In vitro suppression of myelopoiesis by adherent murine splenocytes in experimental disseminated histoplasmosis.

Supernatant culture media obtained from adherent spleen cell preparations of mice experimentally infected with Histoplasma capsulatum yeast cells suppressed in vitro growth of human myeloid cells. This suppression was significantly greater than that obtained when splenocytes from normal mice or those inoculated with killed yeast cells were used. The use of indomethacin partially blocked this suppressive effect. A direct relationship was observed between the levels of prostaglandin E (PGE) per 10(6) adherent cells in the splenocyte preparations and the degree of in vitro myelosuppression. These

[Morphological analysis of spontaneous hemoblastoses in dogs and cats].

Information of the frequency and morphological characteristics of spontaneous hemoblastoses in dogs and cats observed in 1966-1978 are presented. Hemoblastoses were diagnosed in 90 dogs and 12 cats. Lymphoproliferative diseases were most frequent: lymphosarcomas (80 cases) and lymphocytic leukemias (11 cases). Ten dogs had mastocytomas and one--myeloid leukemia. The cell composition of the organs and tissues involved permitted to distinguish 3 cellular variants of lymphosarcoma: poorly differentiated, lymphoblastic, and prolymphocytic; in both species the poorly differentiated type of lymphosa

Histopathology of Endolymphatic subarachnoid shunt surgery for Meniere's disease.

The histopathological findings in the temporal bones of three patients who underwent endolymphatic subarachnoid shunt operation for Meniere's disease are reported. The postoperative follow-up period varied from three to five years. Two patients had a successful operation; the third was unsuccessful. The continuity between th mesothelial lining of the endolymphatic sac and the subarachnoid space was demonstrated in one case; no definite conclusions could be made for the other two cases. Although failure of shunt surgery was presumably due to fibrosis in the shunt area in the reported case, the

[Contusion and permeability of the hemato-aqueous barrier to fluorescein. Effect of a pretreatment by local indometacine (author's transl].

An experimental study, by angiography of the anterior segment of rabbit's eye, estimates the permeability of the hemato-aqueous barrier, using fluorescein. A trauma of the eye (contusion) leads to a rapid and significant increase in this permeability. However, local pretreatment, using an ophthalmic solution of 0,33% indometacine, almost completely stabilizes the hemato-aqueous barrier, even for elements as diffusible as fluorescein. These results suggest that application of a similar preparation to the human eye could be useful in the treatment of ocular contusions, chemical burns and angle-c

A review of the metabolism and toxicology of nickel.

Although nickel is an essential element for animal nutrition, the physiological role of nickel is not yet established. Pathological alterations of nickel metabolism are recognized in several human diseases. The diverse clinical manifestations of nickel toxicology include: (1) acute pneumonitis from inhalation of nickel carbonyl, (2) chronic rhinitis and sinusitis from inhalation of nickel aerosols, (3) cancers of nasal cavities and lungs in nickel workers, and (4) dermatitis and other hypersensitive reactions from cutaneous and parenteral exposures to nickel alloys. The toxicity, carcinogenici

Sub-plasmalemmal linear density: a common structure in globoid cells and mesenchymal cells.

Sub-plasmalemmal linear densities of variable length (0.1 approximately 1.0 mu) were found to be a constant feature of globoid cells in human as well as in canine globoid cell leukodystrophy (GLD). Similar densities were also observed in experimental globoid cells and epithelioid cells in chronic granuloma but not in glial cells. The linear densities always appeared without any relation to basal laminae. These observations together with the other reports of similar structures in lymphoma, fibroma and sarcoidosis suggest that the sub-plasmalemmal density is a structure frequently observed in me

Translational validity of rodent diet-induced obesity models for cognitive impairment in human metabolic disease.

