Metal-ion stoichiometry of the HIV-1 RT ribonuclease H domain: evidence for two mutually exclusive sites leads to new mechanistic insights on metal-mediated hydrolysis in nucleic acid biochemistry.

Crystallographic studies of the Mn(2+)-doped RNase H domain of human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT) have revealed two bound Mn2+ separated by approximately 4A and surrounded by a cluster of four conserved carboxylates. Escherichia coli RNase H is structurally similar to the RNase H domain of HIV-1 RT, but requires one divalent metal cation for its activity, implying either that the HIV-1 RT RNase H domain contrasts in its ability to bind two divalent metal ions, or that the crystallographic data reflect specific use of Mn2+ and/ or the doping technique employed.

Metal ion-tetracycline interactions in biological fluids. Part 3. Formation of mixed-metal ternary complexes of tetracycline, oxytetracycline, doxycycline and minocycline with calcium and magnesium, and their involvement in the bioavailability of these antibiotics in blood plasma.

The equilibria of the complexes formed separately by calcium and magnesium with tetracycline, oxytetracycline, doxycycline and minocycline were studied in the previous parts of this series. Computer simulations run on this occasion showed that the stoichiometry of these species do actually condition the bioavailability of the antibiotics in blood plasma during treatment, the fraction of free base being quite negligible with regard to the metal-bound one. The present paper completes this study with the investigation of the mixed-metal complexes given rise to by calcium and magnesium with the sa

Source-oriented health risk assessment of heavy metals in soil and dust at coal resource-based urban parks in northern China based on bioaccessibilities.

The mining and utilisation of coal release numerous heavy metals (HMs) into the urban environment, but there are few studies expanding on source-oriented health risk assessment based on bioaccessibility for the identification of health priority control pollutants at coal resource-based cities. Hence, dust and soil samples were collected from urban parks in Datong, Shanxi Province, known as the coal capital of China, to evaluate the pollution level, bioaccessibility, and source-specific health risk assessment. The results revealed severe contamination of As, Cd, and Pb in both soil and dust, wi…

N-benzyl-N-methyldithiocarbamate (BMDC) combines with metals to produce antimicrobial and anti-biofilm activity against methicillin-resistant Staphylococcus aureus (MRSA) and Staphylococcus epidermidis.

Methicillin-resistant Staphylococcus aureus (MRSA) is a high-priority microorganism that necessitates the development of new treatments, as it causes a substantial disease burden and economic impact globally. MRSA colonizes the skin and anterior nares and can potentially become invasive, leading to pneumonia and soft tissue infection. Additionally, MRSA can establish chronic infections in wounds and medical implants, partly due to its ability to form biofilms. Likewise, the skin commensal Staphylococcus epidermidis also causes similar infections, particularly through its ability to form a plas

Unveiling the association between dietary metals and prostate health: Identifying benign prostatic hyperplasia using a SHAP-based interpretable machine learning model in US adults.

BACKGROUND: Benign prostatic hyperplasia affects roughly 50 % of men aged 51-60 and 70 % of those aged 61-70. While heavy metals have been linked to prostate cancer and inflammation, little is known about how the dietary intake of trace metals influences BPH risk. This study aimed to establish an interpretable, machine-learning-based framework using U.S. NHANES data to identify key dietary metals associated with BPH risk. METHODS: We analyzed adult male participants from NHANES cycles 2005-2008, extracting daily intake estimates for six trace metals. This study compared six algorithms to eluc…

Maternal pre-pregnancy overweight/obesity, in-utero exposure to toxic heavy metals, and offspring age at peak height velocity: A prospective birth cohort study.

INTRODUCTION: Research on in-utero heavy metal exposure shows inconsistent results on offspring pubertal timing. We aim to contribute to understanding the association between heavy metal exposures and age at peak height velocity (APHV), an indicator of pubertal timing, and to investigate the joint associations of heavy metal concentrations and previously known risk factors. METHODS: We included 956 mother-child dyads from the Boston Birth Cohort. Heavy metal concentrations were measured in maternal red blood cells collected 24-72 h after delivery. Offspring APHV was calculated using mixed-eff

Modulators of mitochondrial ATP-sensitive potassium channel affect cytotoxicity of heavy metals: Action on isolated rat liver mitochondria and AS-30D ascites hepatoma cells.

Heavy metals are ubiquitous environmental pollutants that are extremely dangerous for public health, but the molecular mechanisms of their cytotoxic action are still not fully understood. In the present work, the possible contribution of the mitochondrial ATP-sensitive potassium channel (mK(ATP)), which is usually considered protective for the cell, to hepatotoxicity caused by heavy metals was investigated using polarography and swelling techniques as well as flow cytometry. Using isolated liver mitochondria from adult male Wistar rats and various potassium media containing or not containing p…

Rapid and sensitive determination of Se and heavy metals in foods using electrothermal vaporization inductively coupled plasma mass spectrometry with a novel transportation system.

Rapid, sensitive and simultaneous determination of trace multi-elements in various plant food samples such as grain, oilseed, vegetable and tea is always a challenge thus far. In this work, a rapid determination method for Se, Cd, As and Pb in food samples by inductively coupled plasma mass spectrometry (ICP-MS) using slurry sampling electrothermal vaporization (SS-ETV) was developed. To improve the analytical sensitivity and precision as well as eliminate the memory effect, a gas turbulator line and signal delay device (SDD) were for the first time designed for the graphite furnace (GF) ETV c

Comparison of spatiotemporal burial and contamination of heavy metals in core sediments of two plateau lakes with contrasting environments: implication for anthropogenic-driven processes.

Investigating the impacts of climatic factors and human activities on sedimentary records of heavy metal (HM) contamination in lakes is essential for decision-making in global environmental monitoring and assessment. Spatiotemporal distributions of grain size (GS) and HM (Al, Cr, Mn, Ni, Cu, Zn, As, and Pb) concentrations have been conducted in core sediments that are collected from two adjacent plateau fault-bound lakes in southwest China with contrasting environments, i.e., deep oligotrophic Lake Fuxian (FX) and shallow hypertrophic Lake Xingyun (XY). Results showed that the average value of

Changes in physicochemical, heavy metals and air quality linked to spot Aplocheilus panchax along Mahanadi industrial belt of India under COVID-19-induced lockdowns.

Positive effects of COVID-19-induced lockdowns on environment are well documented although pre-planned experiments for such analyses and appearance of fish species are lacking. We hypothesize that spotting the fish Aplocheilus panchax along the industrial belt of Mahanadi River near Cuttack in a never seen manner could be due to the regenerated environment. Heavy metals, water and air qualities along with spotting A. panchax in up, mid and downstream of Mahanadi River near Jagatpur industrial basins were analysed during pre-(end of March 2020) and after 60 days of lockdowns (last week of May 2…
