Diabetes and neurodegeneration in cognitively unimpaired older adults: implications for cognition.

Type 2 diabetes mellitus is associated with cognitive impairment and greater Alzheimer's disease risk, with cross-sectional studies suggesting that this is driven by neurodegeneration and vascular changes. Longitudinal studies, however, report similar rates of total brain atrophy over time in diabetic and non-diabetic adults. We recently showed that diabetes-related neurodegeneration in cognitively unimpaired adults is regional and may not be evident in longitudinal studies of global brain atrophy. The mechanisms driving diabetes-related neurodegeneration are unknown; glycemic control may play

Clinical patterns in social cognition & personality changes in progressive supranuclear palsy.

Research suggests some individuals with progressive supranuclear palsy (PSP) have socioemotional deficits, typically characterized as reduced emotion recognition, difficulty with perspective taking, and increased apathy. However, the variability of these deficits across individuals and the pattern of impairments across the spectrum of social cognition have not been quantified in larger samples. The present study used multiple face-to-face and informant-based social cognition, neuropsychiatry, and personality measures to identify socioemotional changes occurring as a result of PSP. Data were co…

Healthy dietary patterns, cognition and dementia risk: current evidence and context.

Approximately 60 million individuals worldwide are currently living with dementia. As the median age of the world's population rises, the number of dementia cases is expected to increase markedly, and to affect ∼150 million individuals by 2050. This will create a huge and unsustainable economic and social burden across the globe. Although promising pharmacological treatment options for Alzheimer's disease - the most common cause of dementia - are starting to emerge, dementia prevention and risk reduction remain vital. In this review, we present evidence from large-scale epidemiological studies

Inflammatory Alterations and Multidomain Cognition in Alzheimer Disease and Related Dementias.

OBJECTIVE: To determine how 18kDa Translocator (TSPO) PET, which measures inflammatory alterations, is associated with domain-specific cognition in the presence and absence of elevated amyloid. METHODS: This cross-sectional study uses data collected on 46 adults aged 50 and over who underwent TSPO PET (11C-ER176 SUVR) and amyloid PET (18F-Florbetaben SUVR) as well as medical, neurological, and neuropsychological assessments. Controls (n=21) were amyloid-negative and cognitively unimpaired while individuals with Alzheimer disease or related dementias (ADRD; n=25) were cognitively impaired, reg…

Brain age mediates gut microbiome dysbiosis-related cognition in older adults.

BACKGROUND: Recent studies have focused on improving our understanding of gut microbiome dysbiosis and its impact on cognitive function. However, the relationship between gut microbiome composition, accelerated brain atrophy, and cognitive function has not yet been fully explored. METHODS: We recruited 292 participants from South Korean memory clinics to undergo brain magnetic resonance imaging, clinical assessments, and collected stool samples. We employed a pretrained brain age model- a measure associated with neurodegeneration. Using cluster analysis, we categorized individuals based on th

Prenatal maternal HFD exposure impairs cognition via a hippocampal NMDA mechanism.

Maternal obesity increases the risk of neurodevelopmental disorders and cognitive impairments in offspring later in life. Most animal studies investigated the effects of maternal high-fat diet (HFD) from pre-mating to lactation on offspring neurodevelopment and cognitive function. However, the specific impact of in-utero exposure to maternal HFD on the cognitive function in offspring remains limited. In this study, female dams were fed laboratory chow or HFD for 11 weeks: 8 weeks before conception and during gestation. To isolate the prenatal effects, newborns were reared by foster mothers und

Immersive exposure to simulated visual hallucinations modulates high-level human cognition.

Psychedelic drugs offer valuable insights into consciousness, but disentangling their causal effects on perceptual and high-level cognition is nontrivial. Technological advances in virtual reality (VR) and machine learning have enabled the immersive simulation of visual hallucinations. However, comprehensive experimental data on how these simulated hallucinations affects high-level human cognition is lacking. Here, we exposed human participants to VR panoramic videos and their psychedelic counterparts generated by the DeepDream algorithm. Participants exhibited reduced task-switching costs aft…

Parallel systems for social and spatial cognition reaching the cortical apex.

How are systems supporting high-level cognition organized in the human brain? We hypothesize that cognitive processes involved in understanding people and places are implemented by distinct neural systems with parallel anatomical organization. We test this hypothesis using precision neuroimaging of individual human brains on diverse tasks involving perception and cognition in the domains of familiar people, places, and objects. We find that thinking about people and places elicits responses in distinct areas of high-level association cortex within the default mode network, spanning the frontal…

Underestimated virus impaired cognition-more evidence and more work to do.

