Repurposing of FDA-Approved Drugs to Disrupt Iron Uptake in Mycobacterium abscessus: Targeting Salicylate Synthase as a Novel Approach.

Non-tuberculous mycobacteria (NTM) are opportunistic pathogens that lead to severe, persistent infections, particularly in immunocompromised … roved drugs. In this work, we applied this approach to identify compounds inhibiting iron uptake in Mycobacterium abscessus (Mab). Specifically, we studied the targeting of salicylate synthase, an enzyme that plays a crucial role in the biosynthesis of mycobacterial siderophores necessary for iron acquisition. Performing an in silico virtual screening of three …

The regulatory role of the Notch signaling pathway in mitophagy and macrophage-mediated elimination of Mycobacterium bovis bacillus Calmette-Guérin.

BACKGROUND: The Notch signaling pathway plays a critical role in maintaining immune responses. This study examined the regulatory effect of the Notch signaling pathway on mitophagy and macrophage function in response to Mycobacterium bovis Bacillus Calmette-Guérin (BCG) infection. METHODS: RAW264.7 cells with stable Notch-1 knockdown were divided into three groups: control, siRNA Notch-1, and siRNA Notch-1 + valinomycin. Following BCG infection, we measured reactive oxygen species (ROS) levels, the expression of Notch signaling and mitophagy-related genes, cytokines, and BCG colony count. RE…

Establishment and application of a TaqMan-based quantitative PCR assay for simultaneous detection of bovine Brucella spp. and Mycobacterium spp.

Brucellosis and tuberculosis are two zoonotic, chronic infectious diseases caused by bacteria of the genus Brucella and Mycobacterium, respectively, which pose significant hazards to both animal husbandry and human health. Currently, mixed infections of these two pathogens are prevalent in livestock production; thus, establishing a molecular diagnostic method for the simultaneous detection and analysis of brucellosis and tuberculosis is crucial for the prevention and control of these diseases. By utilizing conserved regions within the genomes of Brucella and Mycobacterium, we designed specific…

A Novel Thiopeptide Exhibits In Vitro and In Vivo Synergistic Antibacterial Activity with Macrolide Against Mycobacterium avium Complex Infection.

The limitations of the current standard therapy for pulmonary diseases caused by Mycobacterium avium complex (MAC) underscore the urgent need for novel therapeutic agents. Among potential candidates, thiopeptide antibiotics have attracted attention due to their antibacterial activity but their poor solubility has limited clinical application. In this study, we developed AJ-099, a thiopeptide derivative with improved solubility and enhanced antibacterial potency. The in vitro antibacterial activity of AJ-099 was confirmed across macrolide-susceptible and- resistant clinical isolates, with minim…

Differential requirement of formyl peptide receptor 1 in macrophages and neutrophils in the host defense against Mycobacterium tuberculosis Infection.

Formyl peptide receptors (FPR), part of the G-protein coupled receptor superfamily, are pivotal in directing phagocyte migration towards chemotactic signals from bacteria and host tissues. Although their roles in acute bacterial infections are well-documented, their involvement in immunity against tuberculosis (TB) remains unexplored. Here, we investigate the functions of Fpr1 and Fpr2 in defense against Mycobacterium tuberculosis (Mtb), the causative agent of TB. Elevated levels of Fpr1 and Fpr2 were found in the lungs of mice, rabbits and peripheral blood of humans infected with Mtb, suggest…

Interleukin-26 expression in tuberculosis disease and its regulatory effect in macrophage polarization and intracellular elimination of Mycobacterium tuberculosis.

Tuberculosis(TB), an infectious disease caused by Mycobacterium tuberculosis (Mtb) infections, remains the leading cause of mortality from a single infectious agent globally. The progression of tuberculosis disease is contingent upon the complex interplay between the host's immune system and the pathogen Mtb. Interleukin-26 (IL-26), the most recently identified cytokine belonging to the IL-10 family, exhibits both extracellular antimicrobial properties and pro-inflammatory functions. However, the precise role of IL-26 in the host immune defense against Mtb infections and intracellular killing …

Correlation between bacterial culture and IS900 qPCR to detect Mycobacterium avium subsp. paratuberculosis in the feces of sika deer.

Mycobacterium avium subsp. paratuberculosis (MAP) infection in deer causes paratuberculosis (PTb; Johne's disease), a slowly progressive chronic granulomatous enteritis. Accurate and rapid detection of MAP shedding by subclinical animals is essential for effective control of infection in deer herds. The VetMAX MAP 2.0 qPCR assay targeting IS900 was developed for cattle, sheep, and goats. However, the performance of the assay in sika deer (Cervus nippon) has not been evaluated. Here, we describe the use of the VetMAX MAP 2.0 qPCR assay on fecal samples from sika deer for the detection of MAP an…

Application of luminescence-producing Mycobacterium tuberculosis strains for evaluating anti-tuberculosis drug and vaccine efficacy in vitro and in vivo.

BACKGROUND: Tuberculosis (TB), caused by Mycobacterium tuberculosis (M.tb), remains a major public health problem worldwide. The development of effective drugs and vaccines remains crucial but the prolonged culture period of M.tb hampers this process. This limitation has been addressed by developing a bioluminescent M.tb strain that can be used to measure bacterial burden in live animals non-invasively and in real time. We investigated whether bioluminescence real-time imaging technologies could be used to monitor the infection process in live mice, with the aim of facilitating TB research. ME…

A minimally instrumented method for the detection of rifampicin resistance-causing mutations in Mycobacterium tuberculosis utilizing lateral flow readout.

BACKGROUND: Genotypic methods for detecting antimicrobial resistance in Mycobacterium tuberculosis (M.tb) typically require expensive instrumentation, limiting their accessibility in peripheral and resource-limited settings. Rifampicin resistance is a key marker of multidrug-resistant tuberculosis (MDR-TB) and is primarily caused by mutations in the rpoB gene. Rapid, cost-effective, and minimally instrumented methods for detecting these mutations are essential for improving tuberculosis diagnostics, particularly in low-resource environments. This study addresses the need for a simple, sensitiv…
