Why does rotavirus vaccine effectiveness vary around the globe?

Rotavirus vaccines have transformed the prevention of severe childhood diarrhea globally, yet their effectiveness remains markedly lower in low- and middle-income countries (LMICs), where rotavirus mortality is greatest. Understanding the biological and environmental determinants of this disparity is critical for optimizing existing vaccination strategies and informing next-generation vaccine development. This review examines the factors contributing to variable rotavirus vaccine effectiveness across different settings, with a focus on maternal antibodies, infant gut health, co-infections, co-

Sex-specific links between stress regulation in early life and anxiety in middle childhood.

Anxiety disorders become more prevalent in females from middle childhood onwards. Yet it is likely that predictors for developing these disorders occur earlier, with emerging evidence of sex differences. We examined whether early life predictors, including maternal depression, placental cortisol bioavailability, and infant cortisol reactivity, predict sex differences in anxiety disorders at age 8. Data are from 149 mothers recruited in the Mercy Pregnancy and Emotional Wellbeing Study (MPEWS) before 20 weeks' gestation and their children followed to 8 years. Maternal measures included the Stru

Maternal high-fat diet programs reproductive function of female offspring rat through IL-6-mediated regulation of Kiss1 gene methylation.

Maternal high-fat diet (HFD) is known to impair the reproductive function of female offspring, but the underlying epigenetic mechanisms of this developmental programming remain unclear. In this study, female rats were fed either a control diet (CD; 10 % kcal from fat) or an HFD (60 % kcal from fat) prior to and during gestation and lactation. After weaning, female offspring were randomly assigned to continue on either a CD or HFD, resulting in four groups: C/C, C/HF, HF/C, and HF/HF. Maternal HFD significantly reduced primordial follicle numbers, increased follicular atresia and granulosa cell

Patterns of Congenital Syphilis in a Large Public Hospital: Maternal Risk Factors and Infant Outcomes.

BACKGROUND: Congenital syphilis (CS) in the United States increased 10-fold in the last decade. At a large public hospital in Los Angeles, the numbers of infants born to mothers with untreated syphilis during pregnancy have continued to rise. METHODS: We compiled a retrospective case series from all infant rapid plasma reagin test results from 2022 to 2023 summarizing CS diagnosis and relevant diagnostic criteria. χ2 and Fisher exact tests were used to examine associations between maternal risk factors and CS diagnosis. RESULTS: Maternal syphilis complicated 97 of 2367 live births (4.1%) at

Maternal investment and early thermal conditions affect performance and antipredator responses.

Exposure to increased temperatures during early development can lead to phenotypic plasticity in morphology, physiology, and behavior across a range of ectothermic animals. In addition, maternal effects are known to be important contributors to phenotypic variation in offspring. Whether the 2 factors interact to shape offspring morphology and behavior is rarely explored. This is critical because climate change is expected to impact both incubation temperature and maternal stress and resource allocation. Using a fully factorial design, and Bayesian multivariate mixed models, we explored how the

Racial inequality in perinatal outcomes in two Brazilian birth cohorts.

This study aimed to estimate and compare racial inequality in low birth weight (LBW), preterm birth (PTB), and intrauterine growth restriction (IUGR) in two Brazilian birth cohorts. This was a cross-sectional study nested within two birth cohorts in Ribeirão Preto (RP) and São Luís (SL), whose mothers were interviewed from January to December 2010. In all, 7430 (RP) and 4995 (SL) mothers were interviewed. The maternal skin color was the exposure variable. Associations were adjusted for socioeconomic and biological covariates: maternal education, per capita family income, family economic classi

Maternal substance use in pregnancy: Differential prediction by childhood adversity subtypes.

Substance use in pregnancy, including alcohol use, drug use, or smoking, is associated with poor health outcomes for both the mother and her unborn child. Building on previous research that has examined the cumulative impact of Adverse Childhood Experiences (ACEs) on maternal binge drinking and alcohol use in pregnancy, the current study sought to examine the association between maternal ACEs and substance use in pregnancy more broadly, including alcohol use, binge drinking, smoking, and drug use. Furthermore, we also examined how different adversity subtypes, including sexual abuse, family vi

Associations between different dimensions of prenatal distress, neonatal hippocampal connectivity, and infant memory.

Prenatal maternal distress-an umbrella concept encompassing multiple negative psychological states including stress, anxiety, and depression-is a substantial prenatal exposure. Consistent across preclinical and human studies, the hippocampus displays alterations due to prenatal distress. Nevertheless, most prenatal distress studies do not focus on multiple dimensions of, have not examined hippocampal functional connectivity in association with, and do not consider observer-based functional outcomes related to distress. We investigated the effects of different dimensions of prenatal distress in

Diminished cerebral inhibition in neonates associated with risk factors for schizophrenia: parental psychosis, maternal depression, and nicotine use.

Diminished inhibitory gating of cerebral auditory evoked responses is transmitted in families with psychoses as an endophenotype related to the genetic risk for these illnesses. To assess whether the endophenotype is already expressed in infants of parents with psychotic illness and to assess effects of other known risk factors for schizophrenia, ie, maternal cigarette smoking and depression, inhibitory gating of cerebral auditory evoked responses was evaluated by comparing the P1 evoked responses to the first and second of paired auditory stimuli. Cerebral evoked responses were recorded durin

Coeliac disease is associated with intrauterine growth and neonatal infections.

