Sex differences in insular cortex function in persistent alcohol drinking despite aversion in mice.

One major hallmark of alcohol use disorder (AUD) is the persistence of drinking despite negative consequences. Among indicators of AUD vulne … c neuron activity was higher during persistent ethanol drinking, specifically in female mice. Using chemogenetics, we revealed that inhibition of aIC glutamatergic neurons reduced intake of bitter solutions independently of the solvent (ethanol or water) in both sexes. In addition, inhibition of pIC glutamatergic neurons exclusively reduced persistent ethanol d …

Gut microbial metabolism via hippocampal indole-AhR signaling regulates emotional symptoms.

Gut microbiota modulate emotion, yet mechanistic insight and therapeutic targets remain limited. Here, we identify reduced gut microbiota-derived indole causally modulating emotion through hippocampal aryl hydrocarbon receptor (AhR). We found that decreased abundance of Alistipes shahii reduced intestinal indole levels via loss of tryptophanase, the rate-limiting step for indole production in microbial tryptophan metabolism, in irritable bowel syndrome (IBS) patients and model mice. In the brain, indole acts via AhR, which is enriched in the ventral dentate gyrus (vDG), a hub in regulating emo

Metformin Restores Innate Fear Deficits in Young Adult Female APP/PS1 Mice via Basolateral Amygdala Activation.

Alzheimer's disease (AD) is a prevalent neurodegenerative disorder characterized by cognitive decline and neuropsychiatric symptoms, among which impaired fear responses are particularly notable. While studies have established that learned fear is compromised in AD, the status of innate fear in this context remains unclear. We found that young adult female APP/PS1 mice exhibited reduced freezing and avoidance responses to 2,5-dihydro-2,4,5-trimethylthiazoline (TMT) compared to their wild-type littermates, indicating an impaired innate fear response. Additionally, c-Fos-positive cells in the bas

Bidirectional Cerebellar Control of Suprasecond Timing in Rats.

The cerebellum is well established in subsecond motor timing, but its role in suprasecond interval timing remains unclear. Here, we investigated how cerebellar output influences time estimation over longer timescales. Male rats performed a nose-poke interval timing task in which reward availability could be predicted either from a fixed 2.5 s auditory cue (cued trials) or had to be estimated internally during uncued 3.5 s trials that demanded self-timing. Chemogenetic inhibition of the lateral cerebellar nucleus (LCN) produced bidirectional effects: delayed action initiation in predictable tri

Activation of D2-Expressing Medium Spiny Neurons in the Nucleus Accumbens Core Mirrors the Delaying Effect of Novelty Exposure on Feeding Onset in Rats.

The nucleus accumbens core (NAcc) plays a central role in integrating reward, motivation, and environmental cues to guide goal-directed behaviors, including feeding. Within the NAcc, D2-expressing medium spiny neurons (D2-MSNs) are implicated in inhibitory control and aversive processing, yet their role in feeding initiation remains poorly understood. Using a chemogenetic approach (DREADDs) in male rats housed in calorimetry-metabolic cages, we investigated the contribution of NAcc D2-MSNs to meal patterning and locomotor activity. Activation of D2-MSNs delayed feeding onset and reduced locomo

A male-biased hypothalamic circuit modulates postmating sleep suppression in mice.

Many species display mating-related sleep suppression to maximize reproductive success, yet the underlying neural mechanisms in mammals remain unclear. Here, we report that male mice exhibit delayed sleep onset and reduced sleep duration in periods following successful mating, while females are unaffected. Single-cell transcriptomics identifies a group of sleep-promoting Rxfp1-expressing neurons in the preoptic area of hypothalamus (POARxfp1 neurons), and fiber photometry reveals selective inhibition of them during and after mating in male mice. Lifting this inhibition via chemogenetic activat

A prelimbic cortex-lateral hypothalamus circuit regulates acquisition of alprazolam-induced conditioned place preference in male mice.

