Effects of 4-aminopyridine on muscle and motor unit force in canine motor neuron disease.

Hereditary Canine Spinal Muscular Atrophy (HCSMA) is an autosomal dominant disorder of motor neurons that shares features with human motor neuron disease. In animals exhibiting the accelerated phenotype (homozygotes), we demonstrated previously that many motor units exhibit functional deficits that likely reflect underlying deficits in neurotrans-mission. The drug 4-aminopyridine (4AP) blocks voltage-dependent potassium conductances and is capable of increasing neurotransmission by overcoming axonal conduction block or by increasing transmitter release. In this study, we determined whether and

Confirmation of clinical diagnosis in requests for prenatal prediction of SMA type I.

The recent discovery of a major SMA-locus in the chromosomal region 5q makes it possible to carry out prenatal DNA studies in families in which a child with SMA type I has been born. Since direct mutation analysis is not yet possible, the reliability of prenatal prediction of SMA type I usually depends on the certainty of the clinical diagnosis in the index patient. Sixteen requests were received for DNA studies in couples who had had a previous child with SMA type I. After re-evaluation, the performance of prenatal diagnosis was rejected in four cases. Among the other twelve families prenatal

Localization of the mutation in an extended family with Charcot-Marie-Tooth neuropathy (HMSN I).

Hereditary motor and sensory neuropathy type I (HMSN I) or Charcot-Marie-Tooth (CMT) disease is an autosomal dominant peripheral neuropathy. In some CMT families linkage has been reported with either the Duffy blood group or the APOA2 gene, both located on chromosome 1q. More recently, linkage has been found in six CMT families with two chromosome 17p markers. We extensively analyzed a multi-generation Charcot-Marie-Tooth family by using molecular genetic techniques in order to localize the CMT gene defect. First, we constructed a continuous linkage group of 11 chromosome 1 markers and definit

[Isolated cervicothoracic spinal amyotrophy caused by hydromyelia. Stabilization by a ventricular shunt].

A 25-year-old man developed marked weakness of the upper limbs and shoulder girdle over a period of 6 months. On examination there was amyotrophy, fasciculations and areflexia of the upper limbs with increased lower limb reflexes. Sensation and CSF were normal. CT scan showed a thickened cord with a central medullary cavity of triangular shape with a ventral base. Myelobulbography showed a type I Arnold-Chiari malformation. Spinal angiography was normal. Isotopic studies showed early ventricular reflux. Although infusion tests gave negative results a ventriculoperitoneal shunt was performed an

Targeted knockdown of Smn in muscle stem cells induces non-cell autonomous loss of motor neurons.

Spinal muscular atrophy (SMA) is due to a deficit in SMN, a ubiquitously expressed protein encoded by the Survival of Motor Neuron 1 (SMN1) gene. Recently, SMN-targeted disease-modifying treatments have greatly improved the clinical outcomes of this neuromuscular disease. However, uncertainties remain regarding their long-term efficacy and non-neuronal tissue involvement in disease progression. Skeletal muscle tissue and the muscle stem cells (MuSC) that sustain its postnatal growth and regenerative capacity are affected by SMN deficit. While a direct contribution of muscle tissue in the disea

Identification of key genes associated with muscle atrophy after spinal cord injury and experimental verification in rats.

BACKGROUND: Spinal cord injury (SCI) is a neurological disease with high morbidity and mortality. Post-SCI muscle atrophy is a cascade response to SCI, and failure to actively prevent its occurrence severely affects patients' mobility and quality of life. Therefore, deeply exploring the correlation between muscle atrophy after SCI and the molecular regulation mechanism is of great significance. METHODS: Download GSE21497 expression profile data from the gene expression omnibus (GEO) database. Perform weighted gene co-expression network analysis (WGCNA) on the obtained differentially expressed

Neuromuscular Electrical Stimulation Combined With Rehabilitation Training Improves Rehabilitation After Spinal Cord Injury.

AIM: Spinal cord injury (SCI), particularly traumatic spinal cord injury (TSCI), is a globally prevalent neurological condition and often causes severe functional and physical disabilities. Neuromuscular electrical stimulation (NMES), when combined with rehabilitation training, has been reported to enhance functional recovery in patients with SCI. However, its specific clinical advantages and safety profile require further validation through robust empirical data. Therefore, this study aims to investigate the synergistic effects of NMES combined with rehabilitation training on improving motor

Ighmbp2 mutations and disease pathology: Defining differences that differentiate SMARD1 and CMT2S.

