Rapid determination of indomethacin and salicylic acid in serum by means of reversed-phase liquid chromatography.

A method for the quantitative analysis of indomethacin and salicylic acid in blood serum and urine by high-performance liquid chromatography is described. A C18-bonded silica was used as the stationary phase and mixtures of ethanol, n-butanol and aqueous buffer as the mobile phase. Before injection the serum is deproteinized and extracted in one step. The recovery of the extraction was found to be 88% and 77% for indomethacin and salicylic acid, respectively. The relative standard deviations of the analysis for 0.5 micrograms indomethacin and 5 micrograms salicyclic acid per ml serum were 3.6%

A selection model for umbilical cord blood units with high isolation yield of total nucleated and CD34+ cells.

BACKGROUND: Cord blood is an important hematopoietic stem cell source for treating over 80 FDA-approved diseases, driving demand for HSC-containing-umbilical cord blood (UCB) unit cryopreservation for future use. Total nucleated cell (TNC) and CD34+ cell amounts are key determinants of transplant success. Predicting processing outcomes is clinically valuable, particularly for public stem cell banks where storing high-quality UCB units is a priority. This study aimed to establish selection models for UCB units with high isolation yields of these critical cellular determinants. METHODS: We firs

Renal volume and cardiovascular risk assessment in normotensive autosomal dominant polycystic kidney disease patients.

Cardiovascular disease, closely related to an early appearance of hypertension, is the most common mortality cause among autosomal dominant polycystic kidney disease patients (ADPKD). The development of hypertension is related to an increase in renal volume. Whether the increasing in the renal volume before the onset of hypertension leads to a major cardiovascular risk in ADPKD patients remains unknown.Observational and cross-sectional study of 62 normotensive ADPKD patients with normal renal function and a group of 28 healthy controls. Renal volume, blood pressure, and renal (urinary albumin

Assessment of Dried Plasma Spots (DPS) as an Appropriate Sample Matrix for the Measurement of Circulating Calretinin, a Biomarker for Mesothelioma-A Proof-Of-Concept Study.

BACKGROUND: Microsampling using filter cards for dried blood spots (DBS) and dried plasma spots (DPS) is a well-established technique offering practical advantages for blood collection and subsequent processing. For DBS/DPS, only a small sample volume is required it enables transport and storage at ambient temperature, and it is well suited for decentralized sample collections and resource-limited settings, as well as collection from elderly individuals, when venous blood collection might be problematic. Many protein-based biomarker assays like enzyme-linked immunosorbent assays (ELISA) depend

Determination of paraquat in human blood plasma using reversed-phase ion-pair high-performance liquid chromatography with direct sample injection.

This report describes the determination of paraquat (PQ) in human blood plasma samples by a direct-injection reversed-phase ion-pair chromatographic method. Blood plasma filtrate was injected directly into the LiChrospher(R) RP-18 alkyl-diol silica (ADS) precolumn integrated in a column switching system using a mixture of 3% 2-propanol and 10 mM sodium octane sulfonate (SOS) in a 0.05 M phosphate buffer (pH 2.8). After washing with this phase, the ADS precolumn was back-flushed with the analytical mobile phase consisting of 40% of methanol and 10 mM SOS in a 0.05 M phosphate buffer (pH 2.8) at

Platelet function investigation by flow cytometry: Sample volume, needle size, and reference intervals.

Flow cytometry is an increasingly used method for platelet function analysis because it has some important advantages compared with other platelet function tests. Flow cytometric platelet function analyses only require a small sample volume (3.5 mL); however, to expand the field of applications, e.g., for platelet function analysis in children, even smaller volumes are needed. Platelets are easily activated, and the size of the needle for blood sampling might be of importance for the pre-activation of the platelets. Moreover, to use flow cytometry for investigation of platelet function in clin

Evaluation of the Tasso-SST® capillary blood microsampling device for the measurement of endogenous uracil levels.

BACKGROUND: Uracil (U) plasma or serum levels can be used as surrogates of dihydropyrimidine dehydrogenase (DPD) activity, which is strongly related to the occurrence of severe or fatal toxicity after administration of fluoropyrimidines (FP) chemotherapy. Obtaining blood plasma or serum for U measurement usually requires a phlebotomy procedure by a qualified professional. An alternative to conventional blood drawn is the use of the Tasso-SST® device, which allows the collection of a small blood volume from skin capillaries. This study aimed to implement a sensitive liquid chromatography-tandem

Is neuropeptide Y a contributor to volume-induced hypertension?

