A rapid and simple assay for cytopathogenicity of axenically cultivated Entamoeba histolytica has been developed employing baby hamster kidney (BHK) or mouse 3T3 cells conventionally tissue cultured. Three of the twelve amebal strains tested produced total destruction of the BHK cell monolayer in 2--3 hours, and these three strains are the three most virulent strains for the newborn hamster liver. In order to study the role of calmodulin (CaM) in the cell cycle, RC 3 cells carring the CaM expression vectors which was constructed by joining the CaM cDNA with a plasmid of mouse mammary tumor virus (MMTV), were used in this experiment. The lectin isolated from Xerocomus chrysenteron (XCL) displays a toxic activity towards insects. In order to assess its possible mode of action and to gather useful data for its potential use in insect-resistant transgenic plants, we investigated the effects of XCL at the cellular level. INTRODUCTION: Obesity contributes to poorer clinical outcomes in patients with acute lymphoblastic leukemia. We have shown that adipocytes cause anthracycline resistance by absorbing and metabolizing them into less-active alcohol metabolites.
METHODS: We hypothesized that mitoxantrone, which has a similar cytotoxic mechanism to anthracyclines but is metabolized through different pathways, might overcome this adipocyte-mediated chemoresistance. We treated human BV173 acute lymphoblastic leukemia cells with daunorubicin and mitoxantrone that had been incubated with or without 3T3-L1 adipocytes.

Gene regulation during in vitro differentiation into adipocytes was examined in rat dental pulp-derived cells. Insulin, 3-isobutyl-1-methylxanthine, and dexamethasone were added to induce adipogenesis. Cells containing lipid droplets were observed after induction as in 3T3 L1 cells. The seminiferous tubules of rat testes contain a protease and heat sensitive factor capable of stimulating (3H)-thymidine incorporation into quiescent cultures of NIH 3T3 mouse fibroblast cells. This mitogenic factor activity was only 4% in the testes of vitamin A deficient-retinoic acid maintained rats as compared to that of normal rats.
