to Marek's disease: listeriosis as a model of study.

The immune response of chickens to Listeria monocytogenes was studied as a potential model for cell-mediated immunocompetence. Chickens genetically resistant and susceptible to Marek's disease (MD) did not differ in their ability to survive Listeria, although during the early stages of infection the bacteria replicated more readily in MD-susceptible chickens. MD-susceptible chickens responded earlier than MD-resistant chickens, and with equal or increased intensity, in assays of various components of the cell-mediated reaction. These assays included T-cell activation, delayed-type hypersensiti

Modulation of the host immune response as a result of Chlamydia psittaci infection.

After intraperitoneal injection of mice with infectious, inactivated, or envelope preparations of the elementary body of Chlamydia psittaci, lymphocyte transformation of spleen cells to the mitogens concanavalin A, phytohemagglutinin, and lipopolysaccharide was significantly reduced 1 and 2 weeks postinjection. Lymphocyte response returned to the control values by 4 weeks. Similarly, transformation of cells by chlamydial antigen was not detected until 4 weeks postinjection. Injection of the noninfectious intracellular reticulate body, in contrast, had little effect on transformation of cells t

[Drug fever caused by the antidepressive agent nomifensine (Alival)].

During 1979 4 patients were observed who developed short episodes of fever as high as 104 degrees F after oral intake of 25--100 mg nomifensine (Alival). In all four cases a clear fever spike was produced by reexposure to the drug. The reaction time was 4--6 hours. this drug-induced fever appeared initially 2--4 weeks after the commencement of Alival therapy. No other cause for the fever was identifiable. In one patient an allergic alveolitis was suspected to be a further reaction. In another patient a concomitant granulomatous hepatitis was possibly also due to this drug. It is probable that

Role of macrophages in defective cell mediated immunity in lepromatous leprosy. I. Factor(s) from macrophages affecting protein synthesis and lymphocyte transformation.

Macrophages from lepromatous leprosy patients specifically show reduced protein synthesis in the presence of M. leprae. They also produce, as a result of interaction with M. leprae, factor(s) that reduce protein synthesis in normal macrophages as well as block lymphocyte transformation in normal leukocyte cultures in the presence of M. leprae as the antigen. These observations implicate a defective macrophage system in lepromatous leprosy patients.

An analysis of cross reactions among mycobacteria by in vivo and in vitro assays of cellular hypersensitivity.

Homologous and heterologous skin test reactions in sensitized guinea pigs to antigens from 11 species of mycobacteria were fitted by a linear model that permitted the ranking of each species on the basis of cross reactivity. When the same species were ranked on the basis of in vitro transformation of sensitive lymphocytes in response to homologous and heterologous antigen preparations, a different ranking order was observed. Reasons for the lack of strict correlation between skin tests and in vitro lymphocyte transformations are discussed.

A comparison of cytogenetics and histopathology in the malignant lymphomata.

The non-Hodgkin's lymphomata are characteristically near diploid with occasional examples having higher chromosome numbers. In contrast, about half the cases of Hodgkin's disease have chromosome numbers in the triploid range. Karyotypes in cases of Hodgkin's disease appear to be more deranged than those found in other lymphomata, even when the cells are in the diploid range. If karyotypically normal cells in the tumours represent some degree of host reaction, then this is most marked in Hodgkin's disease. Long arm deletions of chromosome 18 were identified in cells from 7/31 cases of non-Hodgk
