Role of HMGB1 in Neuroinflammation.

HMGB1 (high mobility group box 1 protein) refers to a chromatin-associated protein localized in the cytosol to be secreted to the extracellular milieu from multiple cell types including immune and non-immune cells. In this context, HMGB1 functions as a proinflammatory cytokine. Microglia as the main resident immune cells in the brain actively express and secrete HMGB1. Accumulative evidence suggests that HMGB1, released not only by microglia but also astrocytes and endothelial cells, plays a central role in mediating inflammatory signaling and driving the progression of neuroinflammation. In t…

Glial cells in chronic inflammation: diversity, dysfunction and therapeutic targeting.

Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system (CNS). Current therapies for relapsing MS primarily target the peripheral immune system but have consistently failed to address mechanisms underlying disease progression, which are thought to involve CNS-resident glial cells such as astrocytes, microglia and oligodendrocytes. Recent technological advances have revealed the functional heterogeneity of these glial cells, highlighting their crucial roles in inflammation, demyelination, remyelination and neurodegeneration. In this Review, we summarize emerging …

Spinal Cord Injury Remyelination: Pathways to Therapies.

Spinal cord injury (SCI) is a debilitating condition that results from a culmination of acute and chronic damage to neural tissue, specifically the myelin sheath, thus impacting neurons' abilities to synergistically perform their physiological roles. This review explores the molecular underpinnings of myelination, demyelination, and remyelination, emphasizing the role of oligodendrocyte progenitor cells (OPCs), astrocytes, and microglia in physiological, and pathophysiological, healing. Furthermore, we link these processes with emerging therapeutic strategies currently under investigation in a…

Autoimmune glial fibillary acidic protein astrocytopathy associated meningoencephalomyelitis and bilateral sensorineuronal deafness.

Autoimmune encephalitis is an important group of disease that can mimic infectious encephalitis, with one of the most severe forms being meningoencephalomyelitis. One of the recently identified biomarkers, glial fibillary acidic protein (GFAP), targets the cytosolic intermediate filament protein of astrocytes and causes a variety of clinical symptoms. Here, we report an adult Chinese woman presented with acute onset of confusion, CSF lymphocytosis, markedly elevated total protein mimicking tuberculosis meningitis with rapid deterioration resulted in coma and respiratory failure. She was diagno…

African trypanosomes activate human fetal brain cells to proliferation and IFN-gamma production.

We addressed the host-parasite interplay and the immunopathogenetic events occurring in the central nervous system (CNS) during human African trypanosomiasis. Human first trimester forebrain cells were stimulated with a trypanosome lymphocyte-triggering factor (TLTF) and studied for their immune response as exemplified by cell proliferation and IFN-gamma production. TLTF induced proliferation of human first trimester forebrain cells and IFN-gamma production at the mRNA and protein levels. Astrocytes are the major producers of IFN-gamma in response toTLTE These data illustrated for the first ti…

The immunohistochemical distribution of protein kinase C isozymes is altered in the canine brain and basilar artery after subarachnoid hemorrhage.

The effects of subarachnoid hemorrhage on immunoreactivities of protein kinase C isozymes (alpha, beta, gamma) were studied in canine brain and basilar artery. Mild and severe constriction of the basilar artery was shown 2 days and 7 days after hemorrhage, respectively. In control brain tissues, the isozymes showed distinct distributions and following hemorrhage, reactive astrocytes with protein kinase C alpha staining emerged in the surface of the pons and hippocampus. The control basilar artery showed protein kinase C alpha staining and its staining was decreased on day 7, consistent with th…

Pinealocytes in rats: connexin identification and increase in coupling caused by norepinephrine.

Dye coupling was observed between pinealocytes in acutely dissected pineal glands of adult rats. Pinealocytes maintained in culture were also electrically coupled. Connexins 26 and 43 and their respective mRNAs were present but neither connexin32 nor its mRNA were detected. Pinealocytes expressed only connexin26 whereas connexin43 was confined to astrocytes. In 5-day-old cultures of pinealocytes the incidence of dye coupling and level of immunodetectable connexin26 were low, and both were increased by norepinephrine (NE). The increase in incidence of coupling was maximal at around 6 h after tr…

Vascular changes in the brains with Alzheimer's disease.

