Hyphal growth is strongly associated with virulence in the human fungal pathogen Candida albicans. While hyphal transcriptional networks have been the subject of intense study, relatively little is known about post-transcriptional regulation. Previous work reported that P-Body (PB) factors Dhh1 and Edc3 were required for C. albicans virulence and filamentation, suggesting an essential role for post-transcriptional regulation of these processes. However, the molecular roles of these factors have not been determined. To further study the function of PB factors in filamentation, we generated homo

Stress granules are biomolecular condensates that form in response to environmental stress and disassemble once normal conditions are restored. However, when disassembly fails, stress granules can persist and solidify. While stress granule solidification has been well documented, the cellular mechanisms underlying the transition from reversible to persistent stress granules remain unclear. Persistent stress granules can seed the formation of pathological aggregates, such as TDP-43 in amyotrophic lateral sclerosis1, 2. Although amyloid and tau aggregates are hallmarks of Alzheimer's disease, a

Non-Saccharomyces yeasts are emerging as promising new probiotics with a beneficial effect equal to or greater than the reference probiotic yeast, Saccharomyces boulardii. Candida intermedia, a non-albicans species not considered a common human pathogen, previously demonstrated probiotic potential. In this work, our objective was to evaluate the immunomodulatory effects of C. intermedia ORQ001 in mice vaccinated with inactivated SARS-CoV-2, seeking further evidence of its probiotic activity. Murine macrophages were stimulated with C. intermedia, followed by mRNA transcription analysis via qPCR…

Candida albicans is an opportunistic fungal pathogen whose growth phase-dependent chemical changes play a critical role in its physiology, pathogenicity, and response to treatment. Here, vibrational spectroscopy combined with chemometric models was applied to investigate differences in spectral signatures of C. albicans yeast cells across different growth phases. C. albicans isolated from different clinical presentations were subjected to spectral analysis at multiple time points over a 24 h period using a Fourier Transform Infrared (FTIR) spectrometer with a built-in Attenuated Total Reflecta

This paper explores the broad probiotic and functional properties of two non-Saccharomyces strains (MI120 and MI125) and one Saccharomyces cerevisiae BB06 strain (as a reference probiotic). Torulaspora delbrueckii MI120 and Starmerella bacillaris MI125 were identified via 5.8S rDNA sequencing. All the strains survived well in simulated gastrointestinal conditions and had strong antioxidant activity (>68%). S. bacillaris MI125 excelled in antimicrobial activity against Gram-positive and Gram-negative bacteria. S. bacillaris MI125 and Sacch. cerevisiae BB06 resisted all the tested antibiotics. N

Ochratoxin A is a mycotoxin commonly found in wine, primarily produced by fungal species from the Aspergillus and Penicillium genera. Due to its nephrotoxic, neurotoxic, immunotoxic, and carcinogenic properties, ochratoxin A contamination in wine is a significant concern for public health. This study investigates the potential of Lachancea thermotolerans in reducing ochratoxin A levels during wine fermentation, evaluating its fermentative performance and impact on key enological parameters. Fermentation trials with 32 L. thermotolerans strains demonstrated considerable variability in fermentat

3-Hydroxypropionic acid (3-HP) is a versatile platform chemical with wide-ranging industrial applications. This study presents a proof-of-concept approach for producing 3-HP from brewer's spent grain (BSG) using a novel one-pot CaO pretreatment method and an engineered, acid-tolerant Issatchenkia orientalis IoDY01H strain. The effects of acid type for pH adjusting of pretreated slurry, nitrogen supplementation, NaHCO3 addition, and BSG deproteinization were evaluated. Results showed that using pure glucose and xylose as carbon sources, yeast extract-peptone (YPDX) medium enhanced 3-HP titers b

Succinic acid (SA), a key platform chemical in biorefineries, faces industrial production challenges stemming from inefficient CO2 fixation and redox imbalances in microbial cell factories. Here, we report a novel strategy to enhance SA biosynthesis in unconventional yeast Yarrowia lipolytica by cooperatively integrating carbonic anhydrase (CA)-mediated CO2 recycling with formate metabolism. Initially, the expression of CA improved the carboxylation efficiency of reductive TCA (rTCA) pathway, resulting in the strain Hi-SA-YlCA, which produced 89.75 g/L of SA with a yield of 1.01 g/g glucose un

