Identification of Novel Biomarkers for Pre-diabetic Diagnosis Using a Combinational Approach.

Reliable protein markers for pre-diabetes in humans are not clinically available. In order to identify novel and reliable protein markers for pre-diabetes in humans, healthy volunteers and patients diagnosed with pre-diabetes and stroke were recruited for blood collection. Blood samples were collected from healthy and pre-diabetic subjects 12 h after fasting. BMI was calculated from body weight and height. Fasting blood glucose (FBG), glycated hemoglobin (HbA1C), triglyceride (TG), total cholesterol, high-density lipoprotein, low-density lipoprotein (LDL), insulin and albumin were assayed by a

Human Epididymis Protein 4 Quantification and Interaction Network Analysis in Seminal Plasma.

BACKGROUND: A well-known tissue marker of ovarian cancer, Human Epididymis protein 4 (HE4) is the member of whey acidic four-disulfide core proteins family. Purified from human seminal plasma and characterized as a cross-class protease inhibitor, HE4 was proposed to shield spermatozoa against proteolytic factors. However, its exact biological function is unknown. Proteins usually function in conjunction with other proteins in the system and thus, identification and analysis of protein networks become essential to decode protein functions. OBJECTIVE: This study was performed to explore possibl

iTRAQ-based quantitative analysis of hippocampal postsynaptic density-associated proteins in a rat chronic mild stress model of depression.

Major depressive disorder (MDD) is a prevalent psychiatric mood illness and a major cause of disability and suicide worldwide. However, the underlying pathophysiology of MDD remains poorly understood due to its heterogenic nature. Extensive pre-clinical research suggests that many molecular alterations associated with MDD preferentially localize to the postsynaptic density (PSD). Here, we used a rodent chronic mild stress (CMS) model to generate susceptible and unsusceptible subpopulations. Proteomic analysis using an isobaric tag for relative and absolute quantitation (iTRAQ) and tandem mass

A salivary histamine-binding protein from Rhipicephalus microplus and its evaluation as an anti-tick vaccine antigen.

The cattle tick Rhipicephalus microplus is an ectoparasite of major economic importance to livestock, particularly in tropical and subtropical regions. The lack of effective vaccines against tick infestation has hindered its control and the identification of new molecular targets for recombinant vaccine development arises as an alternative strategy. Tick saliva is known as a reservoir of potential vaccine targets, including several proteins involved in attachment, immune response modulation and feeding that targeting might affect tick infestations. Among these proteins, histamine binding prote

Downregulation of Nesprin1 by Runx2 deficiency is critical for the development of skeletal laminopathy-like pathology.

Runx2 is a master regulator of bone formation, and its dysfunction causes cleidocranial dysplasia (CCD) in humans. When iPS cells were generated from patients with CCD and Runx2-deficient iPS cells were generated using gene-editing techniques, abnormal laminopathy-like nuclei were observed. Runx2-deficient cells showed reduced Lamin A/C expression, but not protein levels. However, in Runx2-deficient cells, both the gene expression and protein levels of Nesprin1 were reduced, perinuclear actin fibers were sparser, and nuclear stiffness was reduced. Forced expression of Lamin A/C increased nucle

Inherited dystonias: clinical features and molecular pathways.

Recent decades have witnessed dramatic increases in understanding of the genetics of dystonia - a movement disorder characterized by involuntary twisting and abnormal posture. Hampered by a lack of overt neuropathology, researchers are investigating isolated monogenic causes to pinpoint common molecular mechanisms in this heterogeneous disease. Evidence from imaging, cellular, and murine work implicates deficiencies in dopamine neurotransmission, transcriptional dysregulation, and selective vulnerability of distinct neuronal populations to disease mutations. Studies of genetic forms of dystoni

Spectral studies of the Ca2+-dependent interaction of trifluoperazine with S100b.