Alprazolam (Alp), a high-potency benzodiazepine, poses a significant risk of abuse and addiction, yet the precise mechanisms underlying Alp-associated reward memory remain poorly defined. Here, we first established that administration of Alp at 10 mg/kg reliably induces conditioned place preference (CPP) in male mice. Our results indicated that prelimbic cortex (PrL) CaMKⅡα neurons were hyperactive in Alp-trained mice. Chemogenetic manipulation of the activity of PrL CaMKⅡα neurons bidirectionally regulated acquisition of Alp-induced CPP. Furthermore, virus tracing indicated a strong PrL CaMKⅡ

Clocks slide rather than freeze during torpor in the mouse.

Torpor is an energy-saving strategy used by animals in response to environmental challenges. Torpor is subject to circadian influence, but the effect of torpor itself on circadian rhythms is poorly understood. We investigated whether torpor alters circadian behavior by assessing locomotor activity after torpor in mice emerging into constant darkness (without Zeitgebers). Torpor was induced by food deprivation or by chemogenetic re-activation of "torpor-active" preoptic area neurons. In both conditions, torpor did not disrupt circadian rhythmicity, although a significant phase advance of ∼40 mi

Anesthesia unconsciousness recruits dynamic parabrachial activity within a distributed brain-wide ensemble.

Neural circuits underlying unconsciousness remain poorly defined. We test the hypothesis that unconsciousness arises from specific, distributed circuits using general anesthesia in mice as a reproducible model. We identify a cortical-to-subcortical shift in neural activity during isoflurane anesthesia that is organized into nine discrete functional communities mapped at single-cell resolution. The lateral parabrachial nucleus (LPB) emerges as a central hub, exhibiting high interconnectivity, spontaneous firing under anesthesia, and preferential recruitment during reactivation of the brain-wide

Temporal-orbitofrontal pathway regulates choices across physical reward and visual novelty.

Expectation of perceptual novelty shapes our daily decisions and has an impact on how we evaluate primary rewards. To identify the underlying circuitry, we trained subjects to choose between offers associated with varying probabilities of receiving large juice rewards and experiencing novel objects. Anticipated novelty modulated valuation by increasing both choice preference for and motivational value of large rewards. This novelty-reward interaction was reflected in a circuit comprising anterior ventral medial temporal cortex (AVMTC), implicated in detecting and predicting novelty, and orbito

An orexinergic circuit driving migraine relief by lavender essential oil.

Pharmacological interventions can reduce immediate headache, but long-term effectiveness and suitability for chronic migraine remain limited. Although aromatherapy with lavender essential oil (LEO) reportedly alleviates headaches, the neural mechanisms remain uncertain. Here, we found that LEO inhalation exerts antinociceptive effects by activating orexinergic neurons in the lateral hypothalamus (LHOX) of mice with nitroglycerin-induced chronic migraine. Viral tracing, in vivo fiber photometry, and behavior assays revealed that LEO activates glutamatergic inputs from the anterior olfactory nuc

PVT inhibition mitigates neuropathology and cognitive deficits in chronic cerebral hypoperfusion.

Chronic cerebral hypoperfusion (CCH) is a key pathophysiological substrate of vascular cognitive impairment, and the reorganization of neuronal activity across disease progression underlies network dysfunction and intervention strategy. Using immediate-early gene imaging with whole-brain mapping, we characterized spatiotemporal activation patterns in a bilateral common carotid artery stenosis mouse model at early (day 7) and chronic (day 40) stages across 225 brain regions. Weighted gene co-expression network analysis identified the paraventricular thalamus (PVT) as a persistently hyperactive

A Tac1-Expressing Brainstem Pathway Underlies the Pathogenesis of Trigeminal Neuralgia.

