Isothermal microcalorimetry accurately detects bacteria, tumorous microtissues, and parasitic worms in a label-free well-plate assay.

Isothermal microcalorimetry is a label-free assay that allows monitoring of enzymatic and metabolic activities. The technique has strengths, but most instruments have a low throughput, which has limited their use for bioassays. Here, an isothermal microcalorimeter, equipped with a vessel holder similar to a 48-well plate, was used. The increased throughput of this microcalorimeter makes it valuable for biomedical and pharmaceutical applications. Our results show that the sensitivity of the instrument allows the detection of 3 × 10(4) bacteria per vial. Growth of P. mirabilis in Luria Broth med…

The interactions between CdSe quantum dots and yeast Saccharomyces cerevisiae: adhesion of quantum dots to the cell surface and the protection effect of ZnS shell.

The interactions between quantum dots (QDs) and biological systems have attracted increasing attention due to concerns on possible toxicity of the nanoscale materials. The biological effects of CdSe QDs and CdSe/ZnS QDs with nearly identical hydrodynamic size on Saccharomyces cerevisiae were investigated via microcalorimetric, spectroscopic and microscopic methods, demonstrating a toxic order CdSe>CdSe/ZnS QDs. CdSe QDs damaged yeast cell wall and reduced the mitochondrial membrane potential. Noteworthy, adhesion of QDs to the yeast cell surface renders this work a good example of interaction

Thermodynamic characterization of a thermostable antibiotic resistance enzyme, the aminoglycoside nucleotidyltransferase (4').

The aminoglycoside nucleotidyltransferase (4') (ANT) is an aminoglycoside-modifying enzyme that detoxifies antibiotics by nucleotidylating a … Energetic aspects of binding of all aminoglycosides to ANT were determined by isothermal titration calorimetry to be enthalpically favored and entropically disfavored with an overall favorable Gibbs energy. Aminoglycosides in the neomycin class each bind to the enzyme with significantly different enthalpic and entropic contributions, while those of the kanam …

Metabolic assessment of menstruating and nonmenstruating normal weight adolescents.

OBJECTIVE: Resumption of menses (ROM) is a key indicator of recovery in AN, but patients may remain amenorrheic despite weight restoration. The objective of this study is to better understand the mechanism of amenorrhea in patients with eating disorders. METHOD: A retrospective chart review was conducted of 382 normal weight adolescents with a history of anorexia nervosa, bulimia nervosa, or eating disorder not otherwise specified, who had been referred for indirect calorimetry tests. Resting energy expenditure (REE) was compared between amenorrheic (n = 60) and regularly menstruating females…

Thermodynamics and density of binding of a panel of antibodies to high-molecular-weight capsular polysaccharides.

The interaction between antipolysaccharide (anti-PS) antibodies and their antigens was investigated by the use of isothermal titration calorimetry to determine the thermodynamic binding constant (K), the change in the enthalpy of binding (DeltaH), and the binding density (N) to high-molecular-weight PSs. From these values, the change in the entropy of binding (DeltaS) was calculated. The thermodynamic parameters of binding to high-molecular-weight capsular PSs are reported for two monoclonal antibodies (MAbs) with different specificities for meningococcal serogroup C PS, five MAbs specific for…

Micronization and polymorphic conversion of tolbutamide and barbital by rapid expansion of supercritical solutions.

Rapid expansion of supercritical solutions (RESS) was applied to tolbutamide and barbital. The solubility in supercritical CO2 was determined to estimate the extraction efficiency roughly by a simple method and accurately by a direct spectrophotometric technique. The latter revealed that the solubility of tolbutamide was a function of applied pressure and temperature and was proportional to the pressure. No significant difference in solubility between polymorphic Forms I and II of tolbutamide was detected. Tolbutamide and barbital particles produced by the RESS were characterized by size distr

Energy requirements in patients with chronic kidney disease.

Knowledge concerning energy requirements of patients with chronic kidney disease (CKD) is important to providing a sufficient amount of energy to maintain adequate nutritional status for these patients. Data regarding energy expenditures of CKD patients are still scarce, and the results obtained are conflicting, with studies showing energy expenditures to be similar, higher, or lower than those of healthy individuals. More recently, studies focusing the role of the comorbidities and of the dialysis procedure on energy expenditure have been carried out, opening a new field of discussion that ma

Preparation and characterization of the inclusion complex of furosemide with hydroxypropyl-beta-cyclodextrin.

