A scalable Bayesian functional GWAS method accounting for multivariate quantitative functional annotations with applications for studying Alzheimer disease.

Existing methods for integrating functional annotations in genome-wide association studies (GWASs) to fine-map and prioritize potential causal variants are limited to using non-overlapped categorical annotations or limited by the computation burden of modeling genome-wide variants. To overcome these limitations, we propose a scalable Bayesian functional GWAS method to account for multivariate quantitative functional annotations (BFGWAS_QUANT), accompanied by a scalable computation algorithm enabling joint modeling of genome-wide variants. Simulation studies validated the performance of BFGWAS_

Simulating degradation of organic compounds accounting for the growth of microorganisms (Monod kinetics) in a fully Lagrangian framework.

Biologically mediated degradation of organic compounds in porous media is a complex mathematical problem, described by a non-linear differential equation. The organic compound gets in contact with the biomass, and an enzyme-catalysed reaction takes place. The net result is that part of the parent compound degrades into some daughter product, while some of the organic carbon is used for microbial growth. The rate of biomass growth in the presence of a limiting nutrient supply is usually modelled with the experimentally derived Monod equation, i.e., it is proportional to the actual existing biom

Accounting for health literacy and intervention preferences when reducing unhealthy snacking: protocol for an online randomised controlled trial.

INTRODUCTION: Health literacy describes the cognitive and social skills that individuals use to access, understand and act on health information. Health literacy interventions typically take the 'universal precautions approach' where all consumers are presented with simplified materials. Although this approach can improve knowledge and comprehension, its impact on complex behaviours is less clear. Systematic reviews also suggest that health literacy interventions underuse volitional strategies (such as planning) that play an important role in behaviour change. A recent study found volitional s

Accounting for Variance in Concussion Tolerance Between Individuals: Comparing Head Accelerations Between Concussed and Physically Matched Control Subjects.

Researchers have been collecting head impact data from instrumented football players to characterize the biomechanics of concussion for the past 15 years, yet the link between biomechanical input and clinical outcome is still not well understood. We have previously shown that even though concussive biomechanics might be unremarkable in large datasets of head impacts, the impacts causing injury are of high magnitude for the concussed individuals relative to their impact history. This finding suggests a need to account for differences in tolerance at the individual level. In this study, we ident

Accounting for Taste: A Multi-Attribute Neurocomputational Model Explains the Neural Dynamics of Choices for Self and Others.

How do we make choices for others with different preferences from our own? Although neuroimaging studies implicate similar circuits in representing preferences for oneself and others, some models propose that additional corrective mechanisms come online when choices for others diverge from one's own preferences. Here we used event-related potentials (ERPs) in humans, in combination with computational modeling, to examine how social information is integrated in the time leading up to choices for oneself and others. Hungry male and female participants with unrestricted diets selected foods for t

Psychological inflexibility predicts PTSD symptom severity in war veterans after accounting for established PTSD risk factors and personality.

OBJECTIVE AND METHOD: Numerous risk factors for posttraumatic stress disorder (PTSD) have been identified; however, many do not inform treatment. Psychological inflexibility is a modifiable factor that can be targeted in psychological treatment. This study examined whether higher levels of psychological inflexibility predicted unique variance in PTSD symptom severity at 1-year follow-up in 236 U.S. veterans of the wars in Iraq in Afghanistan after accounting for the strongest known risk factors for PTSD. PTSD symptom severity was assessed using the Clinician Administered PTSD Scale. RESULTS: …

Modeling single-vehicle run-off-road crash severity in rural areas: Accounting for unobserved heterogeneity and age difference.

This study investigates factors that significantly contribute to the severity of driver injuries resulting from single-vehicle run-off-road (SV ROR) crashes. A mixed logit model approach is employed to explore the potential unobserved heterogeneous effects associated with each age group: young (ages 16-24), middle-aged (ages 25-65), and older drivers (ages over 65). Likelihood ratio tests indicated that the development of separate injury severity models for each age group is statistically superior to estimating a single model using all data. Based on the crash data collected from 2009 to 2013

Using a modification of the Clavien-Dindo system accounting for readmissions and multiple interventions: defining quality for pancreaticoduodenectomy.