Trigeminal neuralgia (TN) is widely recognized to be one of the most severe pain disorders, severely affects the life quality of patients. However, the neuronal mechanisms underlying TN pathogenesis remain largely unknown. Here, we identify a periphery-to-brain neural circuit that governs TN development in mice. Tachykinin 1 (Tac1)-expressing parabrachial nucleus (PBNTac1) neurons show heightened stimulus-evoked responses, and chemogenetic inhibition of these neurons effectively prevents TN development. Furthermore, PBNTac1 neurons receive projections from the caudal part of the spinal trigemi

Intralaminar thalamus relays basal ganglia output to the insular cortex to drive tic generation.

Motor and vocal tics accompanied by premonitory urges are hallmark symptoms of Tourette syndrome (TS), yet the underlying neuronal mechanisms remain incompletely understood. Here, we establish a mouse model of tic-like movements by unilateral striatal injection of a GABAA receptor antagonist. This model induces c-Fos activation in both motor and limbic structures, including the insular cortex (IC). Fiber photometry reveals tic-associated activity in IC as well as the primary motor cortex (M1). Viral tracing demonstrates that basal ganglia outputs from the substantia nigra pars reticulata are t

Vitamin A deficiency induces sex-specific reward processing alterations through a dysregulation of the mesolimbic dopamine transmission in mice.

Neurodevelopmental psychiatric diseases such as schizophrenia or affective disorders share common symptomatic dimensions, in particular reward processing dysfunctions, associated with dysregulation of dopamine (DA) transmission. Retinoic acid (RA) homeostasis, the active metabolite of vitamin A, is altered across psychiatric disorders but whether impaired developmental RA signaling impacts the functionality of DA-related reward processing at adulthood remains poorly explored. Herein, we found that vitamin A (i.e., retinol) deficiency (VAD) from gestation to adulthood potentiates instrumental r

A GDF-15-GFRAL axis controls autoimmune T cell responses during neuroinflammation.

Inflammatory activity during multiple sclerosis (MS) often improves during pregnancy, suggesting that pregnancy-related immune adaptations affect the disease. Here we show that growth/differentiation factor-15 (GDF-15) increases during pregnancy and correlates with a reduced rate of MS relapses. GDF-15 also accumulates in the inflamed central nervous system, and its absence impairs inflammation resolution in a mouse model of MS. GDF-15 suppresses autoimmune T cell responses through an indirect signaling pathway involving the activation of GDNF family receptor α-like (GFRAL) on brainstem neuron

A lateral hypothalamic region supporting diverse visual processing and modulation of visually-guided behaviour.

Hypothalamic retinal input is traditionally considered distinct from the subcortical pathways supporting vision, specialised to adjust physiology and behaviour alongside variations in ambient illumination. Investigations of retinohypothalamic function have overwhelming focussed on the suprachiasmatic nucleus circadian clock, however. Here we employ multielectrode recording, viral tracing and chemogenetic manipulation in mice to show that another retinohypothalamic target, the anterior lateral hypothalamic area (LHA), displays diverse visual processing capabilities, supporting regulation of mor

Cerebellar Crus II Regulates Recognition and Spatial Memory in Mice.

The cerebellar Crus II is implicated in the late stages of Alzheimer's disease (AD), yet its specific roles in memory regulation and therapeutic potential remain unclear. Using in vivo fiber photometry, we observed robust activation of Crus II neurons in healthy mice during recognition memory tasks. Acute chemogenetic inhibition of Crus II neurons impaired recognition and spatial memory in mice. Polysynaptic circuit tracing revealed that Crus II neurons modulate neural activity in the contralateral prelimbic cortex (PrL) via the Crus II-cerebellar lateral nucleus (LN)-ventromedial thalamus/zon

Ventral hippocampus neurons encode meal-related memory.

The ability to encode and retrieve meal-related information is critical to efficiently guide energy acquisition and consumption, yet the underlying neural processes remain elusive. Here we reveal that ventral hippocampus (HPCv) neuronal activity dynamically elevates between eating bouts during meal consumption and this response is predictive of performance in a foraging-related memory test for the spatial location of a previously consumed meal. Targeted recombination-mediated ablation of HPCv meal-responsive neurons impairs meal location memory without influencing food motivation or spatial me
