The altered expression of inflammation-related microRNAs with microRNA-155 expression correlates with Th17 differentiation in patients with acute coronary syndrome.

MicroRNAs (miRNAs) are a novel class of small, non-coding RNAs that play a significant role in both inflammatory and cardiovascular diseases. Immune cells, especially T helper (Th) cells, are critical in the development of atherosclerosis and the onset of acute coronary syndrome (ACS). To assess whether inflammation-related miRNAs (such as miR-155, 146a, 21, 125a-5p, 125b, 31) are involved in the imbalance of Th cell subsets in patients with ACS, we measured the expression of related miRNAs in patients with acute myocardial infarction (AMI), unstable angina (UA), stable angina (SA) and chest p

Receptor-binding cancer antigen expressed on SiSo cells induces apoptosis via ectodomain shedding.

Receptor-binding cancer antigen expressed on SiSo cells (RCAS1) is a secreted antigen that induces apoptosis in putative receptor-expressing cells, including peripheral lymphocytes and natural killer (NK) cells. RCAS1 expression is associated with aggressive characteristics and poor overall survival for 15 different human malignancies. The putative RCAS1 receptor has not been isolated and the mechanism of RCAS1 apoptosis induction remains unclear. This study explores how RCAS1 is involved in apoptosis initiation. The cell lines SiSo and MCF-7, human uterine carcinoma and breast adenocarcinoma,

Stromelysin-3 mRNA associated with myofibroblasts is overexpressed in aggressive basal cell carcinoma and in dermatofibroma but not in dermatofibrosarcoma.

Stromelysin-3 is produced in the stroma of various malignant tumors, and in breast carcinoma there seems to be a positive correlation between aggressive disease and intensity of stromelysin-3 expression, suggesting that stromelysin-3 participates in the tumor spread. In basal cell carcinoma, previous findings on stromelysin-3 have been inconclusive in this respect. Our study was undertaken to determine the pattern of stromelysin-3 production in relation to different histologic subtypes and stromal reactions in basal cell carcinoma. By in situ hybridization, stromelysin-3 mRNA was detected in s

Characterization of processing requirements and metal cross-reactivities in T cell clones from patients with allergic contact dermatitis to nickel.

Metal ions such as nickel, cobalt, copper and palladium are known to be potent sensitizers in humans, but the antigenic determinants created by these metals as well as the mechanisms of recognition by specific T cell clones are still not elucidated. In this paper, nickel-specific T lymphocyte clones were isolated from four patients exhibiting contact dermatitis to this metal. A panel of 42 independent T cell clones was studied. They were shown to recognize nickel in the context of major histocompatibility complex (MHC) class II molecules and to belong to the CD4 subset. Using fixed autologous

Glucose and glycogen metabolism in erythrocytes from normal and glycogen storage disease type III subjects.

Active glycogen metabolism has been demonstrated in both normal and glycogen-rich erythrocytes taken from patients with type III glycogen storage disease. Activity of all enzymes catalyzing the reactions required for the synthesis and degradation of glycogen have been demonstrated in the mature erythrocytes. Uniformly labeled glucose-(14)C is incorporated into glycogen in intact cells of both types during incubation. Replacement of the glucose-(14)C by unlabeled glucose in the medium resulted in a significant loss of radioactivity from cellular glycogen. In the absence of the substrate a progr

Predictors of mortality among TB-HIV Co-infected patients being treated for tuberculosis in Northwest Ethiopia: a retrospective cohort study.

BACKGROUND: Tuberculosis (TB) is the leading cause of mortality in high HIV-prevalence populations. HIV is driving the TB epidemic in many countries, especially those in sub-Saharan Africa. The aim of this study was to assess predictors of mortality among TB-HIV co-infected patients being treated for TB in Northwest Ethiopia. METHODS: An institution-based retrospective cohort study was conducted between April, 2009 and January, 2012. Based on TB, antiretroviral therapy (ART), and pre-ART registration records, TB-HIV co-infected patients were categorized into "On ART" and "Non-ART" cohorts. A

Idle emissions from heavy-duty diesel vehicles: review and recent data.