Type 2 diabetes and obesity, often modelled in rodents through high-fat diet interventions, are associated with altered brain energy metabolism and cognitive impairments. However, discrepancies between rodent high-fat diet models and human metabolic diseases challenge the translation of findings. This review provides a view on clinical and preclinical studies examining the effects of high-fat diet-induced metabolic dysfunction on brain function, focusing on the limitations of current rodent models, including diet composition, duration, and control diets. It is discussed how these factors influ

Neural correlates of perceptual consciousness from within: A narrative review of human intracranial research.

Despite many years of research, the quest to identify neural correlates of perceptual consciousness (NCC) remains unresolved. One major obstacle lies in methodological limitations: most studies rely on non-invasive neural measures with limited spatial or temporal resolution, making it difficult to disentangle proper NCCs from concurrent cognitive processes. Additionally, the relatively low sensitivity of non-invasive neural measures limits the interpretation of null findings in studies targeting proper NCCs. In this review, we discuss how human intracranial recordings can advance the search fo

Aggregation in gut: on the link between neurodegeneration and bacterial functional amyloids.

Amyloids are insoluble protein aggregates with a cross-beta structure, which are traditionally associated with neurodegeneration. Similar structures, named functional amyloids, expressed mostly by microorganisms, play important physiological roles, e.g., bacterial biofilm stabilization. Using a bioinformatics approach, we identify gut microbiome functional amyloids and analyze their potential impact on human health via the gut-brain axis. The results point to taxonomically diverse sources of functional amyloids and their frequent presence in the extracellular space. The retrieved interactions

There Is More Than Meets the Eye: The Dual Role of Perception in Shaping Color Lexicons.

Color's ultimate physical reality is continuous, and yet human beings "cut" this continuum into a rather small number of categories reflected in their languages' color lexicon. There are striking cross-linguistic differences in the color lexicon, which are primarily attributed to differences in communicative needs, but also striking commonalities, attributed to a common human perceptual system. Here, we challenge this notion of a uniform perceptual structure by experimentally comparing participants from two populations, French from France and Himba from Namibia. They differ in the amount of ex

Delta-9-tetrahydrocannabinol and Cannabidiol for Pain: Preclinical and Clinical Models.

Cannabinoids are increasingly being used to manage pain resulting from a variety of conditions. Both preclinical animal models and human studies have played a crucial role in advancing our knowledge of cannabinoids, their involvement in pain mechanisms, and their potential utility as novel analgesics. This chapter first reviews basic pain neurobiology and the most common experimental pain paradigms, which provide a basis for our discussion of preclinical, human laboratory, and clinical research characterizing the effectiveness of cannabinoids for managing pain. While a substantial body of lite

The crystal structure of human transport and Golgi organization 2 homolog (TANGO2) suggests a cysteine N-terminal nucleophile (Ntn) hydrolase.

Recently, there has been growing interest in the function and physiological importance of human TANGO2 (transport and Golgi organization 2 homolog), particularly whether it acts as a heme-trafficking protein. To address this question, we experimentally determined the three-dimensional structure of TANGO2. Our crystallographic analysis indicates that interactions between heme and TANGO2 are nonspecific. Structural comparison of the TANGO2 crystal structure with known cysteine Ntn-hydrolases allowed us to identify a putative active site, catalytic residues and a substrate-binding cavity that cor

A pericyte-to-myofibroblast transition links APOE4 to cerebrovascular degeneration.

Cerebrovascular disease is a major but poorly understood feature of Alzheimer's disease (AD). The strongest genetic AD risk factor, apolipoprotein E4 (APOE4), is associated with cerebrovascular degeneration, including vascular amyloid deposition and fibrosis. To uncover how APOE4 promotes cerebrovascular pathology, we assembled a single-cell transcriptomic atlas of human brain vasculature. In APOE4 carriers, pericyte abundance was significantly reduced and accompanied by the emergence of a myofibroblast-like cell population co-expressing contraction and extracellular matrix genes. Zebrafish model for studying vascular development and vascular-related disease.