S100b is a calcium-binding protein that will bind to many calmodulin target molecules in a Ca2+-dependent manner. In order to study the Ca2+-dependent binding properties of S100b, its interaction with a calmodulin antagonist, trifluoperazine (TFP), was investigated using [19F]- and [1H]-NMR and UV-difference spectroscopy. It was estimated from [19F]-NMR that in the absence of Ca2+, the k1/2 value of TFP was 130 microM, while its k1/2 value decreased to 28 microM in the presence of Ca2+. The addition of KCl was not antagonistic to the Ca2+-dependent interaction of TFP to S100b. The chemical exc

Excitatory neuron-specific core binding factor β (Cbfβ) deletion causes sex-independent spatial memory deficits and female-specific anxiety and depression-like behaviors in mice.

Core-binding factor subunit β (Cbfβ) serves as a transcriptional cofactor for the Runx transcription factors, with well-established roles in hematopoiesis and skeletal development. However, its function within the central nervous system (CNS) remains largely unexplored. To investigate the role of Cbfβ in the brain, we generated neuron-specific Cbfβ knockout mice (Cbfbbr/br) using the Emx1-Cre system. Behavioral assessments included the Y-maze test for short-term spatial memory, the open field test, and the elevated plus maze (EPM) for anxiety-like behaviors. Depression-like behaviors were eval

From the sequence to the conformation of the unabridged transmembrane domains TM1 and TM2 of the cASIC1a ion channel - a parallel tempering approach.

This work was devised to unravel, along replica-exchange molecular-dynamics (REMD) simulations, the conformation in solution of the TM1 and TM2 transmembrane domains of the homotrimeric cASIC1a ion channel. This includes the head of TM1 and tail of TM2 that had previously defied X-ray diffraction analysis in the crystal. The structure of the open-channel complex of cASIC1a with psalmotoxin 1 (PcTx1) was chosen here as a basis, although, to make the simulations affordable, the procedure was limited to the missing portions, including a few adjacent α-helical turns. The latter were held fixed dur

Mutations associated with hemophilia A in the 558-565 loop of the factor VIIIa A2 subunit alter the catalytic activity of the factor Xase complex.

The 558-565 loop region in the A2 subunit of factor (F) VIIIa forms a direct interface with FIXa. We have expressed and purified B-domainless FVIII (FVIII(WT)) and B-domainless FVIII containing the hemophilia A-associated mutations Ser558Phe, Val559Ala, Asp560Ala, Gln565Arg, and the activated protein C cleavage site mutant Arg562Ala. Titration of FVIIIa in FXa generation assays showed that the mutant and wild-type proteins had similar functional affinities for FIXa (dissociation constant [K(d)] values approximately 5 nM-20 nM and approximately 100 nM-250 nM in the presence and absence of phosp

Structural and mechanistic insights into IL2RG-associated immunodeficiency through computational approaches.

Interleukin-2 receptor γ chain (IL2RG, CD132) act as a signaling subunits for various cytokine receptors, required for lymphocyte growth and maturation. Genetic variants within the IL2RG gene are associated with X-linked Severe Combined Immunodeficiency (X-SCID) disease. This study uses an in-silico method to investigate the structural and functional impacts of two IL2RG variants, Trp240Arg (W240R) and Arg226Cys (R226C), located in the extracellular domain, a region important for receptor stability. Homology modeling was generated for Wild-Type (WT) and Variant IL2RG structures and subsequentl

NADPH gating and F-actin engagement define the structural bases of MICAL1 autoinhibition and activation.

Human MICAL1 is a multidomain protein exhibiting both an oxidase activity, producing H2O2 as a second messenger, and an actin depolymerizing activity through the NADPH-dependent reaction catalyzed by its N-terminal flavoprotein domain. In full-length MICAL1, this domain is inhibited by the following protein regions. Interaction with activating (macro)molecules controls the timing and location of the enzyme activity, explaining its participation in diverse cellular processes. To gain insight into the reaction mechanism and its control, we combined kinetic, cryo-EM, chemical cross-linking and mo

Application of mass spectrometry techniques for analysis of higher order structure of proteins.

