SIGNIFICANCE STATEMENT: Gastric vagal afferent responses to tension are dampened in high fat diet-induced obesity. Endocannabinoids are known to dose-dependently inhibit and excite gastric vagal afferents but their effect on gastric vagal afferents in diet-induced obesity are unknown. In individual gastric vagal afferent neurons of diet-induced obese mice the co-expression of components of the endocannabinoid system, including CB1, GHSR, TRPV1 and FAAH, was increased compared with lean mice. In high fat diet-induced obese mice, methanandamide only inhibited gastric vagal afferent responses to

Several lines of evidence suggest that sensory nerves of the carotid body have an efferent function in addition to their afferent function of conducting chemoreceptive impulses to the brain. However, it has been difficult to document the release of substances from sensory nerve terminals on glomus cells and to determine whether such an efferent function plays a role in chemoreception. By comparison, the phenomenon of neurogenic inflammation has been relatively easy to study in rats and guinea pigs and has proven to be an informative model system for analyzing efferent actions of sensory nerves

Micturition is coordinated at the level of the spinal cord and the brainstem. Spinal cord injury therefore directly interrupts spinal neuronal pathways to the brainstem and results in bladder areflexia. Some time after injury, however, dyssynergic bladder and sphincter function emerges. The changes mediating the appearance of bladder function after spinal cord injury are currently unknown. Primary afferent neurons have been shown to sprout in response to spinal cord injury. Sprouting primary afferents have been linked to the pathophysiology of centrally manifested disorders, such as autonomic

Two types of vagal evoked potentials were recorded in chloralose-anesthetized rats: a positive-negative one and a negative-positive one in granular and agranular insular cortical subareas, respectively, the former being more susceptible to barbiturate. Both potentials consisted of two subtypes which were distinct as to threshold and latency. Peripheral conduction velocities as well as thresholds of these two subtypes corresponded to myelinated and non-myelinated primary afferents, respectively. Thus, these two primary vagal afferents are probably both represented in two insular cortical subare

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease that progressively disrupts voluntary motor command through combined cortical and spinal motor neuron degeneration. However, how spinal circuits reorganize during the disease remains poorly understood, particularly in humans. This study examined the function of excitatory and inhibitory spinal interneuron circuits that control upper and lower limb movements, using non-invasive electrophysiological techniques targeting specific afferent-motoneuron pathways at cervical and lumbar levels. These segments are clinically relevant, as

Cough is comprised of three phases (inspiratory, compressive and expiratory) and serves as a vital defensive mechanism for lung health. It p … ed airway afferents/receptors to cough are still disputed. Plasticity of the peripheral and central afferent pathways in cough has recently been shown to be important in pathological states associated with increased cough. To date, little is known of the developmental aspects of cough.

Postherpetic neuralgia is an unfortunate aftermath of shingles, and is most likely to develop, and most persistent, in elderly patients. Pain, allodynia, and sensory loss in the affected dermatome are the cardinal manifestations of the disorder. The pathophysiology of postherpetic neuralgia is not well known, but recent observations suggest multiple changes in the afferent pathways at both peripheral and central nervous system levels.

PURPOSE OF REVIEW: To review the mechanisms for the perception of dyspnea and to consider the plausibility of interventions that palliate dyspnea after optimal treatment of the underlying disease.
RECENT FINDINGS: Activation of sensory receptors by blood gas abnormalities, mechanical respiratory loads, and hyperinflation provides afferent information to the central nervous system for integration and processing. It has been proposed that a discriminative pathway processes afferent impulses to the somatosensory cortex that reflects the intensity of dyspnea, whereas an affective pathway projects

The projections of coarse and fine axons of the glossopharyngeal (IX) nerve upon the caudal two thirds of the nucleus of the tractus solitarius (NTS) were studied in the cat. These afferents convey the chemo- and baroreceptor activities from the carotid receptors. We applied the Fink-Heimer method on brainstem sections, at different survival times, after a petrosal ganglionectomy. A segregation of fine and coarse fibered components was observed. Degeneration of coarse axons was mostly found in the lateral NTS, while fine fiber degeneration was predominant in regions of the medial and commissur

BACKGROUND: Ghrelin and anandamide (AEA) can regulate the sensitivity of gastric vagal afferents to stretch, an effect mediated via the transient receptor potential vanilloid 1 (TPRV1) channel. High fat diet (HFD)-induced obesity alters the modulatory effects of ghrelin and AEA on gastric vagal afferent sensitivity. This may be a result of altered gastric levels of these hormones and subsequent changes in the expression of their receptors. Therefore, the current study aimed to determine the effects of ghrelin and AEA on vagal afferent cell body mRNA content of cannabinoid 1 receptor (CB1), ghr