BACKGROUND: The Clavien-Dindo system (CD) does not change the grade assigned a complication when multiple readmissions or interventions are required to manage a complication. We apply a modification of CD accounting for readmissions and interventions to pancreaticoduodenectomy (PD). METHODS: PDs done between 1999 and 2009 were reviewed. CD grade IIIa complications requiring more than one intervention and II and IIIa complications requiring significantly prolonged lengths of stay including all 90-day readmissions were classified severe-adverse-postoperative-outcomes (SAPO). CD IIIb, IV, and V …

Single-nucleotide polymorphisms, acute rejection, and severity of tubulitis in kidney transplantation, accounting for center-to-center variation.

BACKGROUND: Acute rejection (AR) is associated with worse renal allograft outcomes. Therefore, this study investigated single-nucleotide polymorphisms (SNPs) to identify genetic variants associated with AR, accounting for center variation, in a multicenter, prospective, observation study. METHODS: We enrolled patients from six transplant centers, five in the United States and one in Canada. A total of 2724 SNPs were genotyped. We accounted for center variation in AR rates by stratifying by transplant center and using novel knowledge discovery methods. RESULTS: There was significant center va…

Explicitly accounting for antiretroviral drug uptake in theoretical HIV models predicts long-term failure of protease-only therapy.

Mathematical models of HIV therapy have traditionally amalgamated the action of antiretroviral drugs, trading the complexity of the situation in favour of simpler-and hence mathematically tractable-models. However, the effects of ignoring such dynamics remain underexamined. In this paper, the traditional method of dosing (where the dose is modelled implicitly as a proportional inhibition of viral infection and production) is compared to a model that accounts for drug dynamics via explicit compartments. Four limiting cases are examined: frequent dosing of both major classes of drugs, absence of

Mapping the Burden of Ischemic Stroke Attributable to Temperature Variation under Climate Change Scenarios Accounting for Long-Term Adaptation.

With the intensification of climate change, temperature variability (TV), a critical climatic dimension, is projected to increase. As a climate-sensitive disease, ischemic stroke may be affected by temperature variability; however, the evidence on the ischemic stroke burdens of short-term temperature variability in a changing climate remains unknown, especially accounting for long-term exposure adaptation. Based on a nationally representative, multicenter data set of over 1.6 million individual-level, hourly data on ischemic stroke onset from 2015 to 2022, we adopted a case-crossover study des…

Association between total cholesterol and depression or depressive symptoms: A systematic review and meta-analysis accounting for comparator comorbidity.

BACKGROUND: The association between total cholesterol (TC) and depression remains uncertain because study populations, depression ascertainment, comparator groups, metabolic comorbidity, and study precision vary substantially. We conducted a systematic review and meta-analysis to quantify group differences in TC and to evaluate the influence of comparator comorbidity, study characteristics, and small-study effects. METHODS: PubMed, Embase, Web of Science, and the Cochrane Library were searched from inception to January 9, 2026. Observational studies reporting TC in participants with depressio

Associations of Cognitive Impairment and Physical Prefrailty with Occupational Dysfunction in Community-Dwelling Older Adults: Accounting for Depressive Tendencies.

This cross-sectional study examined the associations of cognitive impairment and physical prefrailty (PPF) with occupational dysfunction among community-dwelling older adults, adjusting for depressive tendencies. Data from 105 participants were analyzed using two-way ANCOVA. No interaction effects were observed. PPF showed a significant main effect on occupational imbalance. When the Geriatric Depression Scale score was excluded as a covariate, cognitive prefrailty (CPF) was associated with higher occupational deprivation. These findings suggest that depressive tendencies may confound the asso

Synaptic markers are associated with cognitive decline after accounting for amyloid burden among an at-risk Alzheimer's disease cohort.

Amyloid burden impacts cognitive decline in the pre-dementia stages of Alzheimer's disease (AD), but there remains significant variability in cognitive trajectories that may be explainable by markers of synaptic function and neurodegeneration. Leveraging longitudinal data from two harmonized, at-risk but predominantly unimpaired cohorts, we examined how several proteins measured from cerebrospinal fluid (CSF) differ by amyloid status, cognitive status, and impact cognitive decline measured using a Preclinical Alzheimer's Cognitive Composite. Four hundred and thirty-four individuals provided CS
