[Relationship of single nucleotide polymorphism, environmental factors and the hereditary susceptibility of hepatocellular carcinoma].

Resulting by the interaction between the hereditary factor and the environmental factor, the occurrence and development of hepatocellular carcinoma (HCC) is a complicated course with multi-genes and multi-factors. As the third genetic marker, the single nucleotide polymorphism reflects the hereditary difference among individuals, decides the susceptibility to disease and becomes an important tool to study the hereditary susceptibility of HCC.

Concentration and prevalence of mycotoxins in poultry feed and poultry products: a global systematic review and meta-analysis.

Mycotoxins are secondary metabolites produced by fungi that commonly contaminate animal feed, especially poultry feed, impacting poultry productivity and causing adverse effects on animal health. Ochratoxin A, deoxynivalenol, fumonisins, aflatoxins, zearalenone, and trichothecene are of particular concern due to their high toxicity, prevalence, and potential to accumulate as residues in animal-derived foods, such as offal, muscle tissues, and eggs. This study aims to conduct a systematic review and meta-analysis of the occurrence, prevalence, and co-occurrence of mycotoxins in poultry feed and

In vivo detection of aflatoxin-induced lipid free radicals in rat bile.

Aflatoxin B1 (AFB1), a potent hepatotoxin and hepatocarcinogen, is metabolized in the liver via cytochrome P-450 to an AFB1-8,9-epoxide intermediate. The formation of the AFB1-8,9-epoxide correlates with the pathological changes observed in numerous mammalian species. Oxidative damage has been postulated to play a major role in the mechanisms associated with AFB1-induced cytotoxicity and carcinogenecity in mammalian species. The aim of this study was to detect and identify free radical intermediates from the hepatic metabolism of AFB1 in vivo. Rat bile ducts were cannulated and rats were treat…

Mapping Global Research Trends on Aflatoxin M1 in Dairy Products: An Integrative Review of Prevalence, Toxicology, and Control Approaches.

Aflatoxin M1 (AFM1), a hydroxylated metabolite of aflatoxin B1 (AFB1), is a potent hepatotoxic and carcinogenic compound frequently detected in milk and dairy products. Its thermal stability and resistance to processing make it a persistent public health concern, especially in regions prone to fungal contamination of animal feed. This review integrates bibliometric mapping (2015-2025) with toxicological and mitigation perspectives to provide a comprehensive understanding of AFM1. The bibliometric analysis reveals a sharp global rise in research output over the last decade, with Iran, China, an…

Aflatoxin exposure is associated with an increased risk of gallbladder cancer.

Gall bladder cancer (GBC) is common among the socioeconomically deprived populations of certain geographical regions. Aflatoxin is a genotoxic hepatocarcinogen, which is recognized to have a role in the pathogenesis of hepatocellular carcinoma. However, the role of aflatoxin in the pathogenesis of GBC is largely unknown. We determined serum AFB1-Lys albumin adduct (AAA) levels as a marker of aflatoxin exposure in the patients with GBC and compared to those without GBC. The relationship of AAA levels to cytogenetic (TP53mutation&HER2/neu amplification) and radiological characteristics of the tu

CYP2A6: a human coumarin 7-hydroxylase.

Coumarin 7-hydroxylation is catalysed by a high-affinity CYP2A6 enzyme in human liver microsomes. CYP2A6 is the only enzyme catalysing this reaction and consequently the formation of 7-hydroxycoumarin can be used as 'an in vitro and in vivo probe' for CYP2A6. CYP2A6 is a major contributor to the oxidative metabolism of nicotine and cotinine, and it also contributes, to a larger or smaller extent, to the metabolism of a few pharmaceuticals (e.g. fadrozole), nitrosamines, other carcinogens (e.g. aflatoxin B1) and a number of coumarin-type alkaloids. CYP2A6 may be inducible by antiepileptic drugs…

Valorization of Flammulina velutipes residue into a feed additive via co-fermentation to control Escherichia coli.