Neural plasticity occurs within the central and peripheral nervous systems after spinal cord injury (SCI). Although central alterations have extensively been studied, it is largely unknown whether afferent and efferent fibers in pelvic viscera undergo similar morphological changes. Using a rat spinal cord transection model, we conducted immunohistochemistry to investigate afferent and efferent innervations to the kidney, colon, and bladder. Approximately 3-4 weeks after injury, immunostaining demonstrated that tyrosine hydroxylase (TH)-labeled postganglionic sympathetic fibers and calcitonin g

The cerebellum is involved in some forms of motor coordination and learning, and in cognitive and emotional functions. To elucidate the functions of the cerebellum, it is important to unravel the detailed connections of the cerebellar neurons. Although the cerebellar neural circuit structure is generally conserved among vertebrates, it is not clear whether the cerebellum receives and processes the same or similar information in different vertebrate species. Here, we performed monosynaptic retrograde tracing with recombinant rabies viruses (RV) to identify the afferent connections of the zebraf

Cranial sensory placodes derive from discrete patches of the head ectoderm and give rise to numerous sensory structures. During gastrulation, a specialized "neural border zone" forms around the neural plate in response to interactions between the neural and nonneural ectoderm and signals from adjacent mesodermal and/or endodermal tissues. This zone subsequently gives rise to two distinct precursor populations of the peripheral nervous system: the neural crest and the preplacodal ectoderm (PPE). The PPE is a common field from which all cranial sensory placodes arise (adenohypophyseal, olfactory

Somatosensory evoked potentials (SEPs) are widely used in the clinic as well as research to study the functional integrity of the different parts of sensory pathways. However, most general anesthetics, such as isoflurane, are known to suppress SEPs, which might affect the interpretation of the signals. In animal studies, the usage of anesthetics during SEP measurements is inevitable due to which detailed effect of these drugs on the recordings should be known. In this paper, the effect of isoflurane on SEPs was studied in a rat model. Both time and frequency properties of the cortical recordin

The origin of sensory nerves that innervate the submandibular salivary gland was investigated in the rat. After application of wheat germ agglutinin-horseradish peroxidase to the cut endings of the sympathetic and parasympathetic nerve branches at the hilus of the gland, labeled cells were mainly found in the dorsal root ganglia and the trigeminal ganglion, respectively. The labeled neurons in these ganglia were of various sizes compared to unlabeled neurons, suggesting that the sensory nerves of the gland conduct various modalities of sensory information.

Cerebral cortical neural networks associated with eyelid movement play a critical role in facial animation, contribute to the regulation of blink frequency, and help prevent ocular injury. Eyelid closure depends, in part, on motoneurons that innervate the orbicularis oculi (OO) muscles. In this study, OO motoneuron cortical afferents were identified in rhesus monkeys with rabies virus, a retrograde transneuronal tracer. Virus was injected into the right OO muscle and immunohistochemically localized after 4-6 day transport intervals. Labeled motoneurons were limited to dorsal portions of the ip

Until now, most studies concerning active head movements in three dimensions have used the classical rotation vector description. Although this description yields both the orientation of the head rotation axis and the amount of rotation, it is incomplete because it cannot specify the location of this rotation axis in space. The latter is of importance for a proper picture of the vestibular consequences of active head movements and has relevance for the problem of how the brain deals with the inherent kinematic redundancy of the multijoint head-neck system. With this in mind, we have extended t

Horizontal intracortical connections may form one substrate for representational plasticity in somatosensory cortex. Electrophysiological mapping demonstrated the finer details of the representations of the hand, lower jaw, neck, and face in area 3b of normal macaque monkeys. Injections of two fluorescent tracers then defined the extent to which horizontal connections crossed from the face into the hand representations and vice versa in area 3b. Connections are widely distributed within cortical representations of skin areas innervated by cervical nerves or by the trigeminal nerve but do not c

This paper reports a theoretical analysis of the transformation from a tactile stimulus of the face to climbing fiber responses in three regions of the cat cerebellum. The database consisted of climbing fiber receptive fields on the face from 75 responses from the anterior lobe, 33 responses from the paramedian lobule (PML), and 52 responses from the crus IIp of the anesthetized cat. The receptive fields were similar in being composed of discrete areas on the skin, or skin compartments. The regional differences in the configurations of the receptive fields were reflected in which compartments

The direct evaluation of the autonomic nervous system to detect the neurogenic component of erectile dysfunction in men is not possible. Biothesiometry, a vibration perception test, is a simple method for the assessment of the penile somato-afferent system. A nomogram for the evaluation of somato-afferent system has been developed utilizing the biothesiometer in healthy men. This nomogram was used to compare the biothesiometric values of potent and impotent diabetic patients. According to our data, the radix penis is selectively damaged by diabetes mellitus in the first stage of the disease, w
