Short-term anoxia improves postharvest quality and reduces browning-related enzyme activity in edible Astraeus odoratus mushroom.

Astraeus odoratus, a nutritionally and economically valuable wild edible mushroom, is highly perishable because of its high respiration rate and fragile tissue structure. This study evaluated the effects of short-term anoxic treatment prior to low-temperature storage on the postharvest quality of A. odoratus. Fresh mushrooms were exposed to anoxic conditions for 9 h and stored at 10 ± 2 °C. Anoxia significantly reduced weight loss, electrical conductivity, and hydrogen peroxide accumulation compared with the control. Anoxia-treated samples exhibited higher antioxidant capacity as determined by…

Mitochondrial dysfunction and Ca2+ dysregulation in human iPSC-derived neurons carrying presenilin-1 mutation arise under stress via an MCU-1-independent mechanism.

Impaired cellular activities in Alzheimer’s disease (AD) are linked to metabolic defects and Ca²⁺ dysregulation, but the underlying mechanisms in human neurons are unclear. We performed an integrative analysis using human iPSC-derived neurons (iNs) carrying the Presenilin-1 M146L mutation (PS1M146L). Mutant iNs displayed abnormal Ca²+ dynamics, enhanced mitochondrial respiration, and elevated reactive oxygen species (ROS). KCl-evoked depolarization was reduced, indicating a compromised plasma membrane electrochemical gradient. Under thapsigargin-induced stress, mitochondrial Ca²⁺ ([Ca²⁺]m) was…

Lighting up mitochondrial membranes: Reading and writing the physical state of the inner mitochondrial membrane.

The mitochondrial inner membrane (IMM) is depicted as a scaffold housing the enzymes of the electron transport chain, allowing electron transfer and generation of the electrochemical gradient that drives respiration and adenosine triphosphate (ATP) synthesis. However, evidence from imaging, structural biology, and novel chemical probes reveals that the IMM is not a mere passive backdrop: its physical properties change with the metabolic state, and together these changes influence mitochondrial function. Here, we summarize how IMM microviscosity, lipid packing, and mesoscale organization might …

Attachment representations are associated with interoceptive accuracy but not awareness or sensibility.

Interoception - the perception of internal bodily states such as heartbeat, respiration, and visceral sensations - has been proposed to constitute a fundamental component of self-regulation. Current theorizing proposes that interoception consists of several dimensions: interoceptive accuracy, sensibility, and metacognitive awareness. From an attachment perspective, attachment representations are proposed to shape self-regulation across the lifespan, and are thus expected to influence the processing of bodily signals. We investigated the association between attachment representations and dimens…

Multilayer packaging with CO2/O2-selective gas regulation and humidity-responsive release of cinnamon essential oil for cherry tomato preservation.

Fresh produce preservation remains a major challenge due to nutrient loss and microbial spoilage. Here, we reported a biobased paper window that combined gas regulation with humidity-responsive antimicrobial release of cinnamon essential oil (CEO) to address these challenges. The bifunctional paper window exhibited appropriate gas permeability and CO2/O2 selectivity (2.1-6.0). Molecular dynamics simulations revealed the mechanism of selectivity, showing a 16.9-fold stronger CO2 adsorption than O2 at 100 kPa. Furthermore, humidity generated from produce respiration and transpiration triggered s…

Thermal comfort analysis of car drivers through physiological responses in different cabin environments.

A comfortable cabin thermal environment is vital for driver well-being and safety. To establish quantitative guidelines, this study investigated the relationships between physiological signals and subjective thermal perception under realistic vehicular conditions. We collected thermal sensation and comfort votes (TSV/CSV) from a diverse cohort across a range of cabin temperatures, simultaneously measuring five physiological signals: wrist temperature, electrocardiogram, electrodermal activity, electromyogram, and respiration. Our analysis revealed two key findings: 1) Wrist temperature and spe…

Mitochondrial activity promotes neutrophil degranulation and endothelial dysfunction in systemic infections.

Neutrophils are essential for defense against pathogens but excessive activation in systemic infections can drive immunopathology. We show that neutrophil degranulation can induce endothelial dysfunction via degradation of the glycocalyx and increase of endothelial permeability. To identify targetable pathways regulating neutrophil degranulation in severe inflammation, we compared the proteomes of neutrophils isolated from patients with severe malaria and sepsis. We found significant upregulation of mitochondrial pathways, which was accompanied by increased rates of mitochondrial respiration a…

Ergosterol Peroxide Disrupts Triple-Negative Breast Cancer Mitochondrial Function and Inhibits Tumor Growth and Metastasis.

