BACTERIOSTATIC EFFECT OF HUMAN SERA ON GROUP A STREPTOCOCCI : I. TYPE-SPECIFIC ANTIBODIES IN SERA OF PATIENTS CONVALESCING FROM GROUP A STREPTOCOCCAL PHARYNGITIS.

1. Type-specific antibodies were demonstrated by the indirect bacteriostatic test in sera from human adults convalescing from group A streptococcal infection of the upper respiratory tract. The time of appearance of the antibodies varied from 3 to 5 weeks; and they persisted in 2 patients for at least 37 weeks after the onset of the infection. 2. The specificity of the antibody response in one serum was tested with strains of 7 heterologous types; in another, with 6; and in the third, with 2; but in no instance were cross-reactions observed. Moreover, each convalescent serum showed approximate

The efficacy and safety of convalescent plasma for COVID-19 patients: a meta-analysis based on double-blinded parallel-arm randomized placebo-controlled trials.

BACKGROUND: Convalescent plasma (CP) showed promising benefits in previous coronavirus pandemics regarding efficacy and safety. However, the efficacy of CP from COVID-19 patients remains controversial and uncertain based on current randomized controlled trials (RCTs). There is an urgent need to establish the efficacy and safety of CP for COVID-19 patients as soon as possible. OBJECTIVE: To verify the efficacy and safety of CP using high-quality, double-blinded, parallel-arm, placebo-controlled randomized clinical trials, and to provide evidence-based support for the clinical application of CP

Improvement in activities of daily living in postoperative patients with cervical spondylotic myelopathy undergoing intensive rehabilitation in a convalescent rehabilitation ward: A retrospective observational study.

Cervical spondylotic myelopathy (CSM) often leads to persistent impairment in motor function and activities of daily living (ADL) even after surgical decompression. Although postoperative rehabilitation is widely implemented, factors associated with ADL outcomes during the convalescent rehabilitation phase remain unclear. This study aimed to investigate ADL outcomes and their associated factors in postoperative patients with CSM admitted to a convalescent rehabilitation ward. This retrospective observational study included postoperative patients with CSM admitted to a convalescent rehabilitati

Convalescent COVID-19 monocytes exhibit altered steady-state gene expression and reduced TLR2, TLR4 and RIG-I induced cytokine expression.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which causes COVID-19, can induce trained immunity in monocytes. Trained immunity is the result of metabolic and epigenetic reprogramming of progenitor cells leading to an altered inflammatory response to subsequent activation. To investigate the monocyte response 3-6 months post SARS-CoV-2 infection, steady-state gene expression and innate immune receptor stimulation were investigated in monocytes from unvaccinated SARS-CoV-2 naïve individuals and convalescent COVID-19 participants. The differentially expressed genes (DEGs) identif

Antibody responses following COVID-19 vaccination and breakthrough infections in naïve and convalescent individuals suggest imprinting to the ancestral strain of SARS-CoV-2.

The binding and neutralizing activity of SARS-CoV-2 antibodies are important correlates of protection of current COVID-19 vaccines. SARS-CoV-2 exposure status and COVID-19 vaccine types can influence these responses and the breadth of cross-reactivity to variants. In this longitudinal cohort study, we used SARS-CoV-2-specific multiplex Luminex antibody assays and live virus neutralization of ancestral (VIC01/2020), Delta and Omicron (BA1, BA2, and BA5) SARS-CoV-2 variants to compare antigen-specific binding, and neutralizing antibody (nAb) responses to primary vaccination (two doses) of adenov

Memory T cell reactivity to a broad range of conserved SARS-CoV-2-derived ORF1ab epitopes in first wave COVID-19 convalescents.

INTRODUCTION: Understanding T cell responses to SARS-CoV-2 is crucial for developing effective vaccines. Memory T cells, including CD8+ cytotoxic and CD4+ helper T cells, play key roles in controlling viral infections. The open reading frame 1ab (ORF1ab) region of SARS-CoV-2, essential for viral replication, is highly conserved. OBJECTIVE: This study aims to analyze whether memory T cell responses against ORF1ab derived HLA class I peptides are present in convalescent individuals. METHODS: Peripheral blood mononuclear cells (PBMCs) were isolated from 53 staff members, including 20 healthy an

Proteomic Analysis of 442 Clinical Plasma Samples From Individuals With Symptom Records Revealed Subtypes of Convalescent Patients Who Had COVID-19.

After the coronavirus disease 2019 (COVID-19) pandemic, the postacute effects of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection have gradually attracted attention. To precisely evaluate the health status of convalescent patients with COVID-19, we analyzed symptom and proteome data of 442 plasma samples from healthy controls, hospitalized patients, and convalescent patients 6 or 12 months after SARS-CoV-2 infection. Symptoms analysis revealed distinct relationships in convalescent patients. Results of plasma protein expression levels showed that C1QA, C1QB, C2, CFH, CFHR

Identification of neutralizing antibodies against monkeypox virus using high-throughput sequencing of A35+H3L+B cells from patients with convalescent monkeypox.

