Influence of cationic amphiphilic drugs on the phosphatidylcholine hydrolysis by phospholipase A2.

On chronic treatment certain amphiphilic drugs induce a generalized phospholipidosis. This drug side effect has been related to an inhibition of the lysosomal phospholipases due to the interaction of the drugs with phospholipids (PL). In the present experiments, the influence of the amphiphilic drugs ambroxol, imipramine, chloroquine and chlorphentermine on the hydrolysis of dipalmitoyl-phosphatidylcholine (DPPC) unilamellar liposomes by bee venom phospholipase A2 (PLase A2) was studied. Special emphasis was laid on the initial phase and temperature dependence. The activity of PLase A2 was mea…

Effects of phospholipases C on the beta-receptor-adenylate cyclase system of chick erythrocyte membranes.

The beta-adrenergic receptor located in chick erythrocyte membranes was characterized using (-)-[3H]-dihydroalprenolol ([3H]-DHA) with rapid … se activity, but not basal and forskolin-stimulated activities. These results suggest that membrane phospholipids, especially phosphatidylcholine (PC) and phosphatidylinositol (PI), are directly related to the receptor binding and that PI interacts with adenylate cyclase activity.

The phosphatidylcholine metabolism in the renal tissue during experimental acute and chronic pyelonephritis in the rabbit.

In 139 adult male rabbits unilateral pyelonephritis (PN) was induced. In a broad spectrum of enzymes and phospholipids also the content of phosphatidylcholine (PC) was determined. Only these latter values are reported here. PC showed markedly increased levels up to the 20-d-, and to the 31-d-series in the cortex. After 64 and 100 d of experimental PN the content of PC was less than the half of the normal values, and after 261 d of disease, it was only about 1/16 of the normal value (= 22.58 +/- 1.55 micrograms P/g wet weight). In the medulla the PC increased only till the 20-d-series, but show…

Effect of riboflavin deficiency on lipid metabolism of liver and brown adipose tissue of sucking rat pups.

An increase in liver: body-weight and in hepatic triacylglycerol content, together with changes in the fatty acid profiles of hepatic phospholipids, were observed as a result of moderate riboflavin deficiency in sucking rat pups. Oxygen consumption by hepatic mitochondria, with palmitoyl L-carnitine as substrate, was not significantly impaired. Mitochondria from interscapular brown adipose tissue, however, showed a marked impairment of O2 consumption, with palmitoyl L-carnitine as substrate, in the riboflavin-deficient pups. This impairment was also apparent after uncoupling with carbonyl cyan…

Specificity of purified monoacylglycerol lipase, palmitoyl-CoA hydrolase, palmitoyl-carnitine hydrolase, and nonspecific carboxylesterase from rat liver microsomes.

The comparative substrate specificities of five purified serine hydrolases from rat liver microsomes have been investigated, especially their action upon natural lipoids. All enzymes had high carboxylesterase activities with simple aliphatic and aromatic esters and thioesters. The broad pH optima were in the range of pH 6-10. Synthetic amides were less potent substrates. The hydrolytic activities towards palmitoyl-CoA and monoacyl glycerols were generally high, whereas phospholipids and palmitoyl carnitine were cleaved at moderate rates. Acetyl-CoA, acetyl carnitine, and ceramides were not cle…

In vitro and in vivo effects of ethanol on the formation of endoperoxide metabolites in rat platelets.

Preincubation of rat platelet-rich plasma (PRP) with ethanol resulted in dose-dependent inhibition of the formation of endoperoxide metaboli … that it can be reversed as soon as ethanol is eliminated. The level of arachidonic acid in platelet phospholipids of rats fed the ethanol diet was not different from that of the control, indicating that availability of immediate precursor acid would not be a factor for the enhanced synthesis of EPM in the ethanol group. This result suggested that platelets from …

Mechanism of lipid transfer by bridge-like protein VPS13A and the scramblase XK.

In eukaryotes, bridge-like lipid-transfer proteins (BLTPs) are central in mediating vesicle-independent lipid transfer between organelles. BLTPs span the cytosolic space between organelles at contact sites, featuring hydrophobic channels for lipids to travel between membranes. How BLTPs cooperate with partner proteins to orchestrate lipid delivery remains a mystery. Here, we used cryo-electron microscopy to visualize a complex comprising the prototypical BLTP VPS13A and the plasma membrane-localized scramblase XK at near-atomic resolution. VPS13A interacts with XK via its pleckstrin homology d

Curcumin protects against difenoconazole-induced kidney injury in carp by inhibiting ferroptosis through the Nrf2/GPX4 axis.

