Desensitization of lung beta-adrenoreceptors induced by 4 day in vivo isoprenaline administration to rats has been investigated both from a functional a biochemical viewpoint. Chronic isoprenaline treatment significantly reduced the relaxing activity of the beta-agonist when tested ex vivo in lung parenchymal strips and also impaired the adenylate-cyclase system. The effect of ethanol on intestinal hydrolysis of pyridoxal 5'-phosphate (PLP), the first step in PLP absorption, was evaluated in the rat. Models included PLP decay during in vitro incubation and PLP luminal disappearance during in vivo perfusion of jejunal segments. Effects of intracerebroventricular injection of bombesin on gastric mucosal glycoproteins and acid secretion were studied to clarify its action mechanism for experimental ulcers in rats. Bombesin (10 micrograms) prevented stress ulcers and indomethacin ulcers. Aspirin (300 mg/kg p.o.) reduced the high molecular weight glycoproteins extracted from the gastric mucosa, and increased the degraded glycoproteins extracted from gastric juice in pylorus-ligated rats. A subchronic 90-d study was conducted with chlorinated and nonchlorinated humic acids using male Sprague-Dawley rats. Rats were injected in one lateral cerebral ventricle (i.c.v.) with apamin (100 ng per animal). The resulting desynchronisation pattern in the electrocorticogram (ECoG) and the symptoms of poisoning were monitored before and after transsection at different levels, and following morphine. Apamin acts primarily on the brain stem and spinal cord, i.e. structures possessing a sensory input, and then indirectly on the higher integrating systems. A new and rapid method for the production of liver cirrhosis in the rat is described. Rats were treated with carbon tetrachloride for 4 weeks and then injected i.p. with phospholipase D daily for 6 days. Controls received either carbon tetrachloride or phospholipase D only. Following the combined treatment cirrhosis developed rapidly; the liver developed micronodules and severe cirrhosis was accompanied by ascites and splenomegaly.