Zebrafish (Danio rerio) is a powerful vertebrate animal model system for cardiovascular research. Their small size, optical transparency, fecundity, rapid development, remarkable regenerative capacity, and the availability of numerous fluorescent transgenic lines have made them an excellent experimental model for studying vascular development and disease. There is a high degree of conservation in the anatomy and physiology of the zebrafish vascular system relative to other vertebrates, thereby enabling zebrafish to model human vascular pathophysiology. Questions concerning genetic vascular dis

Thymic regeneration and tumor immunotherapy.

The thymus, as a vital immune organ in the human body, provides the essential microenvironment for the development, differentiation, and maturation of T lymphocytes. With advancing age, the thymus gradually undergoes atrophy and degeneration, primarily characterized by structural disruption of thymic epithelial cells, leading to slowed T cell development and reduced output. Experimental studies have identified multiple regenerative pathways, including IL-7, KGF, BMP4, IL-22, RANKL, regulatory T cells, FOXN1-directed approaches, and sex-steroid blockade. Recent human observational studies also

Metabolic and molecular regulation in skeletal muscle dysfunction and regeneration.

Skeletal muscle is an important organ in the human body for maintaining overall strength and mobility. Skeletal muscle has the capability of self-regeneration, which can be achieved by utilizing specific energy pathways. Therefore, understanding the energy metabolism of skeletal muscle is essential to exploring its regenerative mechanisms. This review addresses the current progress in understanding the essential role of metabolic pathways in skeletal muscle function, regeneration, and muscle dysfunction as it relates to diseases such as type 2 diabetes mellitus (T2DM), obesity, and aging (sarc

Intrinsic Gestational Timing Governs Human Cerebellar Development After Preterm Birth.

Intrinsic biological clocks govern human brain development, but whether these programs recover from early disruption, such as premature birth, remains unknown. The cerebellum, with peak maturation in the last trimester, provides a model to address this challenge. We analyzed an unprecedented combination of in-vivo and postmortem cohorts of human postnatal cerebella spanning 22-42 weeks' gestation, integrating longitudinal neuroimaging, spatial transcriptomics, and machine-learning-based histology to capture developmental states inaccessible to experimental models. Gestational age imposed lasti

Functional Reprogramming of Peripheral Eosinophils in Lentivirus-Infected Rhesus Macaques.

As innate immune cells, granulocytic eosinophils form part of the first line of defense against pathogens. While recent studies indicate that granulocytes have additional functions including anti-inflammatory roles, tissue homeostasis maintenance, remodeling, and trained innate immune memory, they remain understudied in viral infections, specifically in human immunodeficiency virus (HIV) infection. Using a rhesus macaque model of simian-human immunodeficiency virus (SHIV) infection, we evaluated the functional responses of peripheral granulocytes using a newly developed whole blood intracellul

Delineation of the Human Germinal Centre Immune Landscape Using Multiplex Imaging Analysis.

Given the role of follicular immune dynamics, especially the germinal centre, for the development of pathogen-specific antibodies, their in situ characterisation is of great importance. We have developed a multiplex immunofluorescence imaging pipeline that allows the analysis of human follicular adaptive and innate immune cell subsets. Our data revealed the in situ phenotypic heterogeneity and differential localisation of follicular helper CD4 T (TFH) cell subsets across follicular areas in tonsils and reactive lymph nodes (LNs). Cell clustering analysis identified specific TFH subsets with di

Mouse Papillomavirus Outbreak in a Research Facility.

The use of mouse papillomavirus (MmuPV1) to study infections, disease outcomes, and vaccine strategies in mice has greatly enhanced our understanding of human papillomavirus. However, as with other species-specific infectious agents used as models for human disease, such studies may pose a risk to facilities that house large numbers of the model agent's natural host, especially when the full natural history of the infection is uncertain. In this study, we describe our recent experience showing that containment of MmuPV1 can be difficult, and that its use in research facilities may cause unexpe