Heavy-duty diesel vehicle idling consumes fuel and reduces atmospheric quality, but its restriction cannot simply be proscribed, because cab heat or air-conditioning provides essential driver comfort. A comprehensive tailpipe emissions database to describe idling impacts is not yet available. This paper presents a substantial data set that incorporates results from the West Virginia University transient engine test cell, the E-55/59 Study and the Gasoline/Diesel PM Split Study. It covered 75 heavy-duty diesel engines and trucks, which were divided into two groups: vehicles with mechanical fuel

The methionyl aminopeptidase from Escherichia coli can function as an iron(II) enzyme.

The identity of the physiologically relevant metal ions for the methionyl aminopeptidase (MetAP) from Escherichia coli was investigated and is suggested to be Fe(II). The metal content of whole cells in the absence and presence of expression of the type I MetAP from E. coli was determined by inductively coupled plasma (ICP) emission analysis. The observed change in whole cell concentrations of cobalt, cadmium, copper, nickel, strontium, titanium, and vanadium upon expression of MetAP was negligible. On the other hand, significant increases in the cellular metal ion concentrations of chromium,

Atopic dermatitis induces the expansion of thymus-derived regulatory T cells exhibiting a Th2-like phenotype in mice.

Atopic dermatitis (AD) is a widespread inflammatory skin disease with an early onset, characterized by pruritus, eczematous lesions and skin dryness. This chronic relapsing disease is believed to be primarily a result of a defective epidermal barrier function associated with genetic susceptibility, immune hyper-responsiveness of the skin and environmental factors. Although the important role of abnormal immune reactivity in the pathogenesis of AD is widely accepted, the role of regulatory T cells (Tregs) remains elusive. We found that the Treg population is expanded in a mouse model of AD, i.e

Broad specificity of GDNF family receptors GFRalpha1 and GFRalpha2 for GDNF and NTN in neurons and transfected cells.

The glial cell line-derived neurotrophic factor (GDNF) family of ligands binds to lipid anchored proteins termed GDNF family receptor (GFR)alphas, and then activates the RET receptor tyrosine kinase, by ligand GFRalpha. The binding of soluble GFRalphas to transfected cells suggested that different GFRalphas were dedicated to particular ligands, with GDNF acting primarily or entirely through GFRalpha1, and neurturin (NTN), through GFRalpha2. More recent evidence has suggested the possibility of cross-talk between these ligands and the two receptors. We examined here whether crosstalk between th

Employing the Role of SLC40A1-Mediated Microglia Inflammatory Response in Diabetic Retinopathy Based on Single-Cell Transcriptomics Technology.

Diabetic retinopathy (DR) is one of the most severe complications of diabetes. Existing treatments are effective but still have many limitations. Thus, it is particularly crucial to uncover novel therapeutic targets and develop more efficacious treatment strategies. In this study, single-cell datasets of retinal and normal retinal tissues of DR patients were obtained from the Gene Expression Omnibus (GEO) database. Using cell annotation, Gene Ontology (GO) pathway enrichment analysis, and cell trajectory analysis, key cell subgroups and their enriched pathways affecting DR progression were sel

Gap junction communication regulates luminal-myoepithelial crosstalk and cell differentiation in a bilayered human mammary epithelial cell model.

Interactions between luminal and myoepithelial cells are essential for proper mammary gland development and function. However, only a few in vitro models allow the study of these interactions and their modulation by extrinsic factors. We developed a layered co-culture system (LCS) that mimics the bilayered architecture of the human breast epithelium, enabling direct contact and bidirectional crosstalk between luminal (MCF-12A) and myoepithelial (MYO1089) cells. We confirmed the formation of adherens and functional gap junctions across the layers. Transcriptomic analysis revealed that co-cultur

MAPK pathway inhibitors enhance radioiodine sensitivity in anaplastic thyroid carcinoma through promoting NIS expression and ARF4-mediated NIS membrane transport.

Anaplastic thyroid cancer (ATC) patient develops resistance to radioactive iodine (RAI) due to diminished sodium-iodide symporter (NIS) expression and mislocalization to the cell membrane. MAPK pathway inhibitors have demonstrated efficacy in enhancing RAI avidity in radioiodine-refractory papillary thyroid carcinoma and ATC. Elucidating this mechanism may lead to novel therapeutic approaches for ATC treatment. The cytotoxic effects of three MAPK pathway inhibitors (selumetinib, vemurafenib, dabrafenib) were assessed in ATC cell lines and xenograft models via viability assays and 18F-FDG PET/C

Immunogenic cell death-primed autophagosome vaccines drive dendritic cell cross-presentation and suppress colon cancer metastasis.

