Study on the role of the combination of quercetin and lutein in alleviating ulcerative colitis in mice.

INTRODUCTION: This study aims to investigate the ameliorative effects and molecular mechanisms of combined quercetin and luteolin intervention on dextran sulfate sodium (DSS)-induced colitis in mice. METHODS: By establishing a DSS-induced colitis model, we systematically evaluated the comprehensive effects of the combined treatment on colonic histopathological damage, inflammatory cytokine expression, intestinal barrier function, and gut microbiota composition. RESULTS: The quercetin-luteolin combination treatment group significantly ameliorated colon shortening, with colon length restored t

[Neonatal small left colon syndrome (author's transl)].

The article reports on the newborn of a diabetic mother. Intestinal obstruction (subileus) occurred in the infant as a result of a neonatal small left colon syndrome. Diagnosis was established by radiological examination. Contrast enema with Gastrografin resulted in a permanent curative effect. Follow-up studies showed normalisation of the colonic lumen demonstrating the functional character of the disturbance. The etiology of the syndrome has not yet been clarified. It appears to be of differential diagnostic importance to make a clear distinction between this syndrome and Hirschsprung's dise

[Parenteral nutrition in surgery preparation in large intestinal diseases. Indication, procedure and results].

The basic problem of colonic surgery is the high septic risk. Especially chronic inflammatory diseases are marked by a high periintestinal infection rate. In addition, there are catabolisms with negative protein balance and immunologic deficiency. In order to avoid there factors the principle of withdrawal of the intestinal function has been applied. Patients are put on a balanced high-caloric and completely absorable diet (2,400-3,600 cal) using a thin duodenal tube for several weeks. Depending on the initial condition the alimentation is carried out in some patients parenterally or as a supp

The Intestinal Mucin Isoform Landscape Reveals Region-Specific Biomarker Panels for Inflammatory Bowel Disease Patient Stratification.

BACKGROUND AND AIMS: Mucosal healing is considered a key therapeutic endpoint in inflammatory bowel diseases (IBD) and comprises endoscopic improvement of inflammation without taking barrier healing into account. Mucins are critical components of the mucosal barrier function that give rise to structurally diverse isoforms. Unraveling disease-associated mucin isoforms that could act as an indication for barrier function would greatly enhance IBD management. METHODS: We present the intestinal mucin RNA isoform landscape in IBD and control patients using a targeted mucin isoform sequencing appro

Mucosal colonization of Mannheimia haemolytica capsular and adhesin mutants in cattle.

Mannheimia haemolytica (Mh) is a normal inhabitant of the upper respiratory tract of ruminants and is associated with bovine respiratory disease. Polysaccharide capsule and surface adhesins are suggested to function in adherence and colonization of M. haemolytica to the mucosa of the upper respiratory tract. M. haemolytica serotype 1 mutant strains containing deletions of either the capsule biosynthetic gene cluster (∆cap) or putative adhesin genes (∆adh123) were created using a temperature-sensitive plasmid and tested for colonization in a calf challenge model. Two treatment groups were used

BpaB and EbfC DNA-binding proteins regulate production of the Lyme disease spirochete's infection-associated Erp surface proteins.

Vector-borne pathogens regulate their protein expression profiles, producing factors during host infection that differ from those produced during vector colonization. The Lyme disease agent, Borrelia burgdorferi, produces Erp surface proteins throughout mammalian infection and represses their synthesis during colonization of vector ticks. Known functions of Erp proteins include binding of host laminin, plasmin(ogen), and regulators of complement activation. A DNA region immediately 5' of erp operons, the erp operator, is required for transcriptional regulation. The B. burgdorferi BpaB and EbfC

Defective IL10 signaling defining a subgroup of patients with inflammatory bowel disease.