A highly hydrophobic drug, furosemide, was treated with 2-Hydroxypropyl-beta-Cyclodextrin in an attempt to produce modified compounds with increased water solubility. Following a freeze-drying process, the interactions of furosemide and cyclodextrin led to the formation of an inclusion complex, whose dissolution rate in water was studied by the solubility method. Furthermore, mechanical mixtures of furosemide and cyclodextrin were also prepared and tested for their solubility. The reaction products were characterized in the solid state by differential scanning calorimetry (DSC), which provided…

Alkali-induced conversion of beta-chitin to alpha-chitin.

Crystal conversion of beta-chitin to alpha-chitin by aq. NaOH treatment was studied for a highly crystalline beta-chitin sample from diatom spine. The minimum NaOH concentration to cause swelling was between 25% and 30% w/w. The alkali-swollen material was poorly crystalline and was regenerated as alpha-chitin on washing with water. This conversion caused total collapse of the original microfibrillar morphology. These features are similar to those of 7 N-8 N HCl treatment reported earlier, but alkali treatment was free from depolymerization or deacetylation.

Accumulation of sunscreens and other compounds in keratinous substrates.

Several cosmetic ingredients, especially sunscreens, should be substantive, which means they are to be adsorbed to specific binding sites within the upper skin layers, particularly keratinized structures of the stratum corneum, and thus show resistance to washing off. We investigated the affinity of 10 non-ionic compounds, among these UV-absorbing chemicals, antioxidants, antimicrobial compounds and a repellent to animal keratin and human callus. In each case a linear relationship between the drug amount, which has accumulated in the respective keratin, and the remaining free concentration of

Increased energy expenditure in infants with cyanotic congenital heart disease.

Infants with cyanotic congenital heart disease (CCHD) often have reduced weight gain compared with infants in control groups. Our purpose was to conduct a longitudinal study of energy intake, resting energy expenditure (REE), and total energy expenditure (TEE) of a group of infants with CCHD. We hypothesized that increased REE and TEE and decreased energy intake in these infants would lead to reduced growth. Ten infants with uncorrected CCHD and 12 infants in a control group were studied at 2 weeks of age and again at 3 months. Indirect calorimetry was used to determine REE; the doubly labeled…

Energy utilization of sodium hydroxide treated or untreated straw supplemented with protein or concentrates by adult sheep. 1. Feed intake, digestibility, metabolizability and net energy value.

NaOH treated (T) or untreated (UT) barley straws were supplemented either with protein (P) or with concentrates based on maize (M) or beet pulp (BP) at rates of 25 or 50% of the diet. Each of the 8 diets was fed to 8 sheep near ad libitum and near maintenance. Energy balances were determined by indirect calorimetry. Straw and feed intakes were doubled by the alkali treatment (P, BP25 and BP50 diets) or increased by 83% (M25 diet) and 63% (M50 diet). Near maintenance, energy digestibility (dE) and metabolizability (q) of the straw were improved by 36% and 40% respectively in the P diet and by 2…

New Insights into the Structure and Thermodynamic Stability of Polymorphs I and II of the Nicotinamide:Adipic Acid Co-Crystal: A BEST-CSP Study.

The polymorphism of the 1:1 nicotinamide:adipic acid (NIC:AA) cocrystal system was re-examined by combining structural and thermodynamic approaches. Differential scanning calorimetry (DSC), solid-state NMR (ssNMR), and variable-temperature powder X-ray diffraction (VT-PXRD) confirmed an enantiotropic relationship between the two polymorphs, evidenced by a direct solid-solid transformation from triclinic form I to monoclinic form II at ∼ 370 K. Solubility data subsequently indicated that this temperature is ∼78 K higher than the equilibrium transition temperature (289.2 ± 0.7 K), consistent wit…

Synthesis and Ring-Opening Polymerization of Mono-Benzoxazines Based on 2-(2-Hydroxyethoxy)phenol and Aniline/Furfurylamine.

Two mono-benzoxazines are synthesized from 2-(2-hydroxyethoxy)phenol, aniline, furfurylamine, and formaldehyde, respectively. The ring-opening polymerization (ROP) of the two mono-benzoxazines is studied with differential scanning calorimetry, Fourier transform infrared spectroscopy, nuclear magnetic resonance, and electrospray ionization mass spectrometry. Compared to phenol-aniline-based benzoxazine, both 2-hydroxyethoxy and N-furfuryl groups can reduce the ROP temperature. Side reactions related to the 2-hydroxyethoxy and N-furfuryl groups are involved in the ROP. The ring-opening reaction …