The global monkeypox virus (MPXV) outbreak in 2022 emphasizes the urgent need for effective and accessible new-generation vaccines and neutralizing antibodies. Herein, we identified MPXV-neutralizing antibodies using high-throughput single-cell RNA and V(D)J sequencing of antigen-sorted B cells from patients with convalescent monkeypox. IgG1-expressing B cells were obtained from 34 paired heavy- and light-chain B cell receptor sequences. Subsequently, three potent neutralizing antibodies, MV127, MV128, and MV129, were identified and reacted with the MPXV A35 protein. Among these, MV129, which

Convalescent human plasma candidate reference materials protect against Crimean-Congo haemorrhagic fever virus (CCHFV) challenge in an A129 mouse model.

Crimean-Congo Haemorrhagic Fever Virus (CCHFV) is spread by infected ticks or direct contact with blood, tissues and fluids from infected patients or livestock. Infection with CCHFV causes severe haemorrhagic fever in humans which is fatal in up to 83 % of cases. CCHFV is listed as a priority pathogen by the World Health Organization (WHO) and there are currently no widely-approved vaccines. Defining a serological correlate of protection against CCHFV infection would support the development of vaccines by providing a 'target threshold' for pre-clinical and clinical immunogenicity studies to ac

Boosted production of antibodies that neutralized different SARS-CoV-2 variants in a COVID-19 convalescent following messenger RNA vaccination - a case study.

Vaccinated convalescents do not develop severe COVID-19 after infection with new SARS-CoV-2 variants. We questioned how messenger RNA (mRNA) vaccination of convalescents provides protection from emerging virus variants. From the cohort of 71 convalescent plasma donors, we identified a patient who developed immune response to infection with SARS-CoV-2 variant of 20A clade and who subsequently received mRNA vaccine encoding spike (S) protein of strain of 19A clade. We showed that vaccination increased the production of immune cells and anti-S antibodies in the serum. Serum antibodies neutralized

Variability in Severe Acute Respiratory Syndrome Coronavirus 2 IgG Antibody Affinity to Omicron and Delta Variants in Convalescent and Community mRNA-Vaccinated Individuals.

The emergence of the omicron and delta variants of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has begun a number of discussions regarding breakthrough infection, waning immunity, need and timing for vaccine boosters, and whether existing mRNA vaccines for the original SARS-CoV-2 strain are adequate. Our work leverages a biosensor-based technique to evaluate the binding efficacy of SARS-CoV-2 S1-specific salivary Abs to the omicron and delta variants using a cohort of mRNA-vaccinated (n = 109) and convalescent (n = 19) subjects. We discovered a wide range of binding efficacies

Early administration of remdesivir plus convalescent plasma therapy is effective to treat COVID-19 pneumonia in B-cell depleted patients with hematological malignancies.

Patients with hematological malignancies (HMs) are at a higher risk of developing severe form and protracted course of COVID-19 disease. We investigated whether the combination of viral replication inhibition with remdesivir and administration of anti-SARS-CoV-2 immunoglobulins with convalescent plasma (CP) therapy might be sufficient to treat B-cell-depleted patients with COVID-19. We enrolled 20 consecutive patients with various HMs with profound B-cell lymphopenia and COVID-19 pneumonia between December 2020 and May 2021. All patients demonstrated undetectable baseline anti-SARS-CoV-2 immun

Limited Correlation between SARS-CoV-2 Serologic Assays for Identification of High-Titer COVID-19 Convalescent Plasma Using FDA Thresholds.

In August 2020, the Food and Drug Administration (FDA) Emergency Use Authorization (EUA) for COVID-19 convalescent plasma (CCP) specified 12 authorized serologic assays and associated assay-specific cutoff values for the selection of high-titer CCP for use in hospitalized patients. The criteria used for establishing these cutoff values remains unclear. Here, we compare the overall agreement and concordance of five serologic assays included in the August 2020 FDA EUA at both the manufacturer-recommended qualitative cutoff thresholds and at the FDA-indicated thresholds for high-titer CCP, using

Regulation of PINX1 expression ameliorates lipopolysaccharide-induced lung injury and alleviates cell senescence during the convalescent phase through affecting the telomerase activity.

PIN2/TERF1-interacting telomerase inhibitor 1 (PINX1) is necessary for telomerase reverse transcriptase (TERT) elements to bind at telomeres and non-telomere sites. We aimed to investigate the role of PINX1 and TERT in lipopolysaccharide (LPS)-induced lung injury during acute stage and convalescent phase. Lung injury rat model was induced, and the expression of PINX1 and TERT in serum and lung tissues was examined using RT-qPCR on day 0 (D0), D3, and D14, respectively. The pathologic changes of lung tissues on D3 and D14 were detected using hematoxylin and eosin staining after TERT overexpress