OBJECTIVES: Early onset inflammatory bowel diseases (EO-IBD) developing during the first year of life are likely to reflect inherited defects in key mechanism(s) controlling intestinal homeostasis, as recently suggested for interleukin 10 (IL10). Thus, we aimed to further elaborate the hypothesis of defective anti-inflammatory responses in patients with IBD. METHODS: The capacities of transforming growth factor β (TGFβ) and IL10 to inhibit proinflammatory cytokine production by monocyte-derived dendritic cells (MoDC) or peripheral blood cells (PBMC) was analyzed in 75 children with IBD, inclu

Impact of Various High Fat Diets on Gene Expression and the Microbiome Across the Mouse Intestines.

High fat diets (HFDs) have been linked to several diseases including obesity, diabetes, fatty liver, inflammatory bowel disease (IBD) and colon cancer. In this study, we examined the impact on intestinal gene expression of three isocaloric HFDs that differed only in their fatty acid composition - coconut oil (saturated fats), conventional soybean oil (polyunsaturated fats) and a genetically modified soybean oil (monounsaturated fats). Four functionally distinct segments of the mouse intestinal tract were analyzed using RNA-seq - duodenum, jejunum, terminal ileum and proximal colon. We found co

Lactose-decorated, alginate-coated chitosan nanoparticles loaded with Coomassie Brilliant Blue G-250 protect colonic barrier integrity in experimental colitis.

Ulcerative colitis involves epithelial injury, crypt distortion, barrier failure, and inflammatory activation within colonic tissue. To support local delivery of the P2X7R antagonist Coomassie Brilliant Blue G-250 (BBG), we developed lactose-decorated, alginate-coated chitosan nanoparticles (ALG-BBG-LCS NPs) and evaluated whether this dual-modified polysaccharide carrier could decouple colonic exposure from systemic exposure while translating into structural protection in dextran sodium sulfate-induced colitis. Lactosylated chitosan was synthesized by the Maillard reaction and confirmed by 1H

Necrotizing colitis complicating necrotized pancreatitis: look out for intestinal pneumatosis.

Acute pancreatitis is a common cause of acute abdominal pain and is associated with a wide variety of complications. Pancreatic necrosis is one of the most important complications and is considered to be the most important indicator of disease severity as the increased frequency of death in acute pancreatitis is directly correlated with the development and extent of pancreatic necrosis. In addition to pancreatic necrosis, wide spectrums of colonic complications have been described, including functional and mechanical ileus, ischemic necrosis and fistula formation. In acute pancreatitis bowel i

Effect of starch malabsorption on colonic function and metabolism in humans.

To study the impact of starch on colonic function and metabolism, 12 healthy volunteers consumed a controlled diet rich in starch for two 4-wk periods. In one of the study periods they received the glucosidase inhibitor acarbose (BAY g 5421) and placebo in the other. Stool wet weight increased by 68%, stool dry weight by 57%, fecal water content by 73%, and the mean transit time by 30% on acarbose. Breath hydrogen was significantly higher on acarbose, indicating stimulated carbohydrate fermentation in the colon. Fecal bacterial mass (+78%), total stool nitrogen (+53%), bacterial nitrogen (+200

Intranasal administration of neuropeptide Y significantly antagonized stress-induced colonic dysmotility via central GABAA receptors in male rats.

Stress plays a major role in the pathogenesis of many diseases. Central neuropeptide Y (NPY) counteracts the biological actions of corticotropin-releasing factor (CRF) and attenuates stress responses. Intracerebroventricular (ICV) administration of NPY significantly antagonized the inhibitory effects of chronic complicated stress (CCS) on gastrointestinal (GI) dysmotility in rats. However, ICV administration is an invasive technique. The effect of intranasal administration of NPY on the hypothalamus-pituitary-adrenal (HPA) axis and GI motility in CCS conditions have not been studied, and the i

5xFAD mice do not have myenteric amyloidosis, dysregulation of neuromuscular transmission or gastrointestinal dysmotility.

BACKGROUND: Alterations in gastrointestinal (GI) function and the gut-brain axis are associated with progression and pathology of Alzheimer's Disease (AD). Studies in AD animal models show that changes in the gut microbiome and inflammatory markers can contribute to AD development in the central nervous system (CNS). Amyloid-beta (Aβ) accumulation is a major AD pathology causing synaptic dysfunction and neuronal death. Current knowledge of the pathophysiology of AD in enteric neurons is limited, and whether Aβ accumulation directly disrupts enteric neuron function is unknown. METHODS: In 6-mo

Colonic dysmotility in postsurgical patients with Hirschsprung's disease. Potential significance of abnormalities in the interstitial cells of Cajal and the enteric nervous system.

