BACKGROUND: Although not every food protein contributes sufficient quantities of all of the indispensable amino acids (AAs) required for human health, the human diet is a mixture of foods. It is therefore important to evaluate protein quality in mixed meals.
OBJECTIVES: The hypothesis was that the true ileal digestible AA content of mixed meals can be predicted from the amounts of digestible AA determined for individual food ingredients, allowing further prediction of the digestible indispensable amino acid score (DIAAS) for meals.
METHODS: Four diets contained either pea protein isolate (PP

BACKGROUND: Sarcopenia is thought to be underlined by age-associated anabolic resistance and dysregulation of intracellular signalling pathways. However, it is unclear whether these phenomena are driven by ageing per se or other confounding factors.
METHODS: Lean and healthy young (n = 10, 22 ± 3 years, BMI; 23.4 ± 0.8 kg/m2) and old men (n = 10, 70 ± 3 years, BMI; 22.7 ± 1.3 kg/m2) performed unilateral resistance exercise followed by intake of essential amino acids (EAA). Muscle biopsies were collected from the rested and the exercised leg before, immediately after and 60 and 180 min after E

Effects of certain groups of nutrients such as glucose, essential amino acids (AA), nonessential AA, vitamins and trace nutrients on the multiplication of various strains of Japanese encephalitis virus (JEV) were studied with an aim to optimise the conditions for cultivation of the virus in porcine stable (PS) kidney cell cultures. Eagle's Minimal Essential Medium (MEM) was modified by addition of the nutrients in different concentrations and combinations. Glucose was found the most important single nutrient in promoting significantly the virus multiplication. Essential AA alone did not influe

In vitro embryo production (IVP) is widely used in cattle breeding, however, pregnancy outcomes remain suboptimal, primarily due to variability in oocyte and embryo quality. This study evaluated the association of donor animal category with amino acid turnover in bovine oocytes and embryos and assessed its relationship with developmental competence. Cumulus-oocyte complexes (COCs) were collected via ovum pick-up from prepubertal heifers (8.4 ± 0.5 mo of age; n = 9), pubertal heifers (16.5 ± 0.5 mo of age; n = 10), and cows (25.5 ± 0.5 mo of age; n = 10). A total of 198, 324, and 286 COCs, resp

Antibiotics alter the gut microbiota and decrease resistance to Clostridium difficile colonization; however, the mechanisms driving colonization resistance are not well understood. Loss of resistance to C. difficile colonization due to antibiotic treatment is associated with alterations in the gut metabolome, specifically, with increases in levels of nutrients that C. difficile can utilize for growth in vitro. To define the nutrients that C. difficile requires for colonization and pathogenesis in vivo, we used a combination of mass spectrometry and RNA sequencing (RNA Seq) to model the gut met

UNLABELLED: The toxicity of the amino acids glycine and L-serine at high concentrations in bacteria was discovered decades ago. In this work, we used deep transposon insertion sequencing (Tn-seq) experiments to determine the genes necessary to tolerate excess L-serine, diglycine or glycine in the human pathogen Staphylococcus aureus . Our results indicate that intracellular accumulation of specific counterbalancing amino acids-such as alanine in excess glycine-is the primary mechanism of resistance to amino acid toxicity. Consistent with this model, specific amino acid and peptide uptake trans

Skeletal muscle is an integral body tissue playing key roles in strength, performance, physical function, and metabolic regulation. It is essential for athletes to ensure that they have optimal amounts of muscle mass to ensure peak performance in their given sport. However, the role of maintaining muscle mass during weight loss and as we age is an emerging concept, having implications in chronic disease prevention, functional capacity, and quality of life. Higher-protein diets have been shown to: (1) promote gains in muscle mass, especially when paired with resistance training; (2) spare muscl

Duchenne muscular dystrophy (DMD) is an X-linked disease caused by the absence of functional dystrophin in the muscle cells. Recent untargeted metabolomics studies identified amino acid metabolism alterations as biochemical pathways potentially involved in DMD pathogenesis. Here, in a well-characterised cohort of DMD children and paediatric controls, we investigated by high-performance liquid chromatography (HPLC) the serum profile of a selected pool of amino acids in D- and L-configuration, including L-glutamate, L-glutamine, glycine, L-aspartate, D-aspartate, L-asparagine, L-serine, and D-se