Mutations in the Immunoglobulin mu DNA binding protein 2 (IGHMBP2) gene result in two distinct diseases, SMA with Respiratory Distress Type I (SMARD1) and Charcot Marie Tooth Type 2S (CMT2S). To understand the phenotypic and molecular differences between SMARD1 and CMT2S, and the role of IGHMBP2 in disease development, we generated mouse models based on six IGHMBP2 patient mutations. Previously, we reported the development and characterization of Ighmbp2D564N/D564N mice and in this manuscript, we examine two mutations: D565N (D564N in mice) and H924Y (H922Y in mice) in the Ighmbp2H922Y/H922Y a

Phenotypes of SMA patients retaining SMN1 with intragenic mutation.

BACKGROUND: Spinal muscular atrophy (SMA) is an autosomal recessive neuromuscular disorder caused by homozygous deletion or intragenic mutation of the SMN1 gene. It is well-known that high copy number of its homologous gene, SMN2, modifies the phenotype of SMN1-deleted patients. However, in the patients with intragenic SMN1 mutation, the relationship between phenotype and SMN2 copy number remains unclear. METHODS: We have analyzed a total of 515 Japanese patients with SMA-like symptoms (delayed developmental milestones, respiratory failures, muscle weakness etc.) from 1996 to 2019. SMN1 and S

Testosterone and Resistance Training Improve Muscle Quality in Spinal Cord Injury.

PURPOSE: Spinal cord injury (SCI) negatively impacts muscle quality and testosterone levels. Resistance training (RT) has been shown to increase muscle cross-sectional area (CSA) after SCI, whereas testosterone replacement therapy (TRT) has been shown to improve muscle quality in other populations. The purpose of this pilot study was to examine if the combined effects of these interventions, TRT + RT, may maximize the beneficial effects on muscle quality after SCI. METHODS: Twenty-two SCI subjects randomized into either a TRT + RT (n = 11) or TRT (n = 11) intervention for 16 wk. Muscle qualit

Clinical and genetic features of Charcot-Marie-Tooth disease 2F and hereditary motor neuropathy 2B in Japan.

Mutations in small heat shock protein beta-1 (HspB1) have been linked to Charcot-Marie-Tooth (CMT) disease type 2F and distal hereditary motor neuropathy type 2B. Only four cases with HSPB1 mutations have been reported to date in Japan. In this study between April 2007 and October 2014, we conducted gene panel sequencing in a case series of 1,030 patients with inherited peripheral neuropathies (IPNs) using DNA microarray, targeted resequencing, and whole-exome sequencing. We identified HSPB1 variants in 1.3% (13 of 1,030) of the patients with IPNs, who exhibited a male predominance. Based on n

Interaction of small heat shock proteins with light component of neurofilaments (NFL).

The interaction of human small heat shock protein HspB1, its point mutants associated with distal hereditary motor neuropathy, and three other small heat shock proteins (HspB5, HspB6, HspB8) with the light component of neurofilaments (NFL) was analyzed by differential centrifugation, analytical ultracentrifugation, and fluorescent spectroscopy. The wild-type HspB1 decreased the quantity of NFL in pellets obtained after low- and high-speed centrifugation and increased the quantity of NFL remaining in the supernatant after high-speed centrifugation. Part of HspB1 was detected in the pellet of NF

SMA screening system using dried blood spots on filter paper: application of COP-PCR to the SMN1 deletion test.

BACKGROUND: Spinal muscular atrophy (SMA) is a common neuromuscular disorder caused by mutations in SMN1. More than 95% of SMA patients carry homozygous SMN1 deletions. Thus, the SMN1 deletion test should be performed initially as part of the diagnostic process. However, SMN2, a highly homologous gene, hampers detection of SMN1 deletion. To differentiate between SMN1 and SMN2, many analysis methods have been developed yet they are not all available worldwide. AIM: To establish a simple but accurate SMN1-deletion detection system that can be used worldwide. METHODS: Fifty DNA samples (29 SMA

Minimum invasive posterior decompression for cervical spondylotic amyotrophy.

BACKGROUND: Cervical spondylotic amyotrophy (CSA), characterized by amyotrophy and muscular weakness of the upper limbs, is caused by damage to anterior spinal root or anterior horn of the spinal cord. Formerly, anterior decompression and fusion were performed for treatment of CSA, but it has recently been reported that posterior decompression is also effective. However, a consensus on the choice of procedure has not yet been reached. Selective laminoplasty as minimally invasive surgery is a posterior decompression procedure that alleviates axial neck pain. Because, for CSA patients, the respo