UNLABELLED: To investigate whether factors in the fetal or neonatal period influence the risk of later development of coeliac disease we conducted a population-based register study. The Swedish Medical Birth Register was linked with the Hospital Discharge Register and identified 3392 singleton infants born in the period 1987-97 who developed coeliac disease. Perinatal data for these children were compared with all children born in these years. Exposure variables: Maternal age, parity and smoking habits in early pregnancy, preeclampsia, pregnancy duration and birthweight, birthweight by gestati

Hippocampal nitric oxide synthase in the fetal guinea pig: effects of chronic prenatal ethanol exposure.

The effects of chronic maternal administration of ethanol on nitric oxide synthase (NOS) activity and the numbers of NOS containing neurons, and CA1 and CA3 pyramidal neurons in the hippocampus of the near term fetal guinea pig at gestational day (GD) 62 were investigated. Pregnant guinea pigs received oral administration of 4 g ethanol/kg maternal body weight (n = 5), isocaloric sucrose/pair feeding (n = 5) or water (n = 5), or no treatment (NT; n = 5) from GD 2 to GD 61. NOS activity in the 25,000 x g supernatant of hippocampal homogenate was determined using a radiometric assay. The numbers

The First 1000 Days: Maternal Nutrient Intake-A Window of Opportunity for Pulmonary Hypertension-A Narrative Review.

The first 1000 days of life, starting from conception to a child's second birthday, constitute a pivotal period for fetal lung and pulmonary vascular development. Maternal nutrition during this period plays an important role in fetal growth, immune programming and organ development, including that of the pulmonary system. This narrative review consolidates evidence linking maternal nutrition and early-life nutrient intake during this period with the development of pulmonary hypertension in the newborn. We examine the influence of both nutrient deficiencies and excesses on fetal lung and vascul

Maternal Overweight and Obesity Alter Neurodevelopmental Trajectories During the First Year of Life: Findings from the OBESO Cohort.

Background/Objectives: Overweight and obesity during pregnancy are metabolic risk factors that may compromise offspring brain development. The first 1000 days of life represent a critical window in which neurodevelopmental trajectories are shaped by intrauterine and early-life exposures. The 6- and 12-month milestones are key checkpoints where deviations may emerge, and interventions are most effective. This study evaluated the association between maternal pregestational weight status and infant neurodevelopment at 6 and 12 months of age. Methods: Mother and infant pairs from the OBESO perinat

Parental age-related aneuploidy in human germ cells and offspring: a story of past and present.

Parental age is the most important aetiological factor in trisomy formation in humans. Cytogenetic studies on germ cells reviewed here imply that (i) 2-4% sperm are aneuploid and 8.6% oocytes from IVF are hyperploid (ii) a paternal age effect may exist, and (iii) oocytes of aged women contain precociously separated chromatids in metaphase II. Trisomy data suggest that most aneuploidy is generated during meiosis I of oogenesis and is maternal age-dependent. Trisomy 18 is unique, originating mostly from maternal meiosis II errors. The extra gonosome in 47, XXY derives mostly from a paternal meio

Impact of prenatal cocaine exposure on child behavior problems through school age.

OBJECTIVE: We examined the trajectory of childhood behavior problems after prenatal cocaine exposure. METHODS: The Maternal Lifestyle Study, a longitudinal cohort study, enrolled children between 1993 and 1995 at 4 centers. Prenatal cocaine exposure was determined from mothers who admitted use and/or meconium results. Exposed children were matched with a group of nonexposed children within site and by gestational age, gender, race, and ethnicity. The study began at the 1-month corrected age with a total of 1388 children enrolled. A total of 1056 were assessed for internalizing, externalizing,

Environmental exposure to tobacco smoke and lung function in young adults.

The relationship between lung function and environmental exposure to tobacco smoke (passive smoking) was studied in 293 nonsmoking young men and women, 15 to 35 yr of age. A self-administered mailed questionnaire was used to assess the lifetime environmental exposure to cigarette smoke at home and at work for each subject. Lung function information used here had been gathered in the course of a previous study of the determinants of lung function in early adulthood. In men, maximal midexpiratory flow rate (FEF25-75) decreased in relation to an index of cumulative lifetime environmental exposure

Markers of Kidney Function in Early Childhood and Association With Maternal Comorbidity.

IMPORTANCE: Kidney functional capacity is low at birth but doubles during the first 2 weeks of life and reaches near-adult levels at age 1 to 2 years. Existing reference intervals for markers of kidney function in newborns are mostly based on preterm newborns, newborns with illness, or small cohorts of term newborns, and the consequences of maternal comorbidities for newborn kidney function are sparsely described. OBJECTIVE: To establish robust reference intervals for creatinine and urea in healthy children in early childhood and to assess whether maternal comorbidity is associated with newbo

The early-life exposome and epigenetic age acceleration in children.

The early-life exposome influences future health and accelerated biological aging has been proposed as one of the underlying biological mechanisms. We investigated the association between more than 100 exposures assessed during pregnancy and in childhood (including indoor and outdoor air pollutants, built environment, green environments, tobacco smoking, lifestyle exposures, and biomarkers of chemical pollutants), and epigenetic age acceleration in 1,173 children aged 7 years old from the Human Early-Life Exposome project. Age acceleration was calculated based on Horvath's Skin and Blood clock