Flammulina velutipes residue (FVR) was valorized into a functional feed additive targeting multi-drug resistant (MDR) Escherichia coli B2 (E. coli B2) via multi-strain co-fermentation. The process, optimized with Lactiplantibacillus plantarum Z-9, concurrently enhanced the nutritional value of FVR by increasing crude protein (83.2 %) and ensured feed safety by degrading aflatoxin B1 (96 %). Crucially, fermentation enriched antimicrobial metabolites, including phenyllactic acid (+2.3-fold), which demonstrated direct inhibition against MDR E. coli. Furthermore, the process effectively reshaped t…

Environmental Influences on the Epigenome: Exposure- Associated DNA Methylation in Human Populations.

DNA methylation is the most well studied of the epigenetic regulators in relation to environmental exposures. To date, numerous studies have detailed the manner by which DNA methylation is influenced by the environment, resulting in altered global and gene-specific DNA methylation. These studies have focused on prenatal, early-life, and adult exposure scenarios. The present review summarizes currently available literature that demonstrates a relationship between DNA methylation and environmental exposures. It includes studies on aflatoxin B1, air pollution, arsenic, bisphenol A, cadmium, chrom…

Mycotoxin-Caused Intestinal Toxicity: Underlying Molecular Mechanisms and Further Directions.

Mycotoxins represent a group of highly toxic secondary metabolites produced by diverse fungal pathogens. Mycotoxin contaminations frequently occur in foods and feed and pose significant risks to human and animal health due to their carcinogenic, mutagenic, and immunosuppressive properties. Notably, deoxynivalenol, zearalenone, fumonisins (mainly including fumonisins B1, B2, and FB3), aflatoxin B1 (AFB1), and T-2/HT-2 toxins are the major mycotoxin contaminants in foods and feed. Undoubtedly, exposure to these mycotoxins can disrupt gut health, particularly damaging the intestinal epithelium in…

Functional characterization of single nucleotide polymorphic variants of DNA repair enzyme NEIL1 in South Asian populations.

The base excision repair (BER) pathway is a precise and versatile mechanism of DNA repair that is initiated by DNA glycosylases. Endonuclease VIII-like 1 (NEIL1) is a bifunctional glycosylase/abasic site (AP) lyase that excises a damaged base and subsequently cleaves the phosphodiester backbone. NEIL1 is able to recognize and hydrolyze a broad range of oxidatively-induced base lesions and substituted ring-fragmented guanines, including aflatoxin-induced 8,9-dihydro-8-(2,6-diamino-4-oxo-3,4-dihydropyrimid-5-yl-formamido)-9-hydroxyaflatoxin B1 (AFB1-FapyGua). Due to NEIL1's protective role again…

Aflatoxins as risk factors for hepatocellular carcinoma in humans.

On a global basis, primary liver cancer (PLC) is a very prevalent form of cancer. Wide variation of PLC incidence in different areas of the world suggests the involvement of environmental factors in its etiology. Two major classes of risk factors have been identified. Extensive evidence indicates the importance of infection by the hepatitis B virus as a major risk factor for PLC. Because many organic chemicals induce liver cancer in experimental animals, those to which human exposure is known to occur are also of interest with respect to their possible involvement as risk factors for PLC. Multiomics Integration Reveals AFB1 Causes Liver Damage Involving the Gut-Microbiota-Lipid Metabolism Axis in Piglet.

Prolonged exposure to aflatoxin B1 (AFB1) poses a significant threat to livestock production. The liver is the main target, but the role of the gut-liver axis and lipid metabolism in pig hepatic toxicity is not well understood. This study evaluates the impact of AFB1 on piglet liver injury via the gut-liver axis using multiomics analysis. Chronic AFB1 exposure significantly impaired the piglet growth and induced liver injury. Meanwhile, AFB1 caused gut microbiota dysbiosis and intestinal barrier damage in the piglets. Fecal microbiota transplantation (FMT) demonstrated that AFB1-altered microb…

AFB1 exacerbates testicular and intestinal inflammation by increasing stearoyl ethanolamide and homocysteine levels.

In environmental toxicology, aflatoxin B1 (AFB1) is recognized for its detrimental effects on reproductive and intestinal health. This study elucidates how AFB1-induced elevations in stearoyl ethanolamide (SEA) and homocysteine (HCY) impact male fertility and intestinal function in mice. AFB1 was found to markedly reduce sperm concentration and exacerbate sperm damage in mice, primarily by increasing serum SEA and HCY levels. These metabolites compromise testicular structure and function, disrupt the blood-testicular barrier, and downregulate critical testicular proteins including DAZL, SYCP1,…