Determining the higher order structure (HOS) of proteins and protein complexes is central to understanding their functions, dynamics, and interactions. Traditional structural biology approaches, such as X-ray crystallography and nuclear magnetic resonance, provide high-resolution snapshots but often require large amounts of homogeneous samples and may miss dynamic or heterogeneous states. Mass spectrometry (MS) has become an indispensable tool for sensitive and rapid analysis of intact proteins and assemblies under native or near-native conditions. This review discusses the major MS-based stra

Context-dependent variability of HIF heterodimers influences interactions with macromolecular and small molecule partners.

Hypoxia inducible factors (HIFs) are heterodimeric transcription factors that coordinate cellular responses to low oxygen levels by binding a short hypoxia response element (HRE) DNA sequence near O2-regulated genes. Prior studies suggest HIF/HRE complexes are augmented by the binding of additional factors nearby, but those interactions are not well understood. Here, we integrated structural and biochemical approaches to investigate several functionally relevant HIF assemblies with other protein, small molecule, and DNA partners. First, we used cryo-electron microscopy (cryo-EM) to establish t

Interaction analysis of tight junction proteins by Förster resonance energy transfer is recommended in live cells rather than after paraformaldehyde fixation.

The tight junction regulates the paracellular passage of solutes and water. It is primarily constituted by the family of claudins which assemble to huge multimers and basically seal the paracellular cleft. Certain claudins, however, can function as subunits of selective paracellular channels and thereby mediate paracellular ion and water flux across epithelia. In the thick ascending limb of Henle's loop (TAL), ion handling depends on the spatial separation of paracellular sodium and magnesium transport. Magnesium is conducted via a claudin-16/19 complex, whereas sodium permeation is enabled by

Proteomic analysis indicates lower abundance of platelet α-granule proteins in Glanzmann thrombasthenia.

BACKGROUND: Glanzmann thrombasthenia (GT) is an inherited platelet function disorder caused by mutations in the fibrinogen receptor αIIbβ3. The deficiency can be quantitative (type I/II) or qualitative (type III). It causes lack of platelet aggregation and leads to a moderate to severe bleeding tendency. Besides the absence or functional alteration of the integrins αIIb and β3, little is known about the proteomic landscape of platelets from GT patients. OBJECTIVES: To evaluate the platelet proteome in GT. METHODS: Label-free quantification of platelet proteins was performed in 13 genetically

SPSB proteins regulate adipocyte differentiation by targeting FOG-2 for proteasomal degradation.

Friend of GATA (FOG)-2 is a transcriptional cofactor that cooperates with GATA family transcription factors to regulate the development of multiple organs and tissues. During adipocyte differentiation, FOG-2 protein is rapidly downregulated, and this reduction is thought to be important for efficient differentiation, but the underlying mechanism has not been elucidated. Here, we show that the ubiquitin-proteasome system degrades FOG-2 during adipogenesis and that SPRY domain- and SOCS box-containing (SPSB) proteins, the substrate-recognition subunits of Cullin-RING ligase 5 (CRL5), mediate thi

Notch signaling modulates Fgf23 expression through crosstalk with hypoxia and PTH pathways in osteogenic cells.

Fibroblast growth factor 23 (Fgf23) is produced by bone and functions primarily as a phosphaturia hormone. We previously reported that overexpression of the Notch intracellular domain (NICD) in osteogenic cells enhances Fgf23 expression in association with osteomalacia in vivo. Here, we investigated the underlying mechanisms using osteogenic cell lines UMR-106 and IDG-SW3 cells. NICD overexpression increased Fgf23 levels in both cell types. Manipulating RBPJ-κ activity, either a dominant-negative or constitutively active form, revealed that Notch-mediated Fgf23 expression is dependent on RBPJ-