PURPOSE: Normal gut muscular function depends on the coordinated activity of both the enteric nervous system (ENS) and the interstitial cells of Cajal (ICC). Hirschsprung's disease (HD) has long been considered a purely neuronal deficit but recent data point to abnormalities in ICC in the proximal ganglionated HD colon. We examined the labeling of ICC and neuronal cells in the proximal ganglionated colon in patients with HD to determine whether abnormalities of ICC and ENS might be associated with a poor clinical outcome. METHODS: Tissue from 11 patients with HD was studied using immunohistoc

Restoring Mucosal Barrier Function and Modifying Macrophage Phenotype with an Extracellular Matrix Hydrogel: Potential Therapy for Ulcerative Colitis.

BACKGROUND AND AIMS: Despite advances in therapeutic options, more than half of all patients with ulcerative colitis [UC] do not achieve long-term remission, many require colectomy, and the disease still has a marked negative impact on quality of life. Extracellular matrix [ECM] bioscaffolds facilitate the functional repair of many soft tissues by mechanisms that include mitigation of pro-inflammatory macrophage phenotype and mobilization of endogenous stem/progenitor cells. The aim of the present study was to determine if an ECM hydrogel therapy could influence outcomes in an inducible rodent

Altered intestinal microbial flora and impaired epithelial barrier structure and function in CKD: the nature, mechanisms, consequences and potential treatment.

Chronic kidney disease (CKD) results in systemic inflammation and oxidative stress which play a central role in CKD progression and its adverse consequences. Although many of the causes and consequences of oxidative stress and inflammation in CKD have been extensively explored, little attention had been paid to the intestine and its microbial flora as a potential source of these problems. Our recent studies have revealed significant disruption of the colonic, ileal, jejunal and gastric epithelial tight junction in different models of CKD in rats. Moreover, the disruption of the epithelial barr

CXCR2-dependent mucosal neutrophil influx protects against colitis-associated diarrhea caused by an attaching/effacing lesion-forming bacterial pathogen.

Enteropathogenic Escherichia coli (EPEC) is a major cause of diarrheal disease in young children, yet symptoms and duration are highly variable for unknown reasons. Citrobacter rodentium, a murine model pathogen that shares important functional features with EPEC, colonizes mice in colon and cecum and causes inflammation, but typically little or no diarrhea. We conducted genome-wide microarray studies to define mechanisms of host defense and disease in C. rodentium infection. A significant fraction of the genes most highly induced in the colon by infection encoded CXC chemokines, particularly

Hyperplastic colonic polyps as a marker for adenomatous colonic polyps.

Hyperplastic colonic polyps are generally regarded as being of little or no clinical consequence. Recently, however, hyperplastic polyps have been found to share numerous functional similarities with colorectal carcinoma. To determine whether the presence of an isolated left-sided colonic hyperplastic (metaplastic) polyp could serve as a marker for more proximal synchronous adenomatous colonic polyps, we retrospectively analyzed all consecutive colonoscopic polypectomies performed over an 18-month period at two medical centers. It is the policy at both institutions to remove or biopsy all poly

Characterisation of bacteria-induced colitis and its modulation by probiotics in naked mole rats: a new mammalian model for acute inflammatory disease.

Enteropathogenic bacteria are a major cause of morbidity and mortality globally. While mouse models have been indispensable in advancing our understanding of infectious enteric diseases, key differences in intestinal microbiota and immunobiology between mice and humans underscore the need for alternative mammalian models that better recapitulate human disease states. The naked mole rat (NMR), the longest-lived rodent and a model of healthy ageing, presents a unique opportunity. It possesses an exceptionally robust intestinal barrier, an abundance of goblet cells, a thicker mucin layer, and red