Ergosterol peroxide (EP) triggers apoptosis pathways by inducing reactive oxygen species (ROS) in TNBC cell lines. Excess ROS production is associated with major damage to mitochondria. We hypothesized that EP may act through ROS-induced mitochondrial dysfunction. Therefore, we performed a series of assays that assessed mitochondrial membrane potential (MMP), cellular respiration, and glycolysis in TNBC models. Cardiomyocytes derived from human-induced pluripotent stem cells were chosen as a non-cancerous model because of their high mitochondrial content. Two in vivo TNBC models were used to q…

Extreme Sensitivity of Greenhouse Gas Production in Mineral Wetland Sediments under Multiple Salinization Regimes.

Wetlands and small inland waters are atmospheric sources of methane (CH4), carbon dioxide (CO2), and nitrous oxide (N2O). Human activities a … ecreasing with NaCl additions, signaling divergent responses of sediment geochemistry and microbial respiration between treatments. Both treatments inconsistently stimulated N2O production. Overall, we have shown that even small increases in salinity (<1 ppt) induce major shifts in wetland GHG production, but that responses depend heavily on salinization chem …

Cellulose cryogel microparticles increase gas permeation of carnauba wax-based edible coating for jujube preservation application.

Wax coating is an important preservation method extending jujube's shelf life. Although wax-based coatings provide a moisture resistant and physical microbial barrier, their dense structure excessively restricts the CO2 and O2 permeation, leading to anaerobic respiration and off-flavor. To increase the CO2 and O2 permeation of wax-based coating while minimizing its impact on inherent properties, microcrystalline cellulose cryogel particles (CCPs) were developed and incorporated into carnauba wax-based edible coating (CCPCW) creating micro gas channel. 1.00 % CCPCW increased the CO2 and O2 perm…

NDP52 deficiency accelerates chondrocyte degeneration through promoting pathogenic mitochondrial ROS via reverse electron transport.

NDP52, a constituent of the selective autophagy receptors (SARs), was recognized for its involvement in facilitating substrate degradation via autophagic bridging. However, its autonomous function apart from autophagy remained largely unexplored. Here, we reported that NDP52 was down-regulated in degenerated chondrocytes. Besides, NDP52 deficiency promoted the extracellular matrix (ECM) degradation, inflammation, cell apoptosis and senescence via its autophagy-independent functions. The absence of NDP52 disrupted the flow of electron respiration chains and led to the production of intracellula…

Copper deficiency disrupts OXPHOS and mitochondrial dynamics through MTCH2-dependent copper trafficking in skeletal muscle.

Copper is an essential trace element required for mitochondrial respiration and cellular metabolism, yet its role in skeletal muscle remains incompletely understood. Here, we show that skeletal muscle-specific deletion of the high-affinity copper importer Ctr1 (SMKO) in mice leads to copper deficiency, resulting in exercise intolerance, metabolic dysfunction, and hallmarks of mitochondrial myopathy, including ragged-red fibers, lactic acidosis, and aberrant mitochondrial morphology. Copper deficiency disrupted electron transport chain proteome and induced mitochondrial hyperfusion. We identifi…

Hydrogen sulfide produced by the gut microbiota impairs host metabolism via reducing GLP-1 levels in male mice.

Dysbiosis of the gut microbiota has been implicated in the pathogenesis of metabolic syndrome (MetS) and may impair host metabolism through harmful metabolites. Here, we show that Desulfovibrio, an intestinal symbiont enriched in patients with MetS, suppresses the production of the gut hormone glucagon-like peptide 1 (GLP-1) through the production of hydrogen sulfide (H2S) in male mice. Desulfovibrio-derived H2S is found to inhibit mitochondrial respiration and induce the unfolded protein response in intestinal L cells, thereby hindering GLP-1 secretion and gene expression. Remarkably, blockin…

P2 Receptor Antagonists Rescue Defective Heme Content in an In Vitro SLC25A38-Associated Congenital Sideroblastic Anemia Cell Model.

Mutations in the SLC25A38 gene are responsible for the second most common form of congenital sideroblastic anemia (CSA), a severe condition for which no effective treatment exists. We developed and characterized a K562 erythroleukemia cell line with markedly reduced expression of the SLC25A38 protein (A38-low cells). This model successfully recapitulated the main features of CSA, including reduced heme content and mitochondrial respiration, increase in mitochondrial iron, ROS levels and sensitivity to oxidative stress. Notably, our study uncovered a new role for extracellular pyridoxal 5'-phos…

Astrocytes produce nitric oxide via nitrite reduction in mitochondria to regulate cerebral blood flow during brain hypoxia.

During hypoxia, increases in cerebral blood flow maintain brain oxygen delivery. Here, we describe a mechanism of brain oxygen sensing that mediates the dilation of intraparenchymal cerebral blood vessels in response to reductions in oxygen supply. In vitro and in vivo experiments conducted in rodent models show that during hypoxia, cortical astrocytes produce the potent vasodilator nitric oxide (NO) via nitrite reduction in mitochondria. Inhibition of mitochondrial respiration mimics, but also occludes, the effect of hypoxia on NO production in astrocytes. Astrocytes display high expression o…
