Sialoside Arrays: New Synthetic Strategies and Applications.

Sialic acid-containing carbohydrates, or sialosides, play critical roles in many biological events and in diseases, including viral and bacterial infections, the immune response, the progression of tumor cell metastasis, etc. Despite the importance, the limited access to complex sialosides had prevented extensive studies on the function and significance of sialic acid structural diversity. However, recent advances in synthetic sialoside chemistry, such as the novel chemoenzymatic or stereochemical approach, have produced homogeneous size- and structure-defined sialosides to create diverse sial

Nicotinamide and the skin.

Nicotinamide, an amide form of vitamin B3, boosts cellular energy and regulates poly-ADP-ribose-polymerase 1, an enzyme with important roles in DNA repair and the expression of inflammatory cytokines. Nicotinamide shows promise for the treatment of a wide range of dermatological conditions, including autoimmune blistering disorders, acne, rosacea, ageing skin and atopic dermatitis. In particular, recent studies have also shown it to be a potential agent for reducing actinic keratoses and preventing skin cancers.

RECQ DNA helicases and osteosarcoma.

The RECQ family of DNA helicases is a conserved group of enzymes that are important for maintaining genomic integrity. In humans, there are five RECQ helicase genes, and mutations in three of them-BLM, WRN, and RECQL4-are associated with the genetic disorders Bloom syndrome, Werner syndrome, and Rothmund-Thomson syndrome (RTS), respectively. Importantly all three diseases are cancer predisposition syndromes. Patients with RTS are highly and uniquely susceptible to developing osteosarcoma; thus, RTS provides a good model to study the pathogenesis of osteosarcoma. The "tumor suppressor" role of

BTG1 expression correlates with the pathogenesis and progression of ovarian carcinomas.

BTG (B-cell translocation gene) can inhibit cell proliferation, metastasis, and angiogenesis and regulate cell cycle progression and differentiation in a variety of cell types. We aimed to clarify the role of BTG1 in ovarian carcinogenesis and progression. A BTG1-expressing plasmid was transfected into ovarian carcinoma cells and their phenotypes and related proteins were examined. BTG1 mRNA expression was detected in ovarian normal tissue (n = 17), ovarian benign tumors (n = 12), and ovarian carcinoma (n = 64) using real-time RT-PCR. Ectopic BTG1 expression resulted in lower growth rate, high…

REG family proteins and immune regulation: current evidence and perspectives.

As a group of small secretory proteins widely expressed in digestive and nondigestive tissues, the regenerating islet-derived (REG) family m … bfamily members on dendritic cell phenotype and function have recently been uncovered in pancreatic carcinogenesis and autoimmune diabetic models. Certain REG proteins also exhibit context-dependent modulatory effects on T cell and neutrophil responses, though current observations remain fragmentary. Research regarding REG-mediated immune regulation is still in …

Pathophysiological affair between protein kinase D and epithelial ovarian cancer: A roadmap towards translating the mechanism from bench to bedside.

Epithelial ovarian cancer, especially high-grade serous ovarian cancer (HGSOC) remains deadliest gynecological malignancy due to lack of early-detection biomarkers and chemoresistance. Protein kinase D (PKD1-3), a member of diacylglycerol (DAG)-targeting, Ca++-Calmodulin family of serine/threonine kinases emerged as a novel, targetable driver of HGSOC. This review synthesizes significance of currently discovered molecular mechanisms of PKD-driven carcinogenesis of the ovary, promise of CRT0066101, a highly selective, potent PKD inhibitor as excellent anti-cancer agent and critically discussed …

Targeting MSR1+ tumor-associated macrophages enhances the therapeutic efficacy of anti-PD-L1 in hepatocellular carcinoma by suppressing NF-κB pathway activation.

MSR1+ tumor-associated macrophages (TAMs) have been implicated in various malignancies; however, their functional role in Hepatocellular car … than either treatment alone. Our findings demonstrate that MSR1+ TAMs contribute to hepatocellular carcinogenesis through the NF-κB/IL-6 signaling axis by promoting MDSCs accumulation and impairing CD8+ T cell responses. Effectively targeting MSR1+ TAMs can overcome resistance to anti-PD-L1 therapy, offering a promising new immunotherapeutic approach for HCC.

The role of pluripotency regulators in triple-negative breast cancer immune response.

Triple-negative breast cancer (TNBC) is defined by the absence of estrogen, progesterone, and HER2 receptor expression. A critical challenge in managing TNBC is its high concentration of cancer stem cells (CSCs), which drives chemotherapy resistance and correlates with poor patient survival. In normal physiology, stem cell pluripotency and differentiation are governed by core transcription factors (such as Oct4, Sox2, Nanog, Klf4, and c-Myc) alongside key signaling networks, including the Notch, Wnt/β-catenin, and Sonic Hedgehog (Shh) pathways. During carcinogenesis, aberrant activation of the…

Di(2-ethylhexyl) phthalate and mono(2-ethylhexyl) phthalate upregulate PPARG and induce metabolic reprogramming in hepatocellular carcinoma cells: a pan-cancer and metabolomics study.

Di(2-ethylhexyl) phthalate (DEHP) and its primary metabolite mono(2-ethylhexyl) phthalate (MEHP) have been implicated in metabolic dysregulation and carcinogenesis, but their potential links to hepatocellular carcinoma (HCC) remain unclear. Using network toxicology and pan-cancer analyses, we identified PPARG as a candidate shared target of DEHP and MEHP. PPARG was upregulated in HCC tissues, and higher expression was associated with poorer overall survival in the TCGA-LIHC cohort. Molecular docking and dynamics simulations predicted a more favorable and stable interaction between MEHP and PPA…

Oral microbiome dysbiosis of Porphyromonas gingivalis in oral leukoplakia, oral squamous cell carcinoma and radiation-induced oral mucositis: a qPCR-based cross-sectional study.

BACKGROUND: Porphyromonas gingivalis is a key periodontal pathogen increasingly implicated in periodontitis and oral carcinogenesis. Among its virulence determinants, fimbriae encoded by the fimA gene exhibit pathogenic potential. While previous studies have mainly focused on fimA II and IV genotypes in periodontitis, limited data exist on the role of fimA I in oral squamous cell carcinoma (OSCC), and radiation-induced oral mucositis (RIOM). The present study aimed to evaluate the relative abundance of the fimA I genotype across different oral disease conditions and to assess its potential rol…

Gastric Cancer Biomarkers: From Cellular Identity to Precision Oncology.

Gastric Cancer (GC) is characterized by remarkable biological heterogeneity, reflected by the continuous expansion of biomarkers with diagnostic, prognostic, predictive, and therapeutic relevance. However, most reviews organize these biomarkers according to signaling pathways or individual molecular alterations, often overlooking the interconnected biological processes and functional states that shape gastric carcinogenesis and tumor evolution. This review proposes a biology-oriented conceptual framework that interprets GC biomarkers through five fundamental biological questions capturing inte…

From Personal Trouble to Public Issue: Early-Onset Colorectal Cancer Among Young Latino Men.

C. Wright Mills' concept of the sociological imagination connects personal troubles to public issues. This editorial, co-authored by a sociologist, a Latino cancer survivor, and a community organizing director, applies that framework to early-onset colorectal cancer (EOCRC) among young Latino men, where molecular epidemiology and structural inequities converge. Recent genomic research identifies colibactin mutational signatures disproportionately present in EOCRC tumors, suggesting early-life microbial exposures may shape later carcinogenesis. Yet biological evidence alone cannot explain why y…