Neurodegenerative disorders (NDs) are chronic neurological diseases that can be of idiopathic, genetic, or potentially infectious origin. Although the exact cause of neurodegeneration is unknown, it might be result of a confluence of age, genetic susceptibility factors, and environmental stresses. The blood-brain barrier shields the brain from the majority of viral infections, however neurotropic viruses are able to breach this barrier and infect central nervous system. Growing research points to a possible connection between viruses and neurodegenerative diseases, indicating that virus-induce

Isolation of Distinct Networks Driving Action and Cognition in Psychomotor Processes.

BACKGROUND: Psychomotor disturbances are observed across psychiatric disorders and often manifest as psychomotor slowing, agitation, disorganized behavior, or catatonia. Psychomotor function includes both cognitive and motor components, but the neural circuits driving these subprocesses and how they relate to symptoms have remained elusive for centuries. METHODS: We analyzed data from the HCP-EP (Human Connectome Project for Early Psychosis), a multisite study of 125 participants with early psychosis and 58 healthy participants with resting-state functional magnetic resonance imaging and clin

White matter correlates of cognition: A diffusion magnetic resonance imaging study.

BACKGROUND: Our comprehension of the interplay of cognition and the brain remains constrained. While functional imaging studies have identified cognitive brain regions, structural correlates of cognitive functions remain underexplored. Advanced methods like Diffusion Magnetic Resonance Imaging (DMRI) facilitate the exploration of brain connectivity and White Matter (WM) tract microstructure. Therefore, we conducted connectometry method on DMRI data, to reveal WM tracts associated with cognition. METHODS: 125 healthy participants from the National Institute of Mental Health Intramural Healthy …

Functional integration and segregation during semantic cognition: Evidence across age groups.

Semantic cognition is underpinned by ventral anterior temporal lobe (vATL) which encodes knowledge representations and inferior frontal gyrus (IFG), which controls activation of knowledge based on the needs of the current context. This core semantic network has been validated in substantial empirical findings in the past. However, it remains unclear how these core semantic areas dynamically communicate with each other, and with other neural networks, to achieve successful semantic processing. Here, we investigated this question by testing functional connectivity in the core semantic network du…

Association between arterial health and cognition in adolescents: The PANIC study.

We investigated the associations of the measures of arterial health with cognition in adolescents and whether physical activity (PA) or sedentary time (ST) confounds these associations. One hundred sixteen adolescents (71 boys) aged 15.9 ± 0.4 participated in the study. PA and ST were assessed using a combined accelerometer/heart rate monitor. Overall cognition was computed from the results of psychomotor function, attention, working memory, and paired-associate learning tests. Pulse wave velocity was measured by impedance cardiography, carotid intima-media thickness, and carotid artery disten…

Atypical semantic cognition in schizotypal personality disorder and borderline personality disorder.

Increased schizotypal traits have previously been associated with atypical semantic cognition in community samples. However, no study has yet examined whether adults diagnosed with schizotypal personality disorder (SPD) display atypical semantic fluency and memory. We hypothesized that 24 adults diagnosed with SPD would name more idiosyncratic words on the semantic fluency task and show decreased semantic recall for animal and fruit category words compared with 29 participants with borderline personality disorder (BPD) and a community sample of 96 age-matched controls. We examined whether atyp…

Monoaminergic degeneration, cognition, and autonomic symptom trajectory in early Parkinson's disease.

INTRODUCTION: Parkinson's disease (PD) encompasses a range of non-motor symptoms attributed to deficits in various neurotransmitter systems. This study aimed to investigate the associations between cognitive and autonomic symptoms and the degeneration of brainstem monoaminergic nuclei, particularly the serotonergic and noradrenergic nuclei, in a prospective cohort of early PD patients. METHODS: Twenty-eight early PD patients (with an average disease duration of approximately three years) underwent baseline [18F]FP-CIT positron emission tomography (PET) scans, Montreal Cognitive Assessment (Mo

Plasma trimethylamine N-oxide (TMAO): associations with cognition, neuroimaging, and dementia.

BACKGROUND: The gut-derived metabolite Trimethylamine N-oxide (TMAO) and its precursors - betaine, carnitine, choline, and deoxycarnitine - have been associated with an increased risk of cardiovascular disease, but their relation to cognition, neuroimaging markers, and dementia remains uncertain. METHODS: In the population-based Rotterdam Study, we used multivariable regression models to study the associations between plasma TMAO, its precursors, and cognition in 3,143 participants. Subsequently, we examined their link to structural brain MRI markers in 2,047 participants, with a partial vali…
