Channel-Guided Engineering of Tryptophan Synthase β-Subunit for High-Level Biosynthesis of the Garlic Bioactive Compound S-Allyl-l-Cysteine.

S-Allyl-l-cysteine (SAC) is a bioactive organosulfur compound from aged garlic that has attracted considerable interest for its health-promoting properties. Here, we developed a channel-guided engineering strategy to enhance SAC biosynthesis using the β-subunit of tryptophan synthase (TrpB). Genome mining guided by channel similarity-identified a thermostable Thermotoga maritima TrpB (TmTrpB) as a suitable scaffold with intrinsic activity toward allyl mercaptan. Engineering the tunnel-lining residues yielded a beneficial mutation, TmTrpB4T, which increased SAC production by 2.5-fold compared w

Discovery and Engineering of a Flavin-Containing Monooxygenase From Bradyrhizobium sp. 200 for the Asymmetric Synthesis of an Indoxacarb Intermediate.

Indoxacarb is an oxadiazine insecticide bearing a single stereogenic center, of which only the (S)-enantiomer exhibits insecticidal activity. Therefore, the synthesis of the key intermediate (S)-5-chloro-2-methoxycarbonyl-2-hydroxy-1-indanone (2) is crucial for the enantioselective total synthesis of Indoxacarb. In this study, a flavin-containing monooxygenase from Bradyrhizobium sp. 200 (BjFMO) was employed for the asymmetric synthesis of this chiral intermediate, offering a biocatalytic approach as a potential complement to conventional chemical routes. Based on molecular docking and sequenc

Computational rational design of unspecific peroxygenase for C-H oxidation.

Computational rational design has emerged as a transformative approach to engineer enzymes with tailored selectivity and efficiency. In the context of carbon-hydrogen oxidation, a key challenge in synthetic chemistry, unspecific peroxygenases (UPOs) directly oxidize unactivated carbon-hydrogen bonds using hydrogen peroxide, yet their utility is limited by low activity and imperfect selectivity. By computational rational design, this study systematically navigated vast sequence spaces to identify mutations that enhance catalytic performance of UPOs, lastly yielded UPO variants with 13-fold enha

Unraveling the Catalytic Promiscuity Mechanism of Triterpene Synthase AaOSC-22030 via Multiscale QM/MM Simulations and Mutagenesis.

The triterpene synthase AaOSC-22030 from Artemisia argyi exhibits remarkable catalytic promiscuity, converting 2,3-oxidosqualene into diverse triterpenoid skeletons. In this study, we employed multiscale molecular dynamics, static QM/MM calculations, and site-directed mutagenesis to elucidate the origins of this diversity. Our analysis identifies residue 728 as a pivotal determinant. We reveal that the substitution of a conserved aromatic residue with a polar serine (S728) creates a relaxed steric environment, permitting broad conformational exploration, while simultaneously serving as a gener

Study on the Asymmetric Synthesis of Chiral 3,3,3-Trifluoro-2-Hydroxypropanoic Acids by Lactate Dehydrogenase.

Chiral 3,3,3-trifluoro-2-hydroxypropanoic acid (3,3,3-trifluorolactic acid, TFLA), which possesses two significant functional groups, is a versatile intermediate in pharmaceutical and material synthesis. A feasible strategy for producing both the enantiomers of chiral TFLAs involves reduction of the corresponding pyruvate using lactate dehydrogenases (LDHs). In this study, ldh genes encoding l-LDHs from animals and d/l-LDHs from lactic acid bacteria are cloned and all the recombinant LDHs are successfully expressed with a histidine tag in Escherichia coli BL21 (DE3). To achieve cofactor regene

Enantioselective Trifluoromethylazidation of Styrenyl Olefins Catalyzed by an Engineered Nonheme Iron Enzyme.

Organofluorines, particularly those containing trifluoromethyl (CF3) groups, play a critical role in medicinal chemistry. While trifluoromethylation of alkenes provides a powerful synthetic route to construct CF3-containing compounds with broad structural and functional diversity, achieving enantioselective control in these reactions remains a formidable challenge. In this study, we engineered a nonheme iron enzyme, quercetin 2,3-dioxygenase from Bacillus subtilis (BsQueD), for the enantioselective trifluoromethylazidation of alkenes. Through directed evolution, the final variant BsQueD-CF3 ex

Nanoflower-structured MnO2/Au composites with enhanced catalytic performance for colorimetric assay of α-glucosidase.

BACKGROUND: α-Glucosidase (α-Glu) is an essential hydrolase that catalyzes the final step of carbohydrate digestion, playing a central role in glucose metabolism. Abnormal α-Glu activity is closely associated with type II diabetes, obesity, and Alzheimer's disease, making its precise monitoring critical for disease diagnosis and therapeutic evaluation. However, conventional detection methods often depend on natural enzymes or sophisticated nanomaterials, which face challenges of high cost, instability, and complicated operation. Thus, developing a simple, robust, and cost-effective strategy fo