Formaldehyde (FA) is a toxic, volatile compound commonly used in industry and frequently found in polluted environments. Its rapid and dependable identification is vital for both environmental assessment and health protection. Microbial biosensors enhanced with nanomaterials offer promising platforms for FA sensing due to their sensitivity, cost-efficiency, and operational simplicity. However, unstable gene expression often limits the practical application of genetically modified microorganisms. To address these challenges, we developed a hybrid biosensing system based on Komagataella phaffii

The oil palm is a prominent tropical oil crop with holds considerable economic value. MYB transcription factors are key regulators in growth and plant stress adaptation mechanisms in plants. However, the roles and operational mechanisms of MYB genes in oil palm are not yet well understood. In this study, EgMYB111 was cloned from oil palm, and its behavior under cold stress was examined in genetically engineered tobacco and oil palm embryoids. Physiological and biochemical analysis demonstrated that genetically engineered lines exhibited substantially greater cold tolerance than control plants.

INTRODUCTION: The development of anastomotic leakage (AL) after esophagectomy is a severe complication, often leading to mediastinitis and systemic infections. Effective empiric antimicrobial therapy is crucial, but there is no consensus on the optimal regimen. This study aimed to document antimicrobial regimens used in the Netherlands and to evaluate culture results from AL after esophagectomy at our center.
METHODS: An online questionnaire about the preferred antimicrobial treatment of AL after esophagectomy was sent to all upper gastrointestinal surgeons in the Netherlands. In addition, dr

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are fatal illnesses forming a neurodegenerative disease continuum. While most ALS/FTD cases are sporadic, a small proportion of cases are linked to mutations in many genes. Among these, hexanucleotide repeat expansions in the C9orf72 gene are the most common and lead to the formation of dipeptide repeat proteins (DPRs), including a proline-arginine dipeptide (PR), which aggregate in the cytoplasm of decaying neurons. As genetics alone fails to explain the etiology of ALS/FTD, it is possible that epigenetic mechanisms - such

Researchers have been fascinated by the remarkable properties of naturally occurring polysaccharides in recent years. We focused on extracting, refining, and analyzing an exopolysaccharide secreted for the first time by the bacterium Paenibacillus mucilaginosus, which was isolated from agricultural soil and identified by 16S rRNA sequence techniques. The polymer's elemental composition was analyzed, focusing on three primary constituents. Carbon emerged as the dominant element, comprising 29.76% of the polymer's total mass. Hydrogen was present at 4.98%, while nitrogen comprised 3.19% of the o

Pathogenic variants in DNM1L, encoding dynamin-like protein-1 (DRP1), cause a lethal encephalopathy. DRP1 defective function results in altered mitochondrial networks, characterized by elongated/spaghetti-like, highly interconnected mitochondria. We validated in yeast the pathogenicity of a de novo DNM1L variant identified by whole exome sequencing performed more than 10 years after the patient's death. Meanwhile, we reviewed the broadness and specificities of DNM1L-related phenotype. The patient, who exhibited developmental delay in her third year, developed a therapy-refractory myoclonic sta

Sporotrichosis is a neglected subcutaneous mycosis of global distribution caused by thermally dimorphic Sporothrix species. Despite their pathogenic potential in vertebrates, the virulence mechanisms underlying their interactions with other organisms remain poorly understood. This study employed the coleopteran insect Tenebrio molitor as an experimental model to evaluate the virulence of seven Sporothrix species from clinical (S. brasiliensis, S. schenckii, S. globosa, S. luriei) and environmental (S. pallida, S. mexicana, S. chilensis) clades. Larvae were inoculated with conidia or yeast cell

Vaginitis is a general term for inflammation of the vagina with symptoms such as vulvovaginal itching, burning, irritation, dyspareunia, odor, or abnormal vaginal discharge. It is a common condition that results in 5 million to 10 million office visits annually. The leading infectious causes of vaginitis are bacterial vaginosis, vulvovaginal candidiasis, and trichomoniasis. Noninfectious causes include atrophic, irritant, and inflammatory vaginitis. Bacterial vaginosis usually presents with a thin, homogenous vaginal discharge and a fishy odor. Vulvovaginal candidiasis often manifests with a w

Candida albicans is a common opportunistic fungus in humans, whose morphological switch between yeast and hyphae forms represents a key virulence trait. Developing strategies to inhibit C. albicans hyphal growth may provide insights into designs of novel antivirulent therapeutics. Importantly, the gut commensal bacterium, Enterococcus faecalis, secretes a bacteriocin EntV which has potent antivirulent and antifungal effects against C. albicans in infection models; however, hampered by the challenges to access large quantities of bioactive EntV, the detailed understanding of its mechanisms on C
