Unraveling the photoluminescence response of light-switching ruthenium(II) complexes bound to amyloid-β.

Photoluminescent molecules are widely used for real-time monitoring of peptide aggregation. In this Article, we detail both experimental and computational modeling to elucidate the interaction between [Ru(bpy)2dppz](2+) and amyloid-β (Aβ(1-40)) aggregates. The transition from monomeric to fibrillar Aβ is of interest in the study of Alzheimer's disease. Concentration-dependent experiments allowed the determination of a dissociation constant of 2.1 μM, while Job plots provided a binding stoichiometry of 2.6 Aβ monomers per [Ru(bpy)2dppz](2+). Our computational approach that combines molecular do

Computational biology in the study of cardiac ion channels and cell electrophysiology.

The cardiac cell is a complex biological system where various processes interact to generate electrical excitation (the action potential, AP) and contraction. During AP generation, membrane ion channels interact nonlinearly with dynamically changing ionic concentrations and varying transmembrane voltage, and are subject to regulatory processes. In recent years, a large body of knowledge has accumulated on the molecular structure of cardiac ion channels, their function, and their modification by genetic mutations that are associated with cardiac arrhythmias and sudden death. However, ion channe

Detection of polybrominated biphenyl ethers (PBDEs) in pediatric hair as a tool for determining in utero exposure.

INTRODUCTION: Cryptorchidism, or undescended/maldescended testis, is the most common birth defect of male genitalia. Its prevalence has been increasing over the past few decades. This may be due to an increase in the prevalence of anti-androgenic chemicals such as polychlorinated biphenyls, organochloride pesticides, plasticizers and fungicides. A newer group of chemicals, brominated flame retardants (BFRs), are being implicated as endocrine-disrupting chemicals. These chemicals are used worldwide in polymers that are incorporated into a variety of consumer products (e.g., textile, computers a

In-silico screening to identify phytochemical inhibitor for hP2X7: A crucial inflammatory cell death mediator in Parkinson's disease.

The second most prevalent neurological disease among the elderly is Parkinson's disease, where neuroinflammation plays a significant role in its pathology. Purinergic signaling mediated by P2X7 plays a significant role in neuroinflammation and pyroptotic cell death pathways through mediators like NLRP3, Caspase-1, and Caspase-3, instigating pyroptotic cell death. No synthetic agent advanced in late-stage clinical trials due to their inefficacy and toxicity. Hence, in this study, we aimed to identify a phytoconstituent inhibitor against the hP2X7 receptor to ameliorate the inflammatory processe

Molecular alterations associated with aneurysmal remodeling are localized in the high hemodynamic stress region of a created carotid bifurcation.

OBJECTIVE: Although elevated hemodynamics has been speculated to play a key role in intracranial aneurysm (IA) initiation, little is known about the specific hemodynamic microenvironment that triggers aneurysmal vascular degradation. We previously demonstrated maladaptive remodeling characteristic of IA initiation occurring in hemodynamic regions of combined high wall shear stress (WSS) and high WSS gradient near the apex of an experimentally created carotid bifurcation. This study examines whether this remodeling recapitulates the molecular changes found in IAs and whether molecular changes a

Disrupted mechanobiology links the molecular and cellular phenotypes in familial dilated cardiomyopathy.

Familial dilated cardiomyopathy (DCM) is a leading cause of sudden cardiac death and a major indicator for heart transplant. The disease is frequently caused by mutations of sarcomeric proteins; however, it is not well understood how these molecular mutations lead to alterations in cellular organization and contractility. To address this critical gap in our knowledge, we studied the molecular and cellular consequences of a DCM mutation in troponin-T, ΔK210. We determined the molecular mechanism of ΔK210 and used computational modeling to predict that the mutation should reduce the force per sa

Targeting tau-mitochondrial crosstalk in Alzheimer's disease with an Integrative multi-omics and artificial intelligence driven tools for the development of disease-modifying therapeutics.

Alzheimer's disease (AD) is a progressive neurodegenerative illness marked by cognitive impairment, synaptic dysfunction and neuronal death. Tau protein abnormalities and mitochondrial dysfunction are key features of its pathogenesis, and both are involved in driving disease development. Emerging evidence suggests that pathogenic tau not only destabilizes microtubules but also directly compromises mitochondrial dynamics, bioenergetics and quality control, ultimately aggravating neurodegeneration. However, the molecular processes by which tau disease causes mitochondrial failure are poorly know

TOLEDO: enhancing Maestro GUI for non-expert users to perform massive MD simulations.

Classical Molecular Dynamics (MD) simulates the dynamical evolution of biological systems at the atomic level. Using MD in conjunction with high-performance computing (HPC) architectures, we can evaluate the possible interactions between a ligand library against one protein target to find a drug that can influence a protein target to cure a disease. Simultaneously, we can also obtain information about their dynamic evolution. One of the primary software packages for MD simulations is Desmond, which employs Maestro for the setup, execution, and analysis of MD through a graphical user interface

Quantitative Structure-Activity Relationship Approaches for Exploring the Anticancer Potential of Flavonoids.

Flavonoids are structurally diverse polyphenolic compounds widely distributed in fruits, vegetables, grains, and beverages, with considerable potential as anticancer agents. Their pleiotropic activities involve modulation of key cancer hallmarks, including oxidative stress, cell-cycle progression, apoptosis, autophagy, angiogenesis, invasion, and metastasis. These effects are mediated through multiple signaling pathways, including PI3K/Akt, MAPK, NF-κB, and STAT3. Understanding the relationship between flavonoid structure and biological activity is therefore essential for rational optimization

A synergistic deep learning and machine learning framework for screening heterocyclic compounds against ALDH1A1.

Aldehyde dehydrogenase 1A1 (ALDH1A1) has emerged as a promising therapeutic target because of its critical roles in cancer stem cell maintenance and chemoresistance. However, the development of highly selective ALDH1A1 inhibitors remains challenging because of the extensive structural conservation shared with the closely related isoforms ALDH2 and ALDH1A2. In this study, we developed an integrated computer-aided drug design (CADD) and artificial intelligence (AI) framework to systematically identify selective ALDH1A1 inhibitors from a heterocyclic compound library. The multistage virtual scree

An experimental-computational approach for measuring charged solute diffusivity through human synovium.

Intra-articular drug delivery has shown promise in targeting arthritic joints, but its therapeutic efficacy is hindered by the synovium, a multilayered connective tissue that rapidly clears locally delivered drugs from the joint space. To better understand the mechanisms behind synovial drug clearance, we previously developed a finite element model of synovium as a multiphasic tissue and used an inverse method to determine the effective diffusivity (Deff) of neutral solutes through synovium, which was found to decrease with increasing molecular weight. Here, we adapted this experimental-comput

Computational and Biological Investigations on Abl1 Tyrosine Kinase: A Review.

Abl1 tyrosine kinase is a validated target for the treatment of chronic myeloid leukemia. It is a form of cancer that is difficult to treat and much research is being done to identify new molecular entities and to tackle drug resistance issues. In recent years, drug resistance of Abl1 tyrosine kinase has become a major healthcare concern. Second and third-generation TKI reported better responses against the resistant forms; still they had no impact on long-term survival prolongation. New compounds derived from natural products and organic small molecule inhibitors can lay the foundation for be
