Microglial activation, tau and amyloid deposition in TREM2 p.R47H carriers and mild cognitive impairment patients: a multi-modal/multi-tracer PET/MRI imaging study with influenza vaccine immune challenge.

BACKGROUND: Microglia are increasingly understood to play an important role in the pathogenesis of Alzheimer's disease. The rs75932628 (p.R47H) TREM2 variant is a well-established risk factor for Alzheimer's disease. TREM2 is a microglial cell surface receptor. In this multi-modal/multi-tracer PET/MRI study we investigated the effect of TREM2 p.R47H carrier status on microglial activation, tau and amyloid deposition, brain structure and cognitive profile. METHODS: We compared TREM2 p.R47H carriers (n = 8; median age = 62.3) and participants with mild cognitive impairment (n = 8; median age =

A further study of relations between motor impairment and social communication, cognitive, language, functional impairments, and repetitive behavior severity in children with ASD using the SPARK study dataset.

Motor impairments are pervasive and persistent in children with autism spectrum disorder (ASD) throughout childhood and adolescence. Based on recent studies examining motor impairments in children with ASD between 5 and 15 years (i.e., SPARK study sample), 87-88% of this population is at-risk for a motor impairment, these problems persisted until 15 years, and related to their core (social communication skills and repetitive behaviors [RBs]) and comorbid (language, cognitive, and functional) impairments. Persistent motor impairments extending into adolescence/adulthood could negatively impact

Essential oils and functional herbs for healthy aging.

As total life expectancy increases, the prevalence of age-related diseases such as diabetes and Alzheimer's disease is also increasing. Many hypotheses about Alzheimer's disease have been developed, including cholinergic neuron damage, oxidative stress, and inflammation. Acetylcholine is a major neurotransmitter in the brain and cholinergic deficits leads to cognitive dysfunction and decline. Recent studies have linked diabetes as a risk factor in developing Alzheimer's disease and other types of dementia. The incidence of patients with type II diabetes and increased levels and activity of α-a

Correlation of specific amyloid-β oligomers with tau in cerebrospinal fluid from cognitively normal older adults.

IMPORTANCE: To improve the ability to develop treatments that prevent incipient Alzheimer disease (AD) from progressing to overt AD, it is important to understand the molecular basis of the earliest pathophysiological abnormalities and to determine how amyloid-β (Aβ) is involved very early in its pathogenesis. OBJECTIVE: To investigate 2 specific Aβ oligomers, Aβ trimers and Aβ*56, in human cerebrospinal fluid (CSF); evaluate the effects of aging and AD; and obtain support for the hypothesis that they may be pathogenic by determining their relationships to CSF tau. DESIGN: A CSF sampling stu

Simultaneous transcutaneous electrical nerve stimulation mitigates simulator sickness symptoms in healthy adults: a crossover study.

BACKGROUND: Flight simulators have been used to train pilots to experience and recognize spatial disorientation, a condition in which pilots incorrectly perceive the position, location, and movement of their aircrafts. However, during or after simulator training, simulator sickness (SS) may develop. Spatial disorientation and SS share common symptoms and signs and may involve a similar mechanism of dys-synchronization of neural inputs from the vestibular, visual, and proprioceptive systems. Transcutaneous electrical nerve stimulation (TENS), a maneuver used for pain control, was found to influ

Cerebral hemodynamic and metabolic effects of chronic alcoholism.

The nervous system is particularly susceptible to the harmful effects of alcohol. These include Wernicke-Korsakoff syndrome, which is related to thiamine deficiency secondary to chronic alcohol abuse. Other neurotoxic effects of alcohol with cognitive impairments include delirium tremens, alcoholic seizures or "rum fits," and alcoholic neuropathies. It has become recognized in recent years that alcohol and its metabolites directly damage the nervous system even in the absence of nutritional deficiencies. Cerebral blood flow (CBF) measurements provide a noninvasive indirect monitor of cerebral

How teacher support shapes reading literacy among urban primary school pupils in China: evidence from achievement emotions and reading self-efficacy.

BACKGROUND: Reading literacy is a fundamental determinant of academic achievement. However, the complex psychological mechanisms linking teacher support to reading literacy during the critical developmental transition of middle childhood remain insufficiently understood. This gap is particularly evident within the Confucian Heritage Culture (CHC) context, where teacher authority and social validation are highly prominent. Grounded in an integration of control-value theory and social cognitive theory, this study aims to investigate the parallel mediating roles of achievement emotions (an affect

Prevalence and risk factors of elopement behavior in children and adolescents with autism spectrum disorder: a systematic review and meta-analysis.

OBJECTIVE: Wandering and elopement are common and potentially life-threatening behaviors in children and adolescents with autism spectrum disorder (ASD). Despite extensive research, debates persist regarding the chronic nature of these behaviors, their relationship with cognitive and linguistic functions, and the primary risk factors. This study aims to quantitatively assess the incidence of elopement in this population and systematically evaluate associated risk factors. METHODS: Systematic searches were conducted in PubMed, Embase, Web of Science, and Cochrane Library databases to gather ob

The AHS-R: A holistic thinking measure with expanded theoretical domains and improved score reliability.

Analytic and holistic thinking represent distinct cognitive styles. The existing Analysis-Holism Scale (AHS) is the most widely used measure of holistic thinking but has faced psychometric challenges and its validation has been largely restricted to a limited cultural context. The purpose of this study was to develop a refined measurement instrument, the AHS-R (Analysis-Holism Revised Scale), that expands the theoretical representation of the holistic cognition domains by differentiating between the Midway (preference for compromise) and Contradiction (tolerance for contradiction) dimensions a

The NeuroImmunoEndocrine Circuit of Umami Peptides: A Systems Biology Approach.

Background/Objectives: Umami peptides enhance flavor and contribute to appetite regulation (satiety) and metabolic health. By signaling to the orbitofrontal cortex, umami has been shown to improve cognitive function in Alzheimer's disease dementia. This taste boosts the immune system and induces saliva secretion. However, the molecular mechanisms linking umami peptides to systemic physiology remain poorly understood. This study provides the first integrated analysis of neurological, immunological, and endocrinological pathways activated by umami peptides. Methods: Novel umami peptides were ide

Glory or cage? A qualitative exploration of moral injury among retired college soldiers in Mainland China.

OBJECTIVE: Moral injury correlates significantly with posttraumatic stress disorder, depression, and suicide, affecting soldiers' combat effectiveness and well-being. Despite growing awareness, it is an emerging research area, mainly studied in Western military cohorts. In China, research on its mechanisms and influencing factors in the unique social-cultural context is scarce. Retired college soldiers are crucial, being high-quality military reserves and national defense educators. Their moral injury during service impacts postretirement adaptation and college conscription. Thus, exploring th

Sex differences in the impact of nerve injury on locus coeruleus function and behaviour in mice.

Chronic pain often coexists with anxiety and depression, particularly in women. The locus coeruleus (LC), a noradrenergic nucleus, modulates pain and emotional states and shows sex-specific structural and functional properties in preclinical studies. Moreover, clinical evidence indicates sex differences in the experience of chronic pain and its emotional impact. These findings highlight the need to explore the LC mechanisms underlying sex-dependent differences in chronic pain. In this study, we examined the sex-specific role of the LC in chronic pain and its emotional and cognitive consequence