Previous studies by our group demonstrated a striking relationship among arginine, nitric oxide production, and salt-sensitive hypertension in the Dahl/Rapp rat. We hypothesized that enzymes of the urea cycle may be involved in this process. We specifically examined the activities of liver and kidney argininosuccinate synthetase (AS), because this enzyme is an essential step of arginine synthesis and a likely control point. We found that salt-sensitive (S) rats on a high-salt diet developed hypertension without change in plasma concentrations of arginine, citrulline, and ornithine. Baseline pl

It is possible that dietary conditions can result in the production of abnormal bone protein. For example, a heavily maize-dependent diet could be deficient in one or more essential amino acids necessary to normal human biochemistry and consequently necessary for normal bone protein synthesis. Amino acid analysis of bone tissues, thus, could provide a useful diagnostic tool in paleopathology. To test this potential we have compared the amino acid analyses of bone samples from a prehistoric Southwest Indian child exhibiting porotic hyperostosis with samples taken from (1) two children's skeleto

Deficient production of nitric oxide may be responsible for the defective defense barrier and persistence of bacterial infection. To gain insight into amino acid-metabolism and L-arginine-nitric oxide system, we studied 34 end-stage renal disease (ESRD) patients on peritoneal dialysis (PD) (20 males, 14 females, with a mean age of 53.5 years and a mean duration on PD of 29.7 months). The concentrations of amino acids, including L-arginine, were measured in peritoneal dialysate and in the serum. The data demonstrated that patients with ESRD on PD have normal serum amino-acid profiles, whereas t

Microalgae-based bioremediation of wastewater enables simultaneous nutrient removal and production of value-added biomass. This study systematically evaluated three geographically distinct Chlorella sp. strains for semi-continuous treatment of food wastewater, addressing critical challenges that hinder practical application. All strains demonstrated substantial nutrient removal (NO3--N > 73%, TP > 78%), with film-forming Chlorella sp. CQ uniquely sustaining complete nitrate elimination. This strain simultaneously produced biomass in which essential amino acids accounted for 40% of total amino

Sarcopenia is a prevalent condition in older adults that involves the progressive loss of muscle mass and function, affecting their autonomy and quality of life. Balanced nutrition, especially adequate protein intake, is essential for preventing and managing this condition. This narrative review analyzes the importance of both the quantity and quality of protein in the diet of older adults to preserve muscle mass and prevent age-related functional decline. It is emphasized that general protein recommendations may be insufficient for this population, requiring higher doses of protein, appropria

Parasitic feather-feeding lice have diets rich in amino acids but deficient in essential B vitamins. Around 22% of feather-feeding louse species are host to vertically inherited endosymbiotic bacteria belonging to the genus Sodalis. These endosymbiotic Sodalis species have undergone genome degeneration, with genes supporting amino acid synthesis being lost, while genes required for B vitamin synthesis are largely retained. However, several species of lice are host to endosymbionts belonging to proteobacteria genera other than Sodalis. It is unknown if these other endosymbionts have undergone a

Generalized pustular psoriasis is a potentially life-threatening skin disease, associated with IL36RN disease alleles. IL36RN encodes the IL-36 receptor antagonist (IL-36Ra), a protein that downregulates the activity of IL-36 cytokines by blocking their receptor (IL-36R). Although generalized pustular psoriasis can be treated with IL-36R inhibitors, the structural underpinnings of the IL-36Ra/IL-36R interaction remain poorly understood. In this study, we sought to address this question by systematically investigating the effects of IL36RN sequence changes. We experimentally characterized the e

L-type amino acid transporter 1 (LAT1) is upregulated in various malignant tumors in humans. LAT1 expression correlates with the grade of cancer and prognosis. LAT1 is responsible for the supply of many essential amino acids to cancer cells. Inhibition of LAT1 reduces the amino acids that enter the cell and inhibits cancer cell growth. Therefore, novel anticancer drugs targeting LAT1 have attracted much attention in recent years. In this study, to explore the applicability of using LAT1 expression in intracranial tumors as a prognostic factor and therapeutic target, we investigated the express

Interaction of the envelope glycoprotein of hepatitis C virus (HCV) with a cellular receptor(s) is thought to be essential for the initial steps of HCV infection. However, the mechanisms of HCV infection remain unclear. The aim of the present study was to determine the features of HCV that enable efficient entry of the virus into human hepatocytes. Variations of hypervariable region 1 (HVR1) sequences in HCV inocula and in infected human hepatoblastoma HepG2 cells were examined. Immunofluorescence of inoculated HepG2 cells with anti-HCV core antibodies demonstrated that HCV structural proteins