Hypertension often occurs with fluid overload. The most common mechanism is considered to be mediated by increased cardiac output. Hemodialysis (HD) patients frequently have large amounts of fluid overload. Neuropeptide Y (NPY) is activated by stress and contributes to hypertension and heart failure. We speculated that NPY may be released by the stress of fluid overload and, by its vasoconstrictor effect, may contribute to hypertension and heart failure. Plasma levels of NPY and other vasoconstrictors were studied in 20 HD patients with varying degrees of fluid overload, and the relationship o

Alternative Sampling Devices to Collect Dried Blood Microsamples: State-of-the-Art.

Dried blood spots (DBS) have been used in newborn screening programs for several years. More recently, there has been growing interest in using DBS as a home sampling tool for the quantitative determination of analytes. However, this presents challenges, mainly because of the well-known hematocrit effect and other DBS-specific parameters, including spotted volume and punch site, which could add to the method uncertainty. Therefore, new microsampling devices that quantitatively collect capillary dried blood are continuously being developed. In this review, we provided an overview of devices tha

Neutrophil volume distribution width: a new automated hematologic parameter for acute infection.

CONTEXT: The number of band forms and immature neutrophils increases during acute bacterial infection. However, the determination of band counts and other neutrophil morphologic changes, such as the presence of toxic granulation, toxic vacuolization, and Dohle bodies in the cytoplasm, is labor intensive and time consuming, as it requires manual examination by an experienced medical technologist. OBJECTIVE: To investigate the value of the neutrophil volume distribution width (NDW), generated by VCS technology of the Coulter LH 750 hematology analyzer, as an additional predictor of acute infect

Chemical blockage of cervical sympathetic trunks for facial palsy.

Impairment of microcirculation of the facial nerves is considered to play an important role in etiology of facial palsy, and the main emphasis of the conservative treatment is aimed at circulatory improvement. Therefore, at the pain clinic, improved blood flow to the facial nerve by chemical blockage of cervical sympathetic trunks (stellate ganglion block; SGB) is employed in treating patients with facial palsy. The circulatory effects of SGB were clinically determined in 20 patients with acute Bell's palsy, and the effects of chemical blockage of cervical sympathetic trunks on the tissue circ

Frequency of low level bacteremia in infants from birth to two months of age.

BACKGROUND: The frequency of low level bacteremia (< or = 10 colony-forming units/ml) in infants from birth to 2 months of age and the optimal volume of blood and number of blood cultures to be collected have not been well-documented. During 1991 guidelines at this hospital for collection of blood for culture from these infants were revised. METHODS: Blood from each infant with suspected bacteremia was usually inoculated into an Isolator 1.5 Microbial Tube (1.5 ml of blood) and a bottle of anaerobic broth (0.5 to 3.0 ml of blood). The use of a second Isolator tube and the total blood volume r

Intra-pleural Instillation of Autologous Blood - Fine-tuning Techniques for Better Success Rate: Two Case Reports.

In the past, many case series have reported the effectiveness of autologous blood patch pleurodesis (ABPP) in recurrent secondary spontaneous pneumothorax (SSP), particularly in those who were unfit for surgery. We describe two cases of persistent air leak in pneumoconiosis and pulmonary fibrosis with bronchiectasis, whereby the techniques employed had improved the success rate of ABPP. The determining factors that lead to the success of ABPP were determined by the volume of autologous blood instilled, Trendelenburg position post instillation, and early chest physiotherapy with mobilisation by

Regulation of water intake.

Here we have reviewed mainly the cerebral regulation of water intake and its relationship with the regulation of the water-retaining antidiuretic hormone (ADH). Much new information of obvious interest has been gained by experiments in conscious animals, by studies in healthy humans, and by clinical investigations. Of particularly great value has been the development of a sensitive radioimmunoassay for determination of plasma ADH (59). The sketchy picture that emerges in light of this new information is as follows. The osmotic regulation of water intake and ADH secretion is exerted by juxtacer

Anti-Müllerian Hormone Concentrations for Determining Resumption of Sertoli Cell Function following Removal of a 4.7 mg Deslorelin Implant in Tomcats.

Background: Deslorelin implant use in cats is a medical alternative to surgical sterilization, and due to its prolonged efficacy, its use has shown growing interest in the veterinary community. In the case of breeding facilities, its removal is often requested for the early restoration of testicular function. As anti-Müllerian hormones (AMH) in males is dependent of testosterone secretion, its assay may determine the restoration of testicular steroid secretion. An average of 3 weeks has been already described for tomcats' testicular function resumption after implant removal, but information ab