Difenoconazole (DFZ) is a persistent triazole fungicide that leads to aquatic toxicity and ecosystem damage due to its environmental persistence and high toxicity. Curcumin (CUR), a natural polyphenol derived from turmeric, exhibits strong anti-inflammatory, antioxidant, and antitumor properties. This study assesses CUR's protective role against DFZ-induced kidney injury in carp and investigates its mechanisms. Carp were exposed to DFZ for 30 days, with dietary supplementation of 400 mg/kg CUR. H&E staining revealed that CUR significantly reduced DFZ-induced histopathological damage in the kid

Oxidative Stress-Related Programmed Cell Death in Male Infertility: Focussing on Ferroptosis.

Oxidative stress is a common occurrence in testicular cells of infertile men and is considered a significant cause of male reproductive dysfunction. The increase in reactive oxygen species (ROS) leads to impaired spermatogenesis by activating caspases and generating free radicals from mitochondria, resulting in DNA damage and cell death through apoptosis. Additionally, oxidative stress in the testis triggers autophagy as an alternative form of programmed cell death in male germ cells. There is a complex interplay between autophagy and apoptosis in response to oxidative stress in male germ cell

Inhibition of Ferroptosis Delays Aging and Extends Healthspan Across Multiple Species.

Ferroptosis, a form of iron-dependent cell death, plays a pivotal role in age-related diseases; yet, its impact on cellular senescence and healthspan in mammals remains largely unexplored. This study identifies ferroptosis as a key regulator of cellular senescence, showing that its inhibition can significantly delay aging and extend healthspan across multiple species. During cellular senescence, ferroptosis is progressively exacerbated, marked by increased lipid peroxidation, oxidative stress, and diminished glutathione peroxidase 4 (GPX4) levels. Ferroptosis inducers such as Erastin and RSL3

Double-edged sword effect of GPX4 in skin homeostasis and diseases.

Glutathione peroxidase 4 (GPX4) is a crucial antioxidant enzyme that plays a vital role in protecting cells from oxidative damage and lipid peroxidation. In the context of skin diseases, GPX4 serves as a key regulator of oxidative stress and inflammation, both of which are significant features of various skin conditions. By preventing lipid peroxidation and maintaining membrane integrity, GPX4 acts as a safeguard against cell death pathways, particularly ferroptosis, in skin diseases. Dysregulation of GPX4 in conditions such as dermatitis, psoriasis, and skin cancer is linked to heightened oxi

Mammalian TMC1 or 2 are necessary for scramblase activity in auditory hair cells.

Sensory transduction in auditory hair cells gates mechanosensitive ion channels, converting sound information into electrical signals (Zheng and Holt, 2021). Previously, we found that Transmembrane channel (TMC) proteins 1 and 2 form the pore of hair cell transduction channels (Pan et al., 2013; 2018). The structure of C. elegans TMC proteins (Jeong et al., 2022; Clark et al., 2024) and predicted mammalian TMC structures (Hahn et al., 2009; Ballesteros et al., 2018; Pan et al., 2018) are reminiscent of TMEM16 proteins, which function as Ca2+-activated ion channels and lipid scramblases. Here,

Ferroptosis and renal fibrosis: mechanistic insights and emerging therapeutic targets.

Ferroptosis is a regulated, iron-dependent form of cell death driven by lipid peroxidation and distinct from apoptosis, necroptosis, and pyroptosis. Recent studies implicate ferroptosis as a central contributor to the pathogenesis of renal fibrosis, a hallmark of chronic kidney disease associated with high morbidity and progression to end-stage renal failure. This review synthesizes current evidence linking ferroptotic signaling to fibrotic remodeling in the kidney, focusing on iron metabolism dysregulation, glutathione peroxidase 4 (GPX4) inactivation, lipid peroxide accumulation, and ferropt

Investigation Into the Inhibition of Ferroptosis by Praeruptorin-A in Sepsis Based on Network Pharmacology.

Praeruptorin A (PA), a monomer derived from the traditional Chinese medicine Peucedani Radix, is known for its therapeutic properties, including heat clearance, phlegm dissolution, and asthma relief. Sepsis, characterized as a systemic inflammatory response syndrome (SIRS), is triggered by a cytokine storm resulting from pathogenic infection and can progress to multi-organ failure. This study investigates and predicts the effective molecular targets and potential mechanisms of PA in the context of sepsis through the application of network pharmacology. The identified targets were subsequently

Targeting Ferroptosis in Tumors: Novel Marine-Derived Compounds as Regulators of Lipid Peroxidation and GPX4 Signaling.

This article reviews the mechanisms by which marine natural products regulate ferroptosis and their potential applications in tumor therapy. Ferroptosis is a form of programmed cell death driven by iron-dependent lipid peroxidation, characterized primarily by the accumulation of lipid peroxides and the failure of antioxidant defense systems. Due to their unique chemical structural diversity, marine natural products demonstrate significant advantages in regulating the ferroptosis pathway. Studies showed that marine compounds target key molecules such as glutathione peroxidase 4 (GPX4) and long-