Sections of temporal and occipital cortex from 14 patients with Alzheimer's disease were examined by light and electron microscopy. Especially, areas without senile plaques or neurofibrillary degeneration were examined in detail. The initial change in the cerebral cortex occurred in the capillaries. The endothelial cells demonstrated degeneration with hypertrophic distorted basement membranes and swelling of the vascular feet. Lipochromes were found in the processes and cell bodies of the astrocytes. A decrease in the number of nerve cells and their processes noted in the cortices may be due t…

Ultrastructural localization of fibroblast growth factor in neurons of rat brain.

The distribution of fibroblast growth factor (FGF) at the ultrastructural level in the brain of young (15- and 20-day-old) and adult (3-month-old) rats was investigated by immunocytochemistry. Strong staining was observed in most neurons of the cortex of young rat brain. In the same brain area of adult rat many neurons were also stained intensely, while others were negative. Neurons in the other parts of the brain and especially in the adult rat, were generally more weakly stained. The reaction product was located in the cytoplasm of the neuronal cell bodies and their processes. Astrocytes, ol…

Human African trypanosomiasis (T.b. gambiense): a study of 16 fatal cases of sleeping sickness with some observations on acute reactive arsenical encephalopathy.

The principal clinical and pathological findings in 16 fatal cases of human African trypanosomiasis caused by T.b. Gambiense are described. The changes in the brain took the form of a non-specific lymphoplasmacytic meningo-encephalitis of varying intensity. Other features included morular cells, diffuse microglial hyperplasia, and large reactive astrocytes in the white matter. Carditis was identified in 10 cases. Acute reactive arsenical encephalopathy appeared to be the principal cause of death in 10 patients. Convulsions figured prominently in this type of encephalopathy in seven patients an…

Neuron-specific mitochondrial degeneration induced by hyperammonemia and octanoic acidemia.

The neuropathological consequences of acute exposure to the neurotoxicants ammonia and octanoic acid were investigated with the isolated, perfused canine brain preparation. After 1 h of combined hyperammonemia and octanoic acidemia, ultrastructural changes were apparent in all brain regions examined. The cell bodies of neurons were the primary sites of these alterations. Neuronal mitochondria were distended, and the lamellae of the mitochondrial cristae were separated. In some cases the lamellae had completely dispersed, leaving only matrix remnants. Mitochondria of adjacent astrocytes appeare…

Central pontine myelinolysis accompanied by multifocal pseudocalcifications.

A 44-year-old diabetic female became unconscious one month prior to death due to acute suppurative meningitis. Although cerebrospinal fluid findings normalized with the administration of antibiotics, the status of unconsciousness did not improve and she expired. Examination of the brain revealed a small, relatively ill-defined, demyelinated lesion in the central part of the anterior pons, with which hypertrophied astrocytes, lymphocytes, macrophages phagocytizing disintegrated myelin were present. Neuronal perikarya and axons were well preserved. The findings are pathognomonic of central ponti…

[Ultrastructural changes in the CNS of monkeys with the chronic form of tick-borne encephalitis].

The central nervous system of monkeys with chronic tick-borne encephalitis (TBE) shows the following ultrastructural alterations which differ from those in acute TBE: widespread destructive changes in neurons, astrocytes, oligodendrocytes, processes, myelin, vascular walls, severe edema of brain tissue with the signs of a so-called spongious degeneration and the absence of cell proliferation and perivascular cell infiltrations, glial nodules as well as circulatory disorders (hyperemia, stasis, hemorrhages). Few virions with or without and altered supercapsid membrane are detected in the neuron…

Measures of cortical microstructure are linked to amyloid pathology in Alzheimer's disease.

Markers of downstream events are a key component of clinical trials of disease-modifying therapies for Alzheimer's disease. Morphological metrics like cortical thickness are established measures of atrophy but are not sensitive enough to detect amyloid-beta (Aβ)- related changes that occur before overt atrophy become visible. We aimed to investigate to what extent diffusion MRI can provide sensitive markers of cortical microstructural changes and to test their associations with multiple aspects of the Alzheimer's disease pathological cascade, including both Aβ and tau accumulation, astrocytic

MSC-derived extracellular vesicles for spinal cord injury: inflammatory microenvironment modulation, stage-dependent repair mechanisms, and translational challenges.

Spinal cord injury is not only an acute mechanical insult but also the beginning of a prolonged secondary pathological process dominated by inflammatory microenvironmental imbalance. Because the primary traumatic event occurs within minutes and offers a narrow therapeutic window, persistent post-traumatic inflammation, blood-spinal cord barrier disruption, edema, oxidative stress, complement activation, inflammasome signaling, astrocyte reactivity, vascular dysfunction, and extracellular matrix remodeling represent more realistic therapeutic targets. In accordance with the Minimal Information
