Tolerance and efficacy of combined diethylcarbamazine and albendazole for treatment of Wuchereria bancrofti and intestinal helminth infections in Haitian children.

This randomized, placebo-controlled trial investigated the tolerance, efficacy, and nutritional benefit of combining chemotherapeutic treatment of intestinal helminths and lymphatic filariasis. Children were infected with Ascaris (30.7%), Trichuris (53.4%), and hookworm (9.7%) with 69.9% having more than one of these parasites. A total of 15.8% of the children had Wuchereria bancrofti microfilariae. Children were randomly assigned treatment with placebo, albendazole (ALB), diethylcarbamazine (DEC), or combined therapy. The combination of DEC/ALB reduced microfilarial density compared with plac…

Pharmacology and efficacy of liposome-entrapped albendazole in experimental secondary alveolar echinococcosis and effect of co-administration with cimetidine.

Encapsulation of the benzimidazole albendazole in multilamellar liposomes results in a preparation in which this normally insoluble anti-hydatid drug is well solublilized in aqueous media. The high entrapment efficiency observed (75-87%) and the stability of the formulation make this a promising delivery vehicle for improved chemotherapy with albendazole. In particular, the high degree of association with phospholipid may give rise to increased oral bioavailability. Oral administration of albendazole in liposomes led to increased concentration and/or altered metabolism of albendazole sulphoxid…

Intraperitoneal and intracranial experimental cysticercosis present different metabolic preferences after treatment with isolated or combined albendazole and nitazoxanide.

Cysticercosis is a zoonotic public health issue especially severe when the parasite is in the central nervous system although it may be found all over the human organism. Taenia crassiceps cysticerci inoculated in mice is the experimental model used to study cysticercosis. The most used cysticercosis treatment is with albendazole (ABZ). Nitazoxanide (NTZ) has been experimentally tested against this parasite. Metabolic analysis has been used to determine drugs impact on the parasite. The aim of this study was to determine the in vivo metabolic impact of the ABZ-NTZ combination in T. crassiceps …

Three day albendazole therapy in patients with a solitary cysticercus granuloma: a randomized double blind placebo controlled study.

A solitary cysticercus granuloma is a benign form of neurocysticercosis. Several reports have suggested albendazole is effective in early resolution of these lesions. A short duration (7 days) albendazole therapy has been found to be effective in treatment. In this study, we evaluated a "three day course" of albendazole in a prospective randomized double- blind trial. Sixty-seven consecutive patients who presented with new-onset seizures and a solitary cysticercus granuloma were randomly allocated to receive either albendazole (15 mg/kg/day for 3 days) or placebo. All patients were treated wit…

Albendazole therapy in alveolar hydatid disease: a report of favorable results in two patients after short-term therapy.

Albendazole was administered preoperatively to two patients with active alveolar hydatid disease for 58 and 84 days. Vesicles of larval Echinococcus multilocularis obtained from surgical tissues were inoculated into red-backed voles for in vivo testing viability. No proliferation of the larval cestode had occurred when the animals were dissected three months post-inoculation. These findings suggest that short-term therapy with albendazole was effective in killing the larval cestode in these two cases. Albendazole was found to be hepatotoxic but resulting transaminase abnormalities have been re…

Amiodarone promoted autophagy and enhanced tissue repair, outperforming albendazole in treating the muscular phase of experimental trichinellosis spiralis.

Targeting autophagy as a therapeutic approach against infections has gained significant attention nowadays. Amiodarone (AMD), the antiarrhythmic drug with autophagy-stimulating property, was used in this study, compared to Albendazole (ALB) (the commonly used drug) against experimental trichinellosis. Our in vivo study used two assessment tools for evaluating the autophagy process: quantitative analysis of local microtubule-associated protein 1 light chain 3 (LC3) autophagy protein marker, in addition to detecting autophagic compartments by transmission electron microscopy (TEM). AMD showed th…