Immunogenic cell death (ICD) releases tumor-associated antigens and damage-associated molecular patterns (DAMPs) but also generates substantial suppressive by-products that limit the translation of ICD-based cancer vaccines into effective therapies. The research bottleneck lies in how to selectively enrich these ICD-derived immunogenic substances and efficiently deliver them to the most potent antigen cross-presenting dendritic cells (cDC1), thereby priming CD8+ T cell immunity. By leveraging the natural autophagy process within cells, this study presents an ICD-Primed Autophagosome Engineerin

Nanoconjugate Strategy Integrating Immunogenic Cell Death Induction and Innate Immune Activation for Potent Anti-Cancer Immunity.

In vivo immune modulation can potentially benefit broader cancer patients. Efficient cross-priming of anti-tumor T cells requires "three-signal" dendritic cells (DCs) that simultaneously integrate antigenic, co-stimulatory, and migratory programs, which remains a major challenge. Here, we developed a carrier-free peptide-cyclic dinucleotide (CDN) nanoconjugate (PCN) that co-activates immunogenic cell death (ICD) in tumors and stimulator of interferon genes (STING) signaling in DCs. Library screening identified ICD-inducing peptides enriched in hydrophobic and cationic residues that promote sel

Evidence that S-phase kinase associated protein 2 (SKP2) is ubiquitinated and degraded via a Rho-related BTB domain containing 1 (RhoBTB1) and Cullin-3 mechanism in placenta.

Rho related BTB domain containing 1 (RhoBTB1) is highly expressed in placenta and functions to deliver protein targets to the Cullin-3 (CUL3) E3 ubiquitin ligase where they are targeted for ubiquitination and degradation. The targets of RhoBTB1 in placenta have not been identified. Using RNAscope, we show that RhoBTB1 is mainly expressed in syncytiotrophoblasts (SCT) in the human and mouse placenta. We employed ascorbate peroxidase 2-mediated targeted proteomics to identify RhoBTB1 binding proteins in immortalized human extravillous trophoblast (HTR8/SVneo) cells. We selected 9 RhoBTB1-interac

BMAL1 regulates the initial step of embryo implantation by modulating endometrial cell adhesion and migration.

Embryo implantation is a critical and tightly regulated process essential for successful human reproduction. Although the role of endometrial receptivity is well established, the molecular mechanisms governing this process are not thoroughly understood. Circadian rhythm regulators, particularly brain and muscle aryl hydrocarbon receptor nuclear translocator-like protein 1 (BMAL1), have emerged as key modulators in reproductive biology. In this study, we investigated the role of BMAL1 in regulating endometrial receptivity and embryo implantation. Ishikawa cells were transfected with BMAL1-speci

Transcriptomic landscape of Gallbladder cancer reveals altered pathways related to cell cycle and Aurora kinase.

Gallbladder cancer (GBC) is a rare but aggressive biliary tract malignancy. This study explores the transcriptomic profile of GBC to identify differentially expressed genes (DEGs) and dysregulated pathways involved in its pathogenesis. RNA sequencing was performed on 13 GBC tumors and 6 matched controls. Functional enrichment analysis (FEA) as well as weighted gene co-expression network analysis (WGCNA) were used to identify dysregulated pathways, functionally relevant gene modules and hub genes. Key targets were validated in patient tissues and cell lines. A total of 1319 DEGs were identified

Inhibiting AIDA suppresses VSMC-derived foam cell formation and atherosclerosis by hindering CD36 membrane translocation.

Vascular smooth muscle cell (VSMC)-derived foam cells critically drive atherosclerotic plaque progression, yet their regulatory mechanisms remain incompletely understood. This study aimed to elucidate the pathophysiological role of the VSMC-enriched factor axin interactor, dorsalization-associated (AIDA) in this process and evaluate its therapeutic potential. We utilized VSMC-specific AIDA knockout in male ApoE-/- mice and adenoviral shRNA silencing in oxLDL-stimulated murine aortic vascular smooth muscle cells (MOVAS), combined with co-immunoprecipitation (Co-IP), cholesterol flux assays, and
